| 1 | 乳腺癌组织miR-23a、miR-1247-5p表达与临床病理特征及预后的关系显示文摘目的检测微小RNA(miR)-23a、miR-1247-5p在乳腺癌组织中的表达情况,并分析miR-23a、miR-1247-5p表达与乳腺癌患者临床病理特征及预后的关系。方法收集2012年1月至2013年12月在昆山市第二人民医院行手术切除的150例乳腺癌患者术后的乳腺癌组织及癌旁正常组织样本,采用RT-PCR法检测组织样本中miR-23a、miR-1247-5p表达水平。单因素分析miR-23a、miR-1247-5p表达水平与患者临床病理特征的关系;Kaplan-Meier生存曲线分析不同miR-23a、miR-1247-5p表达水平患者的术后生存情况;ROC曲线分析miR-23a、miR-1247-5p对患者预后的预测价值。结果与癌旁正常组织相比,乳腺癌组织miR-23a表达水平较高、miR-1247-5p表达水平较低(均P<0.05)。乳腺癌组织miR-23a表达水平与肿瘤直径、分子分型、淋巴结转移、TNM分期有关(均P<0.05),miR-1247-5p表达水平与组织病理分级、分子分型、淋巴结转移、TNM分期有关(均P<0.05)。miR-23a高表达、miR-1247-5p低表达患者的术后5年总生存率及中位生存时间均分别低于miR-23a低表达、miR-1247-5p高表达患者(均P<0.05)。检测乳腺癌组织miR-23a、miR-1247-5p表达水平对患者预后预测的AUC分别为0.709(95%CI:0.625~0.793)、0.690(95%CI:0.606~0.775),灵敏度分别为0.812、0.749,特异度分别为0.637、0.622。结论乳腺癌组织miR-23a表达上调,miR-1247-5p表达下调,且与患者预后有关。检测乳腺癌患者术后癌组织miR-23a、miR-1247-5p表达水平对患者预后预测有一定的临床价值。 | 郭斌 于桑桑 顾蓓 | 2020 | 浙江医学2020,42,23: | 5 |

| 3 | 可视LAMP-荧光检测金黄色葡萄球菌方法的建立显示文摘将环介导等温扩增(loop-mediated isothermal amplification assay,LAMP)技术与荧光检测系统相结合,建立可视LAMP-荧光金黄色葡萄球菌检测方法。根据金黄色葡萄球菌基因保守序列,利用LAMP在线软件设计4条最佳引物,通过优化反应温度,建立LAMP方法,利用3株金黄色葡萄球菌和10株非金黄色葡萄球菌验证方法的特异性。实验结果表明:构建的LAMP-荧光方法优化后的扩增条件为63%,时间lh,菌液最低检测浓度为87cfu/ml,是普通PCR扩增方法的10倍;LAMP方法的特异性良好,其中3株金黄色葡萄球菌均有阳性扩增,6株非金黄色葡萄球菌都没有特异性扩增。LAMP-荧光方法对110份样品(肉类及其制品、枸杞)进行检测,检出10份LAMP阳性,与之前国标法(GB)检测结果一致。 | 郝俊虎 贺生芳 郭澍强 | 2019 | 中国口岸科学技术2019,0,10: | 0 |
| 4 | miR-23a Promoting Cell Proliferation of Human Tongue Squamous Cell Carcinoma Cell through Regulating PPP2R5E显示文摘[Objectives]To investigate the mechanism of miR-23a in the proliferation of human tongue squamous cell carcinoma cells.[Methods]Clinical tissue samples of tongue squamous cell carcinoma were collected and Tca8113 and CAL27 were cultured.Real-time quantitative PCR was performed to detect the expressions of miR-23a and PPP2R5E in clinical tissue samples of tongue squamous cell carcinoma.MTT assay,colony formation assay and growth curve assay were used to detect the effect of miR-23a and PPP2R5E on the proliferation of human tongue squamous carcinoma cell.Luciferase reporter assay verified the regulatory relationship between miR-23a and PPP2R5E.[Results]The expression of miR-23a in tongue squamous cell carcinoma was significantly up-regulated(P<0.01).miR-23a promoted the proliferation of tongue squamous cell carcinoma cells.Bioinformatics prediction and luciferin reporting experiments showed that PPP2R5E was a direct target gene of miR-23a.The expression of PPP2R5E was decreased in tongue squamous cell carcinoma.PPP2R5E inhibited the proliferation of tongue squamous cell carcinoma cells.Overexpression of PPP2R5E can reverse the proliferation promoting effect of miR-23a on tongue squamous cell carcinoma cells.[Conclusions]miR-23a can promote the proliferation of tongue squamous cell carcinoma cells through PPP2R5E and miR-23a plays an oncogene role in the occurrence and development of tongue squamous cell carcinoma. | Xue LIU Yadong TAO Jihong SUN Feng HUO Hongjie GUO | 2020 | Medicinal Plant2020,11,2: | 0 |
