1正交试验优选子午沙鼠继发性骨包虫病的三维适形调强放疗方案显示文摘目的探讨三维适形调强放疗(intensity-modulated radiation therapy,IMRT)计划中对骨包虫杀灭效力高、优化且经济的继发性骨包虫病的放疗方案。方法右侧股骨继发性骨包虫病成年雄性子午沙鼠60只,采用L12(3×23)正交试验表对不同水平的放疗射野、分割剂量、剂量率、射线角度4个因素进行分配,在5周内对鼠股骨继发性包虫病灶行总剂量为20 Gy的IMRT。于放疗24 h后取子午沙鼠股骨包虫包囊,采用Western blot法检测骨包虫包囊组织凋亡因子Fas蛋白相对表达量。结果放疗后,60只鼠均未发现组织放疗损伤;正交实验结果显示,相同放疗总剂量情况下,各因素对鼠股骨继发性骨包虫杀灭作用影响程度依次为单次放疗剂量(R=2.17)、剂量率(R=0.72)、射野数(R=0.48)、射线角度(R=0.11);子午沙鼠股骨骨包虫包囊组织Fas蛋白相对表达量在不同放射野数、不同单次放疗剂量、不同剂量率上差异有统计学意义(P<0.05),在不同射线角度上差异无统计学意义(P>0.05);当单次放疗剂量为5 Gy、剂量率为1.5 Gy/min、射野数为9野、射线角度为等角度时,骨包虫包囊组织Fas蛋白相对表达量最高,对骨包虫的杀灭效果最好。结论IMRT生物安全性好,单次放疗剂量为5 Gy、剂量率为1.5 Gy/min、射野数为9野、射线角度为等角度是最优的骨包虫放疗方案。杨秀恩 地力木拉提·艾克热木 徐万龙 2020中华实用诊断与治疗杂志2020,34,6:2
2Effect of intensity-modulated radiation therapy on sciatic nerve injury caused by echinococcosis显示文摘Conventional radiotherapy has a good killing effect on femoral echinococcosis.However,the sciatic nerve around the lesion is irreversibly damaged owing to bystander effects.Although intensity-modulated radiation therapy shows great advantages for precise dose distribution into lesions,it is unknown whether intensity-modulated radiation therapy can perfectly protect the surrounding sciatic nerve on the basis of good killing of femoral echinococcosis foci.Therefore,this study comparatively analyzed differences between intensity-modulated radiation therapy and conventional radiotherapy on the basis of safety to peripheral nerves.Pure-breed Meriones meridiani with bilateral femoral echinococcosis were selected as the research object.Intensity-modulated radiation therapy was used to treat left femoral echinococcosis of Meriones meridianus,while conventional radiotherapy was used to treat right femoral echinococcosis of the same Meriones meridianus.The total radiation dose was 40 Gy.To understand whether intensity-modulated radiation therapy and conventional radiotherapy can kill femoral echinococcosis,trypan blue staining was used to detect pathological changes of bone Echinococcus granulosus and protoscolex death after radiotherapy.Additionally,enzyme histochemical staining was utilized to measure acid phosphatase activity in the protoscolex after radiotherapy.One week after radiotherapy,the overall structure of echinococcosis in bilateral femurs of Meriones meridiani treated by intensity-modulated radiation therapy disappeared.There was no significant difference in the mortality rate of protoscoleces of Echinococcus granulosus between the bilateral femurs of Meriones meridiani.Moreover,there was no significant difference in acid phosphatase activity in the protoscolex of Echinococcus granulosus between bilateral femurs.To understand the injury of sciatic nerve surrounding the foci of femoral echinococcosis caused by intensity-modulated radiation therapy and conventional radiotherapy,the ultrastructure of sciatic nerves after radiotherapy was observed by transmission electron microscopy.Additionally,apoptosis of neurons was examined using a terminaldeoxynucleotidyl transferase-mediated d UTP nick end labeling assay,and expression of Bcl-2 and Bax in sciatic nerve tissue was detected by immunohistochemical staining and western blot assay.Our results showed that most neurons in the left sciatic nerve of Meriones meridiani with echinococcosis treated by intensity-modulated radiation therapy had reversible injury,and there was no obvious apoptosis.Compared with conventional radiotherapy,the number of apoptotic cells and Bax expression in sciatic nerve treated by intensity-modulated radiation therapy were significantly decreased,while Bcl-2 expression was significantly increased.Our findings suggest that intensity-modulated radiation therapy has the same therapeutic effect on echinococcosis as conventional radiotherapy,and can reduce apoptosis of the sciatic nerve around foci caused by radiotherapy.Experiments were approved by the Animal Ethics Committee of People's Hospital of Xinjiang Uygur Autonomous Region,China(Approval No.20130301A41)on March 1,2013.Wan-Long Xu Dilimulati·Aikeremu Jun-Gang Sun Yan-Jun Zhang Jiang-Bo Xu Wen-Zheng Zhou Xi-Bin Zhao Hao Wang Hong Yuan 2021Neural Regeneration Research2021,16,3:2
3二膦酸盐对人骨肉瘤细胞诱导凋亡作用的实验研究显示文摘目的探讨二膦酸盐对人骨肉瘤细胞的诱导凋亡作用及其可能的作用机制。方法将人骨肉瘤MG-63细胞株培养、传代后将细胞分为两组:二膦酸盐400μg/mL干预组、生理盐水空白对照组,孵育72h后,对两组人骨肉瘤MG-63细胞株行免疫荧光检测,观察两组细胞中凋亡因子Caspase-3、Fas的表达;行流式细胞术检测,观察两组细胞的细胞凋亡率。结果二膦酸盐作用于人骨肉瘤MG-63细胞株72h后,免疫荧光检测结果可见二膦酸盐400μg/mL干预组细胞中凋亡因子Caspase-3、Fas大量表达,而生理盐水空白对照组几乎见不到凋亡因子Caspase-3、Fas的表达;流式细胞术检测结果观察到二膦酸盐400μg/mL干预组细胞的细胞凋亡率为54.00%,远大于生理盐水空白对照组的细胞凋亡率3.10%,差异有统计学意义(P<0.05),存在明显的诱导凋亡现象。结论二膦酸盐对体外人骨肉瘤MG-63细胞株存在很强的诱导凋亡作用。买买提艾力.哈斯木 江仁兵 白靖平 徐万龙 2016重庆医学2016,45,27:1
4三维适形调强放疗对大鼠继发性股骨细粒棘球蚴感染的疗效研究显示文摘目的探讨三维适形调强放疗(IMRT)对大鼠继发性股骨细粒棘球蚴感染的疗效及对包囊的杀灭机制。方法选取股骨继发性细粒棘球蚴感染Wistar雄性大鼠75只,其中50只大鼠为IMRT组,以IMRT方式40 Gy剂量照射左侧股骨棘球蚴病灶区,25只为对照组,不做照射。于放射治疗后1周,取大鼠左、右股骨细粒棘球蚴包囊,提取生发层细胞,采用Hoechst 33258荧光染色观察细胞核的形态变化,免疫组化法检测凋亡相关基因Caspase-3和p53的表达情况;ELISA检测IMRT组和对照组大鼠血清中γ干扰素(IFN-γ)、肿瘤坏死因子-α(TNF-α)、白细胞介素4(IL-4)和IL-10细胞因子水平差异;流式细胞术检测两组大鼠脾淋巴细胞CD4+/CD8+的比值。结果IMRT治疗后1周,IMRT组大鼠血清中Th1细胞因子IFN-γ、TNF-α的吸光度(A490值)分别为322.45±94.42、28.76±6.23,均高于对照组的277.82±65.48、19.63±4.76(P<0.05);Th2细胞因子IL-4的A490值为11.02±2.55,低于对照组的13.22±3.13(P<0.05),IL-10的A490值为16.57±3.02,与对照组(20.51±3.17)的差异无统计学意义(P>0.05)。Hoechst 33258荧光染色可观察到IMRT组大鼠左侧股骨细粒棘球蚴包囊的生发层出现细胞凋亡现象,而右侧生发层细胞正常。免疫组化检测结果显示,IMRT组大鼠左侧股骨细粒棘球蚴包囊的生发层细胞凋亡相关基因Caspase-3和p53大量表达,而右侧生发层细胞未出现。流式细胞术检测结果显示,IMRT组大鼠血清脾淋巴细胞CD4+/CD8+的比值为2.96±1.90,高于对照组的1.65±1.31,差异有统计学意义(P<0.05)。结论IMRT可促进大鼠继发性股骨细粒棘球蚴包囊生发层细胞的凋亡,其作用机制一方面可能是通过调节凋亡相关蛋白Caspase-3和p53而实现,另一方面可能是与射线激发宿主对骨棘球蚴有效的免疫应答有关。周文正 孙俊刚 赵喜滨 曹力 2021中国寄生虫学与寄生虫病杂志2021,39,4:1
5调强放射治疗对鼠骨包虫生发细胞周期及caspase-9的影响显示文摘目的探讨三维适形调强放疗(intensity-modulated radiation therapy,IMRT)的放射线对鼠骨包虫生发细胞的细胞周期及凋亡因子caspase-9的影响。方法骨包虫病子午沙鼠20只,将鼠骨包虫内囊中的生发细胞分离并进行体外培养,将体外培养成功的骨包虫生发细胞分为IMRT组和对照组。IMRT组给予总剂量为40 Gy的IMRT放疗,对照组给予假照射。放疗结束24 h后,采用流式细胞仪检测2组骨包虫生发细胞的细胞周期,采用ELISA法检测骨包虫生发细胞中凋亡相关蛋白caspase-9蛋白水平,采用实时荧光定量PCR法检测骨包虫生发细胞caspase-9 mRNA相对表达量。结果放疗结束24 h后,IMRT组骨包虫生发细胞S期细胞比率[(65.65±7.46)%]明显高于对照组[(32.49±4.23)%],G0/G1期[(32.37±4.94)%]和G2/M期[(1.98±0.19)%]细胞比率明显低于对照组[(53.95±4.55)%、(13.55±2.48)%](P<0.05)。IMRT组生发细胞caspase-9蛋白水平[(0.071±0.012)pmol/L]和caspase-9 mRNA相对表达量(30.21±6.99)明显高于对照组[(0.011±0.004)pmol/L、12.17±3.01](P<0.05)。结论IMRT可改变鼠骨包虫生发细胞的细胞周期,抑制生发细胞的增殖,可能是通过调节凋亡相关蛋白caspase-9水平而实现。宋作成 地力木拉提·艾克热木 徐万龙 2020中华实用诊断与治疗杂志2020,34,6:1
6放射治疗对股骨继发性骨包虫病鼠腹股沟淋巴结T淋巴细胞的影响显示文摘目的探讨三维适形调强放疗(intensity-modulated radiation therapy,IMRT)对股骨继发性骨包虫病鼠腹股沟淋巴结T淋巴细胞的影响及机制。方法股骨继发性骨包虫病子午沙鼠20只,随机分为IMRT组和对照组各10只,IMRT组股骨包虫病灶区给予40 Gy的IMRT放射治疗,对照组给予假照射。放疗结束1周后,采用电镜观察2组鼠骨包虫细胞形态学改变情况,采用免疫荧光法观察鼠腹股沟淋巴结CD4^+、CD8^+T淋巴细胞表达情况,应用流式细胞仪检测鼠腹股沟淋巴结CD4^+、CD8^+T淋巴细胞比率,计算CD4^+/CD8^+。结果放疗1周后,IMRT组鼠股骨包虫细胞大量坏死,对照组鼠股骨包虫细胞形态正常;IMRT组鼠腹股沟淋巴结CD4^+、CD8^+T淋巴细胞大量表达,对照组鼠腹股沟淋巴结CD4^+、CD8^+T淋巴细胞少量表达;IMRT组鼠腹股沟淋巴结CD4^+T淋巴细胞比率[(89.73±12.14)%]和CD4^+/CD8^+(5.29±0.69)高于对照组[(71.16±11.03)%、2.89±0.37],CD8^+T淋巴细胞比率[(16.96±3.14)%]低于对照组[(24.65±4.29)%](P<0.05)。结论IMRT对鼠股骨继发性骨包虫有较好的杀灭效果,其作用机制可能与放疗激发宿主对骨包虫有效的免疫反应有关。宋作成 地力木拉提·艾克热木 徐万龙 2020中华实用诊断与治疗杂志2020,34,6:1
