| 3 | 肝细胞癌患者血浆基质金属蛋白酶的检测及临床意义显示文摘目的探讨基质金属蛋白酶(MMPs)在肝细胞癌(HCC)患者中的表达水平及其与临床病理学特征的关系。方法选取2015年10月-2018年5月在首都医科大学附属北京佑安医院住院治疗的患者131例,其中乙型肝炎肝硬化患者35例,HCC患者96例;另选取同期门诊体检的健康对照者20例;采用悬浮液相芯片技术检测患者治疗前血浆MMP-1、MMP-2、MMP-7、MMP-9、MMP-10水平。非正态分布的计量资料2组间比较采用Mann-Whitney U检验,3组间比较采用Kruskal-Wallis H检验,进一步两两比较采用Dunn-Bonferroni检验,2组样本的相关性检验采用Spearman相关分析。结果 HCC患者血浆MMP-1水平为8134. 84(6147. 94~11 148. 47) pg/ml、MMP-7水平为6541. 58(3906. 63~9033. 12) pg/ml,均高于肝炎肝硬化组及健康对照组,差异均有统计学意义(χ~2值分别为23. 521、66. 112,P值均<0. 001); HCC患者血浆MMP-2水平为103 774. 45(90 485. 91~123 673. 90)pg/ml,高于健康对照组,差异有统计学意义(P <0. 05)。肿瘤直径≥5 cm、伴有远处转移、合并门静脉侵犯、临床分期Ⅲ/Ⅳ期或病理ESⅢ/Ⅳ级组患者血浆MMP-1水平分别高于肿瘤直径<5 cm(Z=-7. 313,P <0. 001)、无远处转移(Z=-6. 569,P <0. 001)、无门静脉侵犯(Z=-6. 051,P <0. 001)、临床分期Ⅰ/Ⅱ期(Z=-5. 844,P <0. 001)、病理ESⅠ/Ⅱ级组(Z=-7. 423,P <0. 001)患者,差异均有统计学意义。伴有远处转移、合并门静脉侵犯、临床分期Ⅲ/Ⅳ期、病理ESⅢ/Ⅳ级组患者血浆MMP-7水平分别高于无远处转移(Z=-3. 454,P=0. 001)、无门静脉侵犯(Z=-3. 846,P <0. 001)、临床分期Ⅰ/Ⅱ期(Z=-2. 285,P=0. 022)、病理ESⅠ/Ⅱ级组(Z=-3. 287,P=0. 001)患者,差异均有统计学意义。结论 HCC患者MMP-1和MMP-7的表达与HCC转移、门静脉侵犯、临床分期及病理学分级相关,可协助判断HCC患者的肿瘤侵袭严重程度及预后。 | 张洪海 孙玉 刘芳 谢立 乔录新 陈德喜 殷继明 | 2019 | 临床肝胆病杂志2019,35,4: | 6 |
| 4 | Post-surgical resection prognostic value of combined OPN, MMP7, and PSG9 plasma biomarkers in hepatocellular carcinoma显示文摘Biomarkers for hepatocellular carcinoma (HCC) following curative resection are not currently sufficient for prognostic indication of overall survival (OS) and disease-free survival (DFS). The aim of this study was to investigate the prognostic performance of osteopontin (OPN), matrix metalloproteinase 7 (MMP7), and pregnancy specific glycoprotein 9 (PSG9) in patients with HCC. A total of 179 prospective patients with HCC provided plasma before hepatectomy. Plasma OPN, MMP7, and PSG9 levels were determined by enzyme-linked immunosorbent assay. Correlations between plasma levels, clinical parameters, and outcomes (OS and DFS) were overall analyzed. High OPN (≥149.97 ng/mL), MMP7 (≥2.28 ng/mL), and PSG9 (≥45.59 ng/mL) were prognostic indicators of reduced OS (P<0.001, P<0.001, and P=0.007, respectively). Plasma PSG9 protein level was an independent factor in predicting OS (P=0.008) and DFS (P=0.038). Plasma OPN+MMP7+PSG9 elevation in combination was a prognostic factor for OS (P<0.001). OPN was demonstrated to be a risk factor-associated OS in stage I patients with HCC and patients with low α-fetoprotein levels (<20 ng/mL). | 5 | Smoc2 potentiates proliferation of hepatocellular carcinoma cells via promotion of cell cycle progression显示文摘AIM To determine the influence of Smoc2 on hepatocellular carcinoma(HCC) cell proliferation and to find a possible new therapeutic target for preventing HCC progression.METHODS We detected expression of Smoc2 in HCC tissues and corresponding non-tumor liver(CNL) tissues using PCR, western blot, and immunohistochemistry methods. Subsequently, we down-regulated and upregulated Smoc2 expression using siR NA and lentivirus transfection assay, respectively. Then, we identified the effect of Smoc2 on cell proliferation and cell cycle using CCK-8 and flow cytometry, respectively. The common cell growth signaling influenced by Smoc2 was detected by western blot assay. RESULTS The expression of Smoc2 was significantly higher in HCC tissues compared with CNL tissues. Overexpression of Smoc2 promoted HCC cell proliferation and cell cycle progression. Down-regulation of Smoc2 led to inhibition of cell proliferation and cell cycle progression. | 6 | OPN基因启动区-156delG多态性与中国HBV相关肝细胞癌的易感性显示文摘【目的】研究骨桥蛋白(OPN)基因启动区多态性与乙型肝炎病毒相关肝细胞癌(HBV-HCC)遗传易感性的关系。【方法】225例确诊为HBV-HCC患者与200例年龄匹配的慢性乙型肝炎患者被纳入研究。OPN基因启动区3个多态性位点(-156del G/G,-443T/C和-616T/G)通过直接测序法确定基因型。【结果】HBV-HCC组-156 del G/del G基因型分布频率高于对照组(P=0.003)。HCC组-156 del G等位基因频率显著增高(P<0.001)。Logistic回归分析显示等位基因-156del G是HBV-HCC的易感因素(OR=1.643,95%CI 1.249-2.161)。两组间-443T/C、-616T/G基因型分布和等位基因频率差异没有统计学意义。【结论】OPN基因启动区-156等位基因可能是中国人群HBV-HCC的易感因素。 | 时红 吴元凯 林国莉 李向永 | 2017 | 中山大学学报(医学科学版)2017,38,5: | 2 |
| 7 | Ku70蛋白在肝细胞癌患者体内的表达规律及其意义显示文摘目的研究Ku70蛋白在肝细胞癌(HCC)患者体内的表达规律及其临床意义。方法采用免疫组织化学染色方法检测106例肝癌、癌旁和92例正常肝组织中Ku70蛋白的表达,探讨Ku70蛋白表达与临床病理参数的关系及与生存情况的关系。结果肝癌组织中Ku70蛋白阳性表达率高于癌旁组织和正常肝组织(P<0.01)。ChildPugh分级、肿瘤大小、肿瘤分化程度、TNM分期是HCC患者肝癌组织中Ku70蛋白表达水平升高的危险因素。生存分析表明低表达组的生存率显著高于高表达组(P<0.05)。结论 Ku70蛋白可作为HCC早期筛查和预后判断的新型肿瘤标志物。 | 杨成林 李俊杰 孙学军 | 2015 | 解放军医药杂志2015,27,4: | 1 |
| 8 | 慢性HBV感染者GPI、MMP-1、MMP-3测定的临床意义显示文摘目的:检测葡萄糖-6-磷酸异构酶(glucose-6-phosphate isomerase,GPI)、基质金属蛋白酶(matrix metalloproteinase,MMP)-1、MMP-3血清标志物在慢性乙型肝炎(chronic hepatitis B,CHB)、肝硬化(liver cirrhosis,LC)及肝细胞肝癌(hepatocellular carcinoma,HCC)患者中的表达水平,探讨其在慢性肝病及肝癌进展过程中的变化及意义。方法:应用酶联免疫吸附试验定量检测受检者血清中GPI、MMP-1、MMP-3的浓度,AU2700全自动生化分析仪测定谷丙转氨酶(alanine aminotransferase,ALT)及谷草转氨酶(aspartate transaminase,AST),分析GPI、MMP-1、MMP-3与ALT、AST的相关性。其中CHB 31例,LC 40例、HCC 46例,健康体检者50例。结果:3组肝病患者中血清GPI、MMP-1、MMP-3表达水平均显著高于健康对照组(均P<0.05);在LC患者中,随着AST的增加MMP-3浓度增高,二者呈弱正相关(r=0.18,P<0.05)。AST>40 IU/L LC患者血清MMP-3浓度显著高于AST≤40 IU/L者(P<0.05)。GPI、MMP-1与ALT、AST均无相关性。结论:血清GPI、MMP-1、MMP-3可作为乙型肝炎病毒感染相关肝病肝损伤的诊断指标。 | 韩刚 倪翠萍 罗文明 吴燕 | 2017 | 南通大学学报(医学版)2017,37,2: | 0 |
