| 1 | Dysregulation of autophagy and mitochondrial function in Parkinson’s disease显示文摘Parkinson’s disease(PD)is the second most common neurodegenerative disease.Increasing evidence supports that dysregulation of autophagy and mitochondrial function are closely related with PD pathogenesis.In this review,we briefly summarized autophagy pathway,which consists of macroautophagy,microautophagy and chaperone-mediated autophagy(CMA).Then,we discussed the involvement of mitochondrial dysfunction in PD pathogenesis.We specifically reviewed the recent developments in the relationship among several PD related genes,autophagy and mitochondrial dysfunction,followed by the therapeutic implications of these pathways.In conclusion,we propose that autophagy activity and mitochondrial homeostasis are of high importance in the pathogenesis of PD.Better understanding of these pathways can shed light on the novel therapeutic methods for PD prevention and amelioration. | Bao Wang Neeta Abraham Guodong Gao Qian Yang | 2016 | Translational Neurodegeneration2016，5，1： | 5 |
| 2 | TRPC3参与α-突触核蛋白引起的线粒体损伤显示文摘目的探讨经典型瞬时受体电位通道3(transient receptor potential canonical channel 3,TRPC3)是否参与α-突触核蛋白(α-synuclein,α-syn)引起的线粒体损伤。方法在α-syn过表达原代培养神经元、α-syn转基因及敲除小鼠模型,利用免疫印迹和免疫荧光技术检测TRPC3和α-syn在线粒体内的定位和表达;在原代培养神经元,运用MTT和乳酸脱氢酶法检测细胞活力和受损情况,JC-1法检测线粒体膜电势,观察敲减TRPC3对α-syn过表达引起的线粒体损伤和细胞损伤的影响。结果 TRPC3和α-syn共同表达于线粒体,过表达α-syn增加TRPC3在线粒体的分布,敲除α-syn则下调TRPC3在线粒体的分布。敲减TRPC3明显减轻过表达α-syn引起的线粒体膜电势下降和细胞毒性。结论 TRPC3可能参与过表达α-syn引起的线粒体损伤。 | 卢勇泉 陈敏 高歌 段春礼 鲁玲玲 杨慧 | 2017 | 首都医科大学学报2017，38，6： | 2 |
| 3 | α-突触核蛋白/A53T突变体损伤SH-SY5Y细胞的MAM并导致线粒体结构异常及自噬显示文摘目的研究α-突触核蛋白(α-syn)A53T突变体对人神经母细胞瘤细胞内质网线粒体相关膜(MAM)的影响,进而探讨α-syn突变后导致神经元毒性的分子细胞机制。方法首先构建过表达α-syn/A53T突变基因的人神经母细胞瘤细胞系(SH-SY5Y)细胞,然后利用分子邻位杂交(PLA)的方法标记MAM并计数MAM的数量。用Rhod2标记线粒体钙离子,动态观测线粒体钙离子变化情况。用激光共聚焦显微镜及电镜技术进一步观察线粒体的结构。结果过表达α-syn/A53T突变体可以导致明显的细胞毒性(P<0.001),MAM的数量显著减少(P<0.001),MAM钙离子转运受到抑制。线粒体变为短棒状、球状,并在神经细胞内出现线粒体空泡以及自噬体。结论α-syn/A53T突变体可以通过破坏SH-SY5Y细胞的MAM导致线粒体结构异常并自噬。 | 岳静静 伍超 高歌 鲁玲玲 | 2023 | 基础医学与临床2023，43，2： | 0 |
