1Implication of Neuronal Versus Microglial P2X4 Receptors in Central Nervous System Disorders显示文摘The P2X4 receptor(P2X4)is an ATP-gated cation channel that is highly permeable to Ca2+and widely expressed in neuronal and glial cell types throughout the central nervous system(CNS).A growing body of evidence indicates that P2X4 plays key roles in numerous central disorders.P2X4 trafficking is highly regulated and consequently in normal situations,P2X4 is present on the plasma membrane at low density and found mostly within intracellular endosomal/lysosomal compartments.An increase in the de novo expression and/or surface density of P2X4 has been observed in microglia and/or neurons during pathological states.This review aims to summarize knowledge on P2X4 functions in CNS disorders and provide some insights into the relative contributions of neuronal and glial P2X4 in pathological contexts.However,determination of the cell-specific functions of P2X4 along with its intracellular and cell surface roles remain to be elucidated before its potential as a therapeutic target in multiple disorders can be defined.Alexia Duveau Eleonore Bertin Eric Boue-Grabot 2020Neuroscience Bulletin2020,36,11:3
2Trafficking pathway between plasma membrane and mitochondria via clathrin-mediated endocytosis显示文摘Endocytosis is a basic cellular process that describes a form of active transport across the plasma membrane into the cell.The endocytic pathway consists of distinct membrane compartments;internalized molecules are delivered to early endosomes,and some of them are recycled back to the surface,whereas other molecules are sent to late endosomes and lysosomes for degradation.However,little is known about how mitochondria are involved in the endocytic pathway.Here,we report that FM dyes, membrane-impermeant fluorescent lipid probes,can traffic to mitochondria directly from the plasma membrane by clathrinmediated endocytosis.FM dye entry into mitochondria uses microtubule-dependent active transport,but the mechanism is different from the classical endocytic pathway.Hence,this study reveals a previously unrealized lipid trafficking pathway from the plasma membrane to mitochondria.Zhongya Wei Wenfeng Su Huifang Lou Shumin Duan Gang Chen 2018Journal of Molecular Cell Biology2018,10,6:2
3Rab27b is Involved in Lysosomal Exocytosis and Proteolipid Protein Trafficking in Oligodendrocytes显示文摘Myelination by oligodendrocytes in the central nervous system requires coordinated exocytosis and endocytosis of the major myelin protein, proteolipid protein(PLP). Here, we demonstrated that a small GTPase,Rab27b, is involved in PLP trafficking in oligodendrocytes.We showed that PLP co-localized with Rab27b in late endosomes/lysosomes in oligodendrocytes. Short hairpinmediated knockdown of Rab27b not only reduced lysosomal exocytosis but also greatly diminished the surface expression of PLP in oligodendrocytes. In addition,knockdown of Rab27b reduced the myelin-like membranes induced by co-culture of oligodendrocytes and neurons.Our data suggest that Rab27b is involved in myelin biogenesis by regulating PLP transport from late endosomes/lysosomes to the cell membrane in oligodendrocytes.Yun-Tian Shen Yun Gu Wen-Feng Su Jing-fei Zhong Zi-Han Jin Xiao-Song Gu Gang Chen 2016Neuroscience Bulletin2016,32,4:2
