| 2 | 黄连素抑制直肠癌SW480细胞增殖的实验研究显示文摘目的观察黄连素抑制直肠癌SW480细胞增殖的效果并探讨其作用机制。方法治疗组用不同浓度的黄连素(0μmol/L、25μmol/L、50μmol/L、100μmol/L、150μmol/L、200μmol/L)干预SW480细胞株。MTT比色法观察最佳作用浓度及细胞增殖效果。结果显示最佳作用浓度为150μmol/L。后续实验治疗组SW480细胞用150μmol/L黄连素干预48小时,对照组用RPMI-1640培养基。流式细胞仪检测两组SW480细胞的细胞周期,Annexin V检测细胞凋亡。免疫印迹法(Western blot)分析治疗组和对照组中Livin、YKL-40、AKT和Cyclin D1蛋白的表达。酶联免疫吸附法检测两组促血管生成素-2(Ang-2)的表达水平。结果黄连素干预SW480细胞后对细胞增殖有明显抑制作用,SW480细胞周期被阻滞在G1期并伴有细胞凋亡比例明显增加。Livin、YKL-40、AKT和Cyclin D1蛋白的表达水平明显降低,并且黄连素可明显抑制YKL-40/AKT/Cyclin D1信号通路和SW480细胞中Ang-2的表达水平。结论黄连素能有效抑制SW480细胞的增殖,其作用机制包括下调YKL-40/AKT/Cyclin D1信号通路从而使细胞周期被阻滞在G1期,通过降低Livin蛋白的表达诱导细胞凋亡,并可下调Ang-2的表达抑制血管生成。 | 彭洪 林中超 彭明沙 祝秀华 刘娟 | 2018 | 西部医学2018,30,2: | 4 |
| 3 | Plasma chitinase 3-like 1 is persistently elevated during first month after minimally invasive colorectal cancer resection显示文摘AIM: To assess blood chitinase 3-like 1(CHi3L1) levels for 2 mo after minimally invasive colorectal resection(MICR) for colorectal cancer(CRC). METHODS: CRC patients in an Institutional Review Board approved data/plasma bank who underwent elective MICR for whom preoperative(PreO p), early postoperative(PostO p), and 1 or more late PostO p samples [postoperative day(POD) 7-27] available were included. Plasma CHi3L1 levels(ng/m L) were determined in duplicate by enzyme linked immunosorbent assay. RESULTS: PreOp and PostOp plasma sample were available for 80 MICR cancer patients for the study. The median PreOp CHi3L1 level was 56.8 CI: 41.9-78.6 ng/mL(n = 80). Significantly elevated(P < 0.001) median plasma levels(ng/mL) over PreOp levels were detected on POD1(667.7 CI: 495.7, 771.7; n = 79), POD 3(132.6 CI: 95.5, 173.7; n = 76), POD7-13(96.4 CI: 67.7, 136.9; n = 62), POD14-20(101.4 CI: 80.7, 287.4; n = 22), and POD 21-27(98.1 CI: 66.8, 137.4; n = 20, P = 0.001). No significant difference in plasma levels were noted on POD27-41. CONCLUSION: Plasma CHi3L1 levels were significantly elevated for one month after MICR. | 5 | Novel methylxanthine derivative-mediated anti-inflammatory effects in inflammatory bowel disease显示文摘Family 18 chitinases have a binding capacity with chitin, a polymer of N-acetylglucosamine. Recent studies strongly suggested that chitinase 3-like 1(CHI3L1, also known as YKL-40) and acidic mammalian chitinase, the two major members of family 18 chitinases, play a pivotal role in the pathogenesis of inflammatory bowel disease(IBD), bronchial asthma and several other inflammatory disorders. Based on the data from highthroughput screening, it has been found that three methylxanthine derivatives, caffeine, theophylline, and pentoxifylline, have competitive inhibitory effects against a fungal family 18 chitinase by specifically interacting with conserved tryptophans in the active site of this protein. Methylxanthine derivatives are also known as adenosine receptor antagonists, phosphodiesterase inhibitors and histone deacetylase inducers. Anti-in-flammatory effects of methylxanthine derivatives have been well-documented in the literature. For example, a beneficial link between coffee or caffeine consumption and type 2 diabetes as well as liver cirrhosis has been reported. Furthermore, theophylline has a long history of being used as a bronchodilator in asthma therapy, and pentoxifylline has an immuno-modulating effect for peripheral vascular disease. However, it is still largely unknown whether these methylxanthine derivativemediated anti-inflammatory effects are associated with the inhibition of CHI3L1-induced cytoplasmic signaling cascades in epithelial cells. In this review article we will examine the above possibility and summarize the biological significance of methylxanthine derivatives in intestinal epithelial cells. | 6 | 久泻宁颗粒联合白头翁汤治疗慢性非特异性溃疡性结肠炎疗效观察及对血清几丁质酶-3样蛋白-1含量的影响显示文摘目的观察久泻宁颗粒联合白头翁汤治疗慢性非特异性溃疡性结肠炎(CUC)临床疗效,以及对血清几丁质酶-3样蛋白-1(Chtinase-3-like-1 protein,YKL-40)含量的影响。方法将60例CUC患者随机分为2组,治疗组30例予久泻宁颗粒联合白头翁汤治疗,对照组30例予美沙拉嗪颗粒剂治疗。2组均治疗8周后统计综合疗效、中医证候疗效及肠镜下黏膜疗效,并观察2组治疗前后症状积分、疾病活动指数评分(DAI)及YKL-40情况。结果临床综合疗效:治疗组总有效率90.0%,对照组总有效率76.7%,2组比较差异无统计学意义(P>0.05)。中医证候疗效:治疗组总有效率90.0%,对照组总有效率76.7%,2组比较差异有统计学意义(P<0.05),治疗组疗效优于对照组。肠镜下黏膜疗效:治疗组总有效率83.33%,对照组总有效率73.33%,2组比较差异无统计学意义(P>0.05)。2组治疗后症状总积分与本组治疗前比较差异均有统计学意义(P<0.05),但2组治疗后症状总积分组间比较差异无统计学意义(P>0.05)。2组治疗后主要症状积分均降低(P<0.05),且治疗组治疗后腹泻、里急后重症状积分降低优于对照组(P<0.05)。2组治疗后DAI评分与本组治疗前比较差异均有统计学意义(P<0.05),但2组治疗后组间比较差异无统计学意义(P>0.05)。2组治疗后血清YKL-40水平与本组治疗前比较差异均有统计学意义(P<0.05),但2组治疗后组间比较差异无统计学意义(P>0.05)。结论久泻宁颗粒联合白头翁汤治疗CUC疗效确切,并可改善患者的临床症状及中医症状,同时降低血清YKL-40水平。 | 彭洪 林中超 祝秀华 苏强 | 2017 | 河北中医2017,39,12: | 2 |
| 7 | Increased expression and possible role of chitinase 3-like-1 in a colitis-associated carcinoma model显示文摘AIM:To investigate the possible role of chitinase3-like-1(CHI3L1)in the progression of colitis-associated carcinoma(CAC).METHODS:Thirty-four Balb/c mice were randomly assigned to five groups,including the control,CAC control,CAC+caffeine,colitis control and colitis+caffeine.Three animals were sacrificed every two weeks for blinded macroscopic inspection,histological analysis,and total RNA extraction.An immunofluorescent assay was performed using specimens from the colitis control and colitis+caffeine groups to investigate whether the protective effect of caffeine was associated with less oxidative DNA damage.In vitro,HT29 cells prestimulated with different concentrations of recombinant CHI3L1 protein and H2O2 were loaded with the DCFHDA fluorescent probe to determine the effect of CHI3L1on intracellular reactive oxygen species production.RESULTS:CHI3L1 mRNA was increased during the progression of colon carcinogenesis.Tumors were mostly located in the distal end of the colon where the expression of CHI3L1 was higher than in the proximal colon.Caffeine-treated mice developed fewer tumors and milder inflammation than untreated mice.CHI3L1protein increased reactive oxygen species in HT29 cells when exposed to H2O2.CONCLUSION:Caffeine reduces tumor incidence by decreasing oxidative DNA damage.CHI3L1 may contribute to CAC by increasing reactive oxygen species production. | Jia-Yi Ma Run-Hua Li Kun Huang Gao Tan Chen Li Fa-Chao Zhi | 2014 | World Journal of Gastroenterology2014,20,42: | 2 |
| 8 | 跑台运动对腹主动脉缩窄诱导的心衰大鼠心肌及骨骼肌CHI3L1表达的影响及机制显示文摘目的探讨跑台运动对心衰大鼠心肌、骨骼肌和循环中CHI3L1表达的影响以及可能机制。方法健康雄性SD大鼠随机分成假手术对照组(CON,n=8)、腹主动脉缩窄手术组(AAC,n=8)、AAC加跑台运动干预组(AAC+E,n=8),在手术4周后进行连续4周,每周5 d,每天40 min的跑台有氧运动干预,跑台速度12 m/min。在运动结束后次日进行超声心动测试观察大鼠心脏的结构及功能;采用RT-PCR检测心肌、骨骼肌中相关指标的mRNA表达水平;Western-blotting检测心肌组织中CHI3L1的蛋白表达;ELISA检测血清中CHI3L1、IL-6、IL-10及TGF-β1的表达水平。结果(1)与CON组相比,8周AAC大鼠左心室射血分数和左心室缩短分数明显下降(P<0.001);与AAC 8周心衰大鼠相比,4周跑台有氧运动使心衰大鼠(AAC+E组)左心室舒张末期内径、左心室收缩末期内径、左心室舒张末期前壁厚度明显下降(P<0.01、P<0.05、P<0.001),左心室射血分数显著升高(P<0.01);与CON组相比,AAC组大鼠心肌ANP和BNP的mRNA表达明显升高(P<0.05);与AAC组相比,AAC+E组大鼠心肌ANP和BNP的mRNA表达显著降低(P<0.01,P<0.05)。(2)与CON组相比,AAC组大鼠血清CHI3L1水平明显升高(P<0.05);心肌CHI3L1的mRNA及蛋白表达显著上升(P<0.05);与AAC组相比,AAC+E组大鼠血清CHI3L1以及心肌CHI3L1的表达水平明显下降(P<0.05);与CON组相比,AAC组大鼠腓肠肌和比目鱼肌中CHI3L1的mRNA表达均显著上升(P<0.05);与AAC组相比,AAC+E组大鼠腓肠肌中CHI3L1的mRNA表达水平升高超过2倍(P<0.01),比目鱼肌中CHI3L1的表达水平也明显升高(P<0.05)。(3)与CON组相比,AAC组大鼠血清IL-6升高近3倍(P<0.01);与AAC组相比,AAC+E组大鼠血清IL-6水平显著下降(P<0.001);与CON组相比,AAC组大鼠血清IL-10水平略下降;与AAC组相比,AAC+E组大鼠血清IL-10明显上调(P<0.01);与AAC组相比,AAC+E组大鼠血清TGF-β1明显升高(P<0.05)。结论跑台运动降低了AAC诱导的心力衰竭大鼠循环CHI3L1水平,抑制了心肌CHI3L1的表达,运动可能通过减轻心衰时炎症水平,发挥对CHI3L1的调节作用。 | 郭兰兰 邢正 李灵杰 李世田 张靓 | 2022 | 西安体育学院学报2022,39,5: | 0 |
| 9 | CAF的分泌组学在免疫联合疗法中的应用显示文摘癌相关成纤维细胞(CAF)是肿瘤微环境中最丰富的细胞群,在肿瘤进展中起着至关重要的作用。CAF在肿瘤微环境中能分泌一些趋化因子,如IL-6、Chi3L1、SDF-1、CXCL12,这些因子通过旁分泌作用促血管生成、肿瘤侵袭及转移。这些因子不仅影响肿瘤免疫,还在肿瘤微环境中协助肿瘤细胞进行免疫逃逸,从而促进肿瘤进展和免疫耐受等。近年来,免疫检查点抑制剂在部分肿瘤晚期患者中应用,取得了较好的临床效果,部分患者得以长期存活。因此,靶向肿瘤微环境中特定的分泌因子与免疫检查点抑制剂联合应用,成为了研究的热点。本文综述了CAF的分泌组学及生物学功能的研究进展,并阐述其临床应用前景和面临的现实挑战。 | 张梦琰 石然 何敬东(指导) | 2020 | 中国免疫学杂志2020,36,22: | 0 |
