1索拉菲尼联合紫杉醇对HepG2细胞的抑制作用及其对cyclinD1表达水平的影响显示文摘目的探讨索拉菲尼(SOR)单药、紫杉醇(PTX)单药以及两药联合对肝癌细胞株Hep G2增殖的抑制作用及其对周期素依赖性蛋白cyclin D1表达的影响,探讨其协同作用机制。方法以不同浓度的SOR和不同浓度的PTX,分别组成单药组和联合用药组,另设不加药的对照组,作用于Hep G2细胞。MTT法检测SOR、PTX单药及联用后Hep G2细胞的增殖抑制率,流式细胞仪分析细胞的凋亡率;RT-PCR检测各组细胞中cyclin D1表达。结果 SOR、PTX单药与联用均能抑制Hep G2细胞增殖,且呈明显时间-剂量依赖效应,两药联用有协同效应,可使Hep G2细胞增殖率明显下降,细胞数减少,联合组与对应的单药组间差异显著(P<0.05);药物作用48 h后,给药组cyclin D1 mRNA的表达较对照组明显减少(P<0.05),两药联用cyclin D1表达更显著,与单药组比较有显著差异(P<0.05)。结论 SOR和PTX联用可协同下调Hep G2细胞中cyclin D1表达,增强了抑制Hep G2细胞增殖及诱导其凋亡效应。孙炜 宋祥和 卞勇华 俞敏 李玉华 2015中国老年学杂志2015,35,2:6
2Evaluation of antiangiogenic efficacy in advanced hepatocellular carcinoma: Biomarkers and functional imaging显示文摘Many years after therapeutic wilderness, sorafenib finally showed a clinical benefit in patients with advanced hepatocellular carcinoma. After the primary general enthusiasm worldwide, some disappointments emerged particularly since no new treatment could exceed or at least match sorafenib in this setting. Without these new drugs, research focused on optimi-zing care of patients treated with sorafenib. One challenging research approach deals with identifying prognostic and predictive biomarkers of sorafenib in this population.
