1Targeting the insulin-like growth factor pathway in hepatocellular carcinoma显示文摘Hepatocellular carcinoma(HCC) is the third leading cause of cancer-related deaths worldwide. Only 30%-40% of the patients with HCC are eligible for curative treatments, which include surgical resection as the first option, liver transplantation and percutaneous ablation. Unfortunately, there is a high frequency of tumor recurrence after surgical resection and most HCC seem resistant to conventional chemotherapy and radiotherapy. Sorafenib, a multi-tyrosine kinase inhibitor, is the only chemotherapeutic option for patients with advanced hepatocellular carcinoma. Patients treated with Sorafenib have a significant increase in overall survival of about three months. Therefore, there is an urgent need to develop alternative treatments. Due to its role in cell growth and development, the insulin-like growth factor system is commonly deregulated in many cancers. Indeed, the insulin-like growth factor(IGF) axis has recently emerged as a potential target for hepatocellular carcinoma treatment. To this aim, several inhibitors of the pathway have been developed suchas monoclonal antibodies, small molecules, antisense oligonucleotides or small interfering RNAs. However recent studies suggest that, unlike most tumors, HCC development requires increased signaling through insulin growth factor Ⅱ rather than insulin growth factor Ⅰ. This may have great implications in the future treatment of HCC. 2人原发性肝癌组织中AEG-1、β-catenin和C-myc的表达及其意义显示文摘目的研究星形胶质细胞上调基因-1(AEG-1)、β-连环蛋白(β-catenin)和C-myc在原发性肝癌组织中的表达情况,以探讨AEG-1、β-catenin、C-myc蛋白在肝癌发生、发展中的可能作用。方法以40例原发性肝癌患者术后肝癌组织及癌旁组织作为研究组,8例正常肝组织标本作为对照组,采用免疫组织化学染色及western blot方法测定各组研究对象中AEG-1、β-catenin及C-myc蛋白的表达水平。结果在肝癌组织中AEG-1、β-catenin和C-myc蛋白的表达水平显著高于癌旁组织及正常肝组织,其组间差异有统计学意义(P<0.05);在肝癌组织中AEG-1蛋白和β-catenin蛋白的表达呈正相关(r=0.420),β-catenin蛋白和C-myc蛋白的表达呈正相关(r=0.388),AEG-1蛋白和C-myc蛋白的表达呈正相关(r=0.332),P<0.05。结论 AEG-1、β-catenin和C-myc蛋白在肝癌组织中的表达水平明显升高,提示AEG-1可能通过Wnt信号传导途径调控下游的β-catenin和C-myc蛋白,促进肿瘤细胞增殖从而参与肝细胞肝癌的发生发展。程云娟 罗新华 程明亮 2015贵州医药2015,39,11:3
