| 1 | 黄芪总苷对脾虚大鼠胃黏膜保护机制探讨显示文摘目的:探讨黄芪总苷对冷水-束缚应激状态下脾虚大鼠胃黏膜H+,K+-ATP酶活性及mRNA表达变化、胃蛋白酶活性及髓过氧化物MPO活性的影响。方法:SD大鼠70只分为7组,即正常对照组,脾虚组,模型(脾虚应激)组,黄芪总苷(200mg/kg、100mg/kg、50mg/kg)组,补中益气丸组。采用经典大黄致脾虚法复制脾虚模型大鼠,水浸-束缚法给予脾虚大鼠应激,观察胃黏膜损伤指数,无机磷法检测胃黏膜H+,K+-ATP酶、胃蛋白酶及MPO活性的变化,RT-PCR法测定H+,K+-ATP酶mRNA表达变化。结果:正常对照及脾虚组大鼠胃黏膜未见溃疡损伤,经冷水-束缚应激后的模型组、补中益气丸组、黄芪总苷(高、中、低)组大鼠胃黏膜可见点状或条索状溃疡损伤,且与模型组相比黄芪总苷中剂量及高剂量组、补中益气丸组明显降低(P<0.01)。与正常组比较,模型组大鼠胃黏膜H+,K+-ATP酶活性、胃蛋白酶及髓过氧化物MPO活性值,H+,K+-ATP酶mRNA表达明显增高(P<0.05),而脾虚组降低(P<0.05)。与模型组相比,补中益气丸组、黄芪总苷(中、高)剂量组H+,K+-ATP酶活性、胃蛋白酶及髓过氧化物MPO活性值,H+,K+-ATP酶mRNA表达均有所下降(P<0.05)。结论:黄芪总苷对脾虚大鼠胃黏膜的保护作用机制之一与影响胃蛋白酶、H+,K+-ATP酶活性及其mRNA表达有关。 | 米红 李燕舞 王晓燕 祁燕 巫燕莉 王汝俊 | 2012 | 中药药理与临床2012,28,5: | 23 |
| 2 | 白藜芦醇对大鼠脊髓损伤后炎症反应的影响显示文摘目的探讨白藜芦醇对大鼠脊髓损伤后炎症反应的影响。方法 63只SD大鼠随机分为假手术组、损伤组、白藜芦醇处理组,每组21只。假手术组仅行手术暴露,不给予打击及治疗;损伤组采用Allens打击法建立SD大鼠脊髓损伤模型;白藜芦醇组模型建立后给予白藜芦醇200mg/kg腹腔注射。术后24、48、72h采用ELISA法检测各组脊髓组织白介素1β(IL-1β)、白介素10(IL-10)、肿瘤坏死因子α(TNF-α)含量;比色法测定髓过氧化物酶(MPO)活性;免疫组化及Western blot法检测术后72h脊髓组织IL-1β、IL-10、TNF-α、MPO蛋白表达的变化。结果 24、48、72h各时间点白藜芦醇组较损伤组IL-1β、IL-10、TNF-α表达及MPO活性降低(P<0.05);72h免疫组化染色及Western blot检测显示,白藜芦醇组较损伤组TNF-α、IL-1β、IL-10及MPO的表达明显降低(P<0.05)。结论白藜芦醇可通过降低大鼠脊髓损伤后脊髓组织中炎性介质活性发挥对脊髓损伤的保护作用。 | 刘长江 毕文超 张晨 时志斌 王坤正 | 2013 | 西安交通大学学报(医学版)2013,34,6: | 12 |
| 3 | 白藜芦醇对大鼠急性脊髓损伤后MPO和NOS活性的影响显示文摘为探讨白藜芦醇(Resveratrol,Res)对大鼠急性脊髓损伤后脊髓组织中髓过氧化物酶(MPO)和一氧化氮合酶(NOS)活性的影响,将120只SD大鼠随机区组分成4组:假手术组、损伤组、白藜芦醇(Res)组和甲基强的松龙(MPSS)组。假手术组切除椎板但不损伤脊髓,术后腹腔注射等量生理盐水,其余三组采用Allen's法以10g×5cm制作大鼠脊髓损伤模型,术后立即分别给予等量生理盐水Res(100mg/kg)和MPSS(100mg/kg)腹腔注射。术后8h、12h、24h、72h、168h取损伤脊髓标本,利用分光光度计测定MPO,NOS吸光度来检测其活性变化。结果SCI后脊髓组织中各时间点MPO和NOS活性均增加(P<0.01),损伤后3d达高峰;Res组和MPSS组在伤后各时间点MPO和NOS活性均小于损伤组(P<0.05);Res组和MPSS组比较,72h、168h时间点MPO活性无统计学意义;24h、72h、168h时间点NOS活性无统计学意义。说明白藜芦醇能抑制脊髓损伤后中性粒细胞浸润,抑制NOS的活性,减轻继发性脊髓损伤,对SCI具有保护和治疗作用。 | 赵训明 刘世清 梅红军 张弩 马永刚 邓明 | 2010 | 科学技术与工程2010,10,14: | 4 |
| 4 | 桑叶与菊花配伍退热机制实验研究显示文摘目的:研究桑叶与菊花配伍对发热大鼠解热、抗炎等方面的影响。方法:将64只大鼠随机分为空白组、模型组、阿司匹林组、桑叶组、菊花组、桑叶菊花5∶2组、桑叶菊花2∶1组、桑叶菊花1∶1组。除空白组外,其余各组采用15%干酵母混悬液背部皮下注射法建立发热大鼠模型,观察桑叶、菊花以及两药不同配伍比例的中药煎剂对发热大鼠的体温及血清中白细胞介素-1β(IL-1β)、一氧化氮(NO)、髓过氧化物酶(MPO)含量的影响。结果:给药后2h,除桑叶组外各药物组大鼠体温均有明显的降低(P<0.01)。给药后4h除桑叶组、菊花组外各药物组大鼠体温均有明显的降低(P<0.01)。与空白组比较,各组大鼠体内IL-1β含量均有明显的升高(P<0.01)。与模型组比较,各药物组大鼠体内IL-1β含量均有明显的降低(P<0.01),与空白组比较,各组大鼠体内NO含量均有明显的升高(P<0.01)。与模型组比较,各药物组大鼠体内NO含量除桑叶组、菊花组均有明显的降低。与空白组比较,各组大鼠体内MPO含量均有明显的升高(P<0.01)。与模型组比较,各药物组大鼠体内MPO含量均有明显的降低(P<0.01),以阿司匹林组最为显著,其次为桑叶菊花1∶1组,再次为桑叶菊花5∶2组、桑叶菊花2∶1组、桑叶组、菊花组。结论:桑叶与菊花以及不同配伍比例的中药煎剂在降低大鼠体温及血清中IL-1β、NO、MPO的含量均具有一定的协同作用,其中发挥解热、抗炎等作用的最佳配伍比例是桑叶、菊花1∶1。 | 秦雪娟 马成 | 2013 | 新中医2013,45,11: | 3 |
| 5 | 白藜芦醇灌胃给药对大鼠脊髓损伤炎症反应中MPO和CCO的作用显示文摘目的探讨白藜芦醇灌胃(管饲)后对脊髓损伤后炎症反应中的髓过氧化物酶和细胞色素C氧化酶活性的影响。方法 2008年2月至2012年2月在武汉大学人民医院动物实验中心将96只Sprague-Dawley健康成年雄性大鼠随机分成4组,根据Allen’s法制成中度脊髓损伤模型,术后按照100 mg/kg立即管饲白藜芦醇或甲基强的松龙;通过比色法观察脊髓损伤8 h,1d,3 d及7 d后白藜芦醇组脊髓髓过氧化物酶和细胞色素C氧化酶活性的变化,并与甲基强的松龙组进行疗效对比。结果在脊髓损伤管饲后8 h,1 d,3 d及7 d白藜芦醇组髓过氧化物酶和细胞色素C氧化酶活性与损伤组统计学上具有显著性差异(P<0.01),显示白藜芦醇管饲后对脊髓损伤具有明显神经细胞保护作用,而且损伤后1d白藜芦醇组与甲基强的松龙组间统计学上具有显著性差异(P<0.01);结论在脊髓损伤后管饲白藜芦醇,能够有效降低髓过氧化物酶活性的升高幅度,并提高细胞色素C氧化酶活性,对损伤后脊髓起保护作用。 | 梅红军 刘洋 张卉 | 2014 | 中国骨质疏松杂志2014,20,6: | 2 |
| 6 | 乙型肝炎肝硬化患者不同临床时期血清sCR1含量变化及意义显示文摘目的研究乙型肝炎肝硬化患者代偿期和失代偿期血清sCR1含量变化及临床意义。方法选择104例代偿期、54例失代偿期乙型肝炎肝硬化患者作为研究对象,选择肝脏功能正常的40例慢性乙型肝炎患者为疾病对照组,50例健康体检人群作为正常对照组,分别采用酶联免疫定量检测试剂盒测定研究对象血清sCR1含量,血清胆碱酯酶(CHE)采用AU5400检测,血浆凝血酶原活动度(PTA)采用ACL9000测定。结果失代偿期乙型肝炎肝硬化患者血清sCR1含量[(122.4±40.9)ng/ml]显著高于代偿期肝硬化患者[(66.3±23.2)ng/ml,P<0.01];不同时期的肝硬化患者血清sCR1含量均显著高于慢性乙型肝炎患者疾病对照组[(22.1±9.5)ng/ml]和正常人群对照组[(19.3±6.6)ng/ml,P<0.01]。失代偿期肝硬化患者血清sCR1的含量变化与CHE和PTA的变化明显负相关(r=-0.82和r=-0.80,P<0.01),但肝脏功能正常的慢性乙型肝炎患者血清sCR1含量变化与正常人群比较无明显差异(P>0.05)。结论乙型肝炎后肝硬化患者血清sCR1含量变化与肝脏功能损伤程度关系密切,可作为临床评估肝脏功能损伤的参考指标。 | 庞君丽 朱剑功 李筱涵 马洪滨 李永利 刘立明 王雪飞 王海滨 | 2012 | 中华临床医师杂志(电子版)2012,6,23: | 0 |
| 7 | Effects of transplantation of microencapsulated rabbit sciatic nerve on nuclear factor-kappa B expression after spinal cord injury in rats显示文摘BACKGROUND:It has been reported that nuclear factor-kappa B(NF-κB),activated after spinal cord injury in rats,plays a key role in inflammatory responses in the central nervous system. OBJECTIVE:To investigate the effects of transplantation of microencapsulated rabbit sciatic nerve on NF-κB expression and motor function after spinal cord injury in rats,and to compare the results with the transplantation of rabbit sciatic nerve alone. DESIGN,TIME AND SETTING:This completely randomized,controlled study was performed at the Department of Neurobiology,Medical College of Nanchang University between December 2007 and July 2008. MATERIALS:A rabbit anti-NF-κB P65 monoclonal antibody was made by the Santa Cruz Company,USA and a streptavidin peroxidase immunohistochemical kit was provided by the Sequoia Company,China. METHODS:Eight rabbits were used to prepare a sciatic nerve cell suspension that was divided into two parts:one stored for transplantation,and the other mixed with a 1.5%sodium alginate solution.One hundred and twenty adult Sprague Dawley rats weighing 220-250 g were randomly divided into four groups:the microencapsulated cell group(n = 36),the non-encapsulated cell group(n = 36),the saline group(n = 36) and the sham operation group(n = 12).The first three groups underwent a right hemisection injury of the spinal cord at the T_(10) level,into which was transplanted a gelatin sponge soaked with 10μL of a microencapsulated nerve tissue/cell suspension(microencapsulated cell group),a tissue/cell suspension(non-encapsulated cell group) or physiological saline(saline group).In the sham operation group the vertebrae were exposed,but the spinal cord was not injured,and no implantation was given. MAIN OUTCOME MEASURES:Pathological changes were detected using hematoxylin-eosin staining;NF-κB expression was quantified using immunohistochemical staining;motor function was assessed using the Basso,Beattie and Bresnahan(BBB) scale. RESULTS:Spinal cord injuries,such as neuronal death and inflammatory cell infiltration,were found in the microencapsulated cell group,the non-encapsulated cell group and the saline group. However,the damage in the microencapsulated cell group was milder than in the non-encapsulated cell or saline groups.NF-κB expression in the microencapsulated cell group, the non-encapsulated cell group and the saline group was increased after spinal cord injury;it reached a peak after 24 hours,gradually decreased after 3 days,and was close to normal levels after 7 days.NF-κB expression in the microencapsulated cell group was significantly lower than in the saline group and the non-encapsulated cell group(P< 0.05).With time,the motor function of the animals in each group improved to a certain extent,but did not reach normal levels.There were no significant differences in BBB scores between the different groups on post-operative day 3;however,the BBB scores for the microencapsulated cell group and the non-encapsulated cell group were significantly higher than the saline group on post-operative day 7(P < 0.05).In addition,the motor function recovered better in the microencapsulated cell group than in the non-encapsulated cell group(P< 0.05).
