2Gas Hydrates in the Qilian Mountain Permafrost, Qinghai, Northwest China显示文摘Qilian Mountain permafrost, with area about 10×104 km2, locates in the north of Qinghai-Tibet plateau. It equips with perfect conditions and has great prospecting potential for gas hydrate. The Scientific Drilling Project of Gas Hydrate in Qilian Mountain permafrost, which locates in Juhugeng of Muri Coalfield, Tianjun County, Qinghai Province, has been implemented by China Geological Survey in 2008–2009. Four scientific drilling wells have been completed with a total footage of 2059.13 m. Samples of gas hydrate are collected separately from holes DK-1, DK-2 and DK-3. Gas hydrate is hosted under permafrost zone in the 133–396 m interval. The sample is white crystal and easily burning. Anomaly low temperature has been identified by the infrared camera. The gas hydrate-bearing cores strongly bubble in the water. Gas-bubble and water-drop are emitted from the hydrate-bearing cores and then characteristic of honeycombed structure is left. The typical spectrum curve of gas hydrate is detected using Raman spectrometry. Furthermore, the logging profile also indicates high electrical resistivity and sonic velocity. 3Genome Editing Enables Next-Generation Hybrid Seed Production Technology显示文摘The next-generation hybrid seed technology enables the successful production of sortable hybrid seeds from genic male sterile(GMS)lines and maintainers;however,it requires multiple laborious and complicated steps.Here,we designed a simple next-generation hybrid seed production strategy that takes advantage of the CRISPR/Cas9 technology to create a Manipulated GMS Maintainer(MGM)system via a single transformation.Under this schema,the maize male fertility gene ZmMS26 was nullified by removal of its fifth exon using the CRISPR/Cas9 system on a vector,and a second vector carrying a functional ZmMS26 cDNA was co-transformed to restore fertility.The second vector also contains a male gametophyte inactivation gene(ZmAA1)encoding maizeα-amylase driven by the pollen-specific promoter PG47 and an endosperm fluorescent marker(DsRED)driven by the barley endosperm aleurone-specific promoter Ltp2.The derived single-copy hemizygous MGM lines bore a mutated MS26 gene,leading to complete male sterility but normal vegetative growth and grain yield.The MGM system could prevent genetic transmission of the MGM elements via male gametophytes,providing an efficient method for sorting maintainer seeds labeled by DsRED.This strategy can be extended to any GMS gene and to hybrid crops other than maize.Xiantao Qi Congsheng Zhang Jinjie Zhu Changlin Liu Changling Huang Xinhai Li Chuanxiao Xie 2020Molecular Plant2020,13,9:18
4Testing method and distribution characteristics of spatial pesticide spraying deposition quality balance for unmanned aerial vehicle显示文摘In order to explore the spatial pesticide spraying deposition distribution,the downwash flow field characteristics for unmanned aerial vehicle(UAV)pesticide application with accurate flight height and velocity and the relationship of these two aspects,a novel measurement method was proposed in this paper.A model‘3WQF80-10’single-rotor diesel UAV was tested using this method in wheat field and the effects of flight direction,flight parameters and crosswind on the distribution of spatial spraying deposition quality balance(SSDQB)and the downwash flow field distribution were researched.A cuboid aluminum sampling frame of spatial spraying deposition quality balance(SFSSDQB)with monofilament wires was made for collecting the droplets in four directions,and a set of multi-channel micro-meteorology measurement system(MMMS)was applied for measuring the downwash wind speed in three directions.Besides,BeiDou Navigation Satellite System(BNSS)was used for controlling and recording the working height,velocity and track of this model of single-rotor UAV.The results showed the distribution of the spatial spray deposition and the downwash flow field of UAV could be measured effectively at exact flight height and velocity via this method.When the average wind speed was 0.9 m/s,the average temperature was 31.5℃ and the average relative humidity was 34.1%,and the average distribution ratios of spraying deposition for model‘3WQF80-10’UAV on the upwind part,the top part,the downwind part and the bottom part were 4.4%,2.3%,50.4%and 43.7%,respectively.The flight directions of forward and backward had an impact on droplet deposition distribution and the working effect of flying backwards,with 60%of deposition ratio of the bottom part of the SFSSDQB,was better than flying forward.There was a linear negative correlation between the coefficient of variation(CV)of the bottom part and the flight height and the coefficient of determination was 0.9178,which means that the deposition distribution becomes more uniform with the increase of height.Additionally,there was a linear positive correlation between weighted mean deposition rate and crosswind speed and the coefficient of determination was 0.9684,which shows the deposition distribution gets more concentrated towards the downwind part with the rise of crosswind speed.Therefore,according to the results of tests of downwash airflow speed,it is shown that regardless of the flight direction and height and the crosswind,all these factors influence the droplet deposition distribution via weakening the intensity of the downwash airflow field in the direction perpendicular to the ground.The results can provide valuable information for the research of UAV pesticide application techniques and the establishment of the standard of spraying deposition and drift tests of UAV in crop field.Changling Wang Xiongkui He Xiaonan Wang Zhichong Wang Shilin Wang Longlong Li Jane Bonds Andreas Herbst Zhiguo Wang 2018International Journal of Agricultural and Biological Engineering2018,11,2:14
5Undrained shear strength evaluation for hydrate-bearing sediment overlying strata in the Shenhu area, northern South China Sea显示文摘The undrained shear strength of shallow strata is a critical parameter for safety design in deep-water operations.In situ piezocone penetration tests(CPTU) and laboratory experiments are performed at Site W18-19 in the Shenhu area, northern South China Sea, where China's first marine hydrate exploitation operation is due to be located. The validation of the undrained shear strength prediction model based on CPTU parameters. Different laboratory tests, including pocket penetrometer, torvane, miniature vane and unconsolidated undrained triaxial tests, are employed to solve empirical cone coefficients by statistical and mathematical methods. Finally, an optimized model is proposed to describe the longitudinal distribution of undrained shear strength in calcareous clay strata in the Shenhu area. Research results reveal that average empirical cone coefficients based on total cone resistance, effective resistance, and excess-pore pressure are 13.8, 4.2 and 14.4, respectively. The undrained shear strength prediction model shows a good fit with the laboratory results only within specific intervals based on their compaction degree and gas-bearing conditions. The optimized prediction model in piecewise function format can be used to describe the longitudinal distribution of the undrained shear strength for calcareous clay within all depth intervals from the mud-line to the upper boundary of hydrate-bearing sediments(HBS). The optimized prediction result indicates that the effective cone resistance model is suitable for very soft to firm calcareous clays,the excess-pore pressure model can depict the undrained shear strength for firm to very stiff but gas-free clays,while the total cone resistance model is advantageous for evaluating the undrained shear strength for very stiff and gassy clays. The optimized model in piecewise function format can considerably improve the adaptability of empirical models for calcareous clay in the Shenhu area. 6Cooperative effects of hydrogen sulfide and nitric oxide on delaying softening and decay of strawberry显示文摘Hydrogen sulfide(H2S)and nitric oxide(NO)as two signal molecules play prominent roles in plant response to biotic stress.The combination effects of these two molecules on ripening and decay of postharvest strawberry were investigated.Strawberries were first dipped in distilled water,0.8 mmol/L NaHS solution,5μmmol/L sodium nitroprusside(SNP)solution,and combinative solutions of 0.8 mmol/L NaHS and 5μmmol/L SNP for 10 min,respectively,and then stored at 20°C.Results showed that the effects of the co-treatment of 0.8 mmol/L NaHS solution and 5μmmol/L SNP solution were superior to that of other treatments.The former could suppress the percentage of fruit decay,inhibit respiration rate,maintain the original crust color,and slow down the decrease of firmness and the change of relative conductivity,which preserved the fruit quality.Further investigation showed that co-treatment of hydrogen sulfide and nitric oxide maintained higher activities of chitinase(CHI)and beta-1,3-glucanase(GNS),and lowered activities of pectin methylesterase(PME),polygalacturonase(PG)and endo-β-1,4-glucanase(EGase).The study indicated that the synergistic function of H2S and NO could effectively prolong the shelf life and reduce the decay rate of harvested strawberry.Zhang Chang Shi Jingying Zhu Liqin Li Changle Wang Qingguo 2014International Journal of Agricultural and Biological Engineering2014,7,6:12

8Precise base editing of non-allelic acetolactate synthase genes confers sulfonylurea herbicide resistance in maize显示文摘Single-nucleotide polymorphisms contribute to phenotypic diversity in maize. Creation and functional annotation of point mutations has been limited by the low efficiency of conventional methods based on random mutation. An efficient tool for generating targeted single-base mutations is desirable for both functional genomics and precise genetic improvement. The objective of this study was to test the efficiency of targeted C-to-T base editing of two non-allelic acetolactate synthase(ALS) in generating sulfonylurea herbicide-resistant mutants. A CRISPR/Cas9 nickase-cytidine deaminase fused with uracil DNA glycosylase inhibitor(UGI) was employed to achieve targeted conversion of cytosine to thymine in ZmALS1 and ZmALS2. Both protoplasts and recovered mutant plants showed the activity of the cytosine base editor, with an in vivo efficiency of up to 13.8%. Transgene-free edited plants harboring a homozygous ZmALS1 mutation or a ZmALS1 and ZmALS2 double mutation were tested for their resistance at a dose of up to 15-fold the recommended limit of chlorsulfuron, a sulfonylurea herbicide widely used in agriculture. 9Study of the Surface Morphology of Gas Hydrate显示文摘Study of the surface morphology of gas hydrate is of great importance in understanding its physical properties and occurrence.In order to investigate the surface morphology of different types(sI and sII)and occurrences(pore-filling and fracture-filling)of gas hydrate,both lab-synthesized and drilled-gas hydrate samples were measured using cryo-scanning electron microscopy(cryo-SEM).Results showed that the surface of s I hydrate was relatively smooth,and spongy nano-pores(200–400 nm)gradually occurred at the surface during continuous observation.The surface of sII hydrate was more compact,showing a tier-like structure.Hydrate occurred in quartz sand and usually filled the pores of the sediments and both hydrate and sediments were cemented with each other.SEM observation of the gas hydrates collected from the South China Sea showed that the surface morphology and contact relation with sediments varied with hydrate occurrence.For instance,hydrates dispersed in sediments mainly filled the pores of the sediments.The existence of microorganism shells,such as foraminifera,was beneficial to the formation of gas hydrate.When hydrate occurred as a massive or vein structure,it was easily distinguished from the surrounding sediments.The surface of hydrate with massive or vein structure showed two distinct characters:one was dense and smooth,the other is porous(several to tens of micrometers in diameter).The occurrence of different hydrate morphologies was probably caused by the supplement rates of methane gas.SUN Jianye LI Chengfeng HAO Xiluo LIU Changling CHEN Qiang WANG Daigang 2020Journal of Ocean University of China2020,19,2:9
10Factors affecting genomic selection revealed by empirical evidence in maize显示文摘Genomic selection(GS) as a promising molecular breeding strategy has been widely implemented and evaluated for plant breeding, because it has remarkable superiority in enhancing genetic gain, reducing breeding time and expenditure, and accelerating the breeding process. In this study the factors affecting prediction accuracy(rMG) in GS were evaluated systematically, using six agronomic traits(plant height, ear height, ear length, ear diameter,grain yield per plant and hundred-kernel weight) evaluated in one natural and two biparental populations. The factors examined included marker density, population size, heritability,statistical model, population relationships and the ratio of population size between the training and testing sets, the last being revealed by resampling individuals in different proportions from a population. Prediction accuracy continuously increased as marker density and population size increased and was positively correlated with heritability; rMGshowed a slight gain when the training set increased to three times as large as the testing set. Low predictive performance between unrelated populations could be attributed to different allele frequencies, and predictive ability and prediction accuracy could be improved by including more related lines in the training population. Among the seven statistical models examined, including ridge regression best linear unbiased prediction(RR-BLUP), genomic BLUP(GBLUP), Bayes A, Bayes B, Bayes C, Bayesian least absolute shrinkage and selection operator(Bayesian LASSO), and reproducing kernel Hilbert space(RKHS), the RKHS and additive-dominance model(Add + Dom model) showed credible ability for capturing non-additive effects, particularly for complex traits with low heritability. 11QTL analysis of ear leaf traits in maize(Zea mays L.)under different planting densities显示文摘Modem maize varieties have become more productive than ever, owing largely to increased tolerance of high plant density. However, the genetics of ear leaf traits under different densities remains poorly understood. In this study, Zhongdan 909 recombinant inbred lines(RILs) derived from a cross between Z58 and HD568 were genotyped for 3072 single-nucleotide polymorphisms(SNPs), and phenotyped for leaf length(LL), leaf width(LW), and leaf angle(LA) of the uppermost ear leaf under three planting densities(52,500,67,500, and 82,500 plants ha-1, respectively). A genetic map was then constructed using1358 high-quality SNPs. The total length of the linkage map was 1985.2 cM and the average interval between adjacent markers 1.46 cM. With increasing density, LL and LW decreased from 63.68 to 63.02 cm and from 8.56 to 8.21 cm, respectively, while LA increased from19.42° to 19.66°. All three traits had high heritabilities, of 0.75, 0.78, and 0.84, respectively.Using inclusive composite interval mapping, 23, 25, and 17 quantitative trait loci(QTL) were detected for LL, LW, and LA, respectively. Of these, 35 were simultaneously detected under two or three plant densities, while 30 were detected under only one. 12Influence of foraminifera on formation and occurrence characteristics of natural gas hydrates in fine-grained sediments from Shenhu area, South China Sea显示文摘Marine gas hydrates accumulate primarily in coarse-grained, high-permeability layers; however, highly saturated natural gas hydrates have been discovered in the fine-grained sediments of Shenhu area, South China Sea(SCS). This may be explained by key factors, such as the great abundance of foraminifera shells. In this paper, by analyzing the SCS foraminifera structure and performing hydrate formation experiments in the foraminifera shells, the contribution of foraminifera to hydrate accumulation in the SCS was investigated from a microscopic point of view. Simulations of hydrate formation were carried out in both pure SCS foraminifera shells and the host sediments. Pore structures in typical foraminifera were studied by use of micro-focus X-ray computed tomography(CT) and scanning electron microscopy(SEM). Hydrate growth and occurrence characteristics in the foraminifera shells were observed in-situ. The results showed that the presence of foraminifera significantly enhanced the effective porosity of the SCS sediments. Moreover, while the hydrates grew preferentially in the chambers of the coarse-grained foraminifera by adhering to the inner walls of the foraminifera shells, no apparent hydrate accumulation was observed in the fine-grained or argillaceous matrix. 14Insulin-like growth factor 2 as a candidate gene influencing growth and carcass traits and its bialleleic expression in chicken显示文摘We have identified DNA polymorphisms in the gene of insulin-like growth factor 2 by PCR-SSCP in a resource population, which was generated by Silky reciprocally crossing to Broilers. A C→G mutation was detected in the exon 2 (at position 71) by sequencing. This single nucleotide polymorphism (SNP) was found to be associated with production traits. Chicken with BB genotype showed more chest angle width but less 3 week body weight and glandular stom- ach weight than chicken with AA genotype (P<0.05); while the heterozygote (AB genotype) chicken had more abdominal fat weight, eviscerated yield with giblet than AA homozygote chicken. Further analysis showed that there were different genetic effects on some traits between heterozygote AB (paternal allele given first) and heterozygote BA: chickens with genotype BA had more birth weight and breast weight but less abdominal fat weight than chickens with geno- type AB (P<0.05), which could be hypothetically contributed by genome imprinting. Therefore, Silky chickens were selected for production of heterozygotes to confirm whether IGF2 locus was imprinting. Progeny from heterozygote × homozygote reciprocal cross was assayed for ex- pression after the genotype was determined. The transcription of IGF2 was detected by RT-PCR-SSCP. IGF2 gene was expressed bialleleically in 1-day-old neonatal liver and 90-day-old liver, kidney, heart, and muscle of both heterozygote AB and BA chickens. Therefore, IGF2 was not an imprinting gene in chicken. 15Microstructure and properties of indium tin oxide thin films deposited by RF-magnetron sputtering显示文摘Tin-doped indium oxide (ITO) thin films were prepared using conventional radio frequency (RF) planar magne-tron sputtering equipped with IR irradiation using a ceramic target of In2O3/SnO2 with a mass ratio of 1:1 at various IR irradiation temperatures TI (from room temperature to 400°C). The refractive index, deposited ratio, and resistivity are functions of the sputtering Ar gas pressure. The microstructure of ITO thin films is related to IR TI, the crystalline seeds appear at TI = 300°C, and the films are amorphous at the temperature ranging from 27°C to 400°C. AFM investigation shows that the roughness value of peak-valley of ITO thin film (Rp-v) and the surface microstructure of ITO thin films have a close relation with TI. The IR irradiation results in a widening value of band-gap energy due to Burstein-Moss effect and the maximum visible transmittance shifts toward a shorter wavelength along with a decrease in the film’s refractive index. The plasma wavelength and the refractive index of ITO thin films are relative to the TI. XPS investigation shows that the photoelectrolytic properties can be deteriorated by the sub-oxides. 18A phase 3,double-blind,randomized placebo-controlled efficacy and safety study of abiraterone acetate in chemotherapynaı¨ve patients with mCRPC in China,Malaysia,Thailand and Russia显示文摘Objective:This double-blind,placebo-controlled phase 3 study was designed to compare efficacy and safety of abiraterone acetate+prednisone(abiraterone)to prednisone alone in chemotherapy-naı¨ve,asymptomatic or mildly symptomatic metastatic castrationresistant prostate cancer(mCRPC)patients from China,Malaysia,Thailand and Russia.Methods:Adult chemotherapy-naı¨ve patients with confirmed prostate adenocarcinoma,Eastern Cooperative Oncology Group(ECOG)performance status(PS)grade 0e1,ongoing androgen deprivation(serum testosterone<50 ng/dL)with prostate specific antigen(PSA)or radiographic progression were randomized to receive abiraterone acetate(1000 mg,QD)t prednisone(5 mg,BID)or placebo t prednisone(5 mg,BID),until disease progression,unacceptable toxicity or consent withdrawal.Primary endpoint was improvements in time to PSA progression(TTPP).Results:Totally,313 patients were randomized(abiraterone:n Z 157;prednisone:n Z 156);and baseline characteristics were balanced.At clinical cut-off(median follow-up time:3.9 months),80% patients received treatment(abiraterone:n Z 138,prednisone:n Z 112).Median time to PSA progression was not reached with abiraterone versus 3.8 months for prednisone,attaining 58%reduction in PSA progression risk(HR=0.418;p<0.0001).Abirateronetreated patients had higher confirmed PSA response rate(50%vs.21%;relative odds=2.4;p<0.0001)and were 5 times more likely to achieve radiographic response than prednisonetreated patients(22.9%vs.4.8%,p=0.0369).Median survival was not reached.Most common(≥10% abiraterone vs.prednisone-treated)adverse events:bone pain(7%vs.14%),pain in extremity(6%vs.12%),arthralgia(10%vs.8%),back pain(7%vs.11%),and hypertension(15%vs.14%).Conclusion:Interim analysis confirmed favorable benefit-to-risk ratio of abiraterone in chemotherapy-naı¨ve men with mCRPC,consistent with global study,thus supporting use of abiraterone in this patient population.Dingwei Ye Yiran Huang Fangjian Zhou Keji Xie Vsevolod Matveev Changling Li Boris Alexeev Ye Tian Mingxing Qiu Hanzhong Li Tie Zhou Peter De Porre Margaret Yu Vahid Naini Hongchuan Liang Zhuli Wu Yinghao Sun 2017Asian Journal of Urology2017,4,2:3
19Source Recovery in Underdetermined Blind Source Separation Based on Artificial Neural Network显示文摘We propose a novel source recovery algorithm for underdetermined blind source separation, which can result in better accuracy and lower computational cost. On the basis of the model of underdetermined blind source separation, the artificial neural network with single-layer perceptron is introduced into the proposed algorithm. Source signals are regarded as the weight vector of single-layer perceptron, and approximate ι~0-norm is taken into account for output error decision rule of the perceptron, which leads to the sparse recovery. Then the procedure of source recovery is adjusting the weight vector of the perceptron. What's more, the optimal learning factor is calculated and a descent sequence of smoothed parameter is used during iteration, which improves the performance and significantly decreases computational complexity of the proposed algorithm. 20BROADCAST SCHEDULING WITH MIMO LINKS IN MULTI-HOP AD HOC NETWORKS显示文摘As the current medium access control protocols with Multiple Input Multiple Output (MIMO) links only bear point to point service, broadcast scheduling algorithm in ad hoc networks with MIMO links is proposed. The key to the proposed broadcast scheduling algorithm is the time slot scheduling algorithm which guarantees collision-free transmissions for every node and the mini- mum frame length. The proposed algorithm increases the simultaneous transmissions of MIMO links efficiently. Due to the interference null capacity of MIMO links, the interference node set of each node can decrease from two-hop neighbors to one-hop neighbors possibly.
