3Identification and Fine Mapping of a Gene Related to Pale Green Leaf Phenotype near the Centromere Region in Rice(Oryza sativa)显示文摘A thermo-insensitive pale green leaf mutant (pgl2) was isolated from T-DNA inserted transgenic lines of rice (Oryza sativa L. subsp. japonica cv. Nipponbare). Genetic analysis indicated that the phenotype was caused by a recessive mutation in a single nuclear-encoded gene. To map the PGL2 gene, an F2 population was constructed by crossing the mutant with Longtefu (Oryza sativa L. subsp. indica). The PGL2 locus was roughly linked to SSR marker RM331 on chromosome 8. To finely map the gene, 14 new InDel markers were developed around the marker, and PGL2 was further mapped to a 2.37 Mb centromeric region. Analysis on chlorophyll contents of leaves showed that there was no obvious difference between the mutant and the wild type in total chlorophyll (Chl) content, while the ratio of Chl a / Chl b in the mutant was only about 1, which was distinctly lower than that in the wild type, suggesting that the PGL2 gene was related to the conversion between Chl a and Chl b. 4Oxidative Stress and Apoptotic Changes of Rat Cerebral Cortical Neurons Exposed to Cadmium in Vitro显示文摘Objective To investigate the cytotoxic mechanism of cadmium(Cd) on cerebral cortical neurons.Methods The primary cultures of rat cerebral cortical neurons were treated with different concentrations of cadmium acetate(0,5,10,and 20 μmol/L),and then the cell viability,apoptosis,ultrastructure,intracellular [Ca2+]i and reactive oxygen species(ROS) levels,mitochondrial membrane potential(ΔΨ),activities of catalase(CAT) and superoxide dismutase(SOD) were measured.Results A progressive loss in cell viability and an increased number of apoptotic cells were observed.In addition,Cd-induced apoptotic morphological changes in cerebral cortical neurons were also demonstrated by Hoechst 33258 staining.Meanwhile,ultrastructural changes were distortion of mitochondrial cristae and an unusual arrangement.Simultaneously,elevation of intracellular [Ca2+]i and ROS levels,depletion of ΔΨ were revealed in a dose-dependent manner during the exposure.Moreover,CAT and SOD activities in the living cells increased significantly.Conclusion Exposure of cortical neurons to different doses of Cd led to cellular death,mediated by an apoptotic mechanism,and the apoptotic death induced by oxidative stress may be a potential reason.And the disorder of intracellular homeostasis caused by oxidative stress and mitochondrial dysfunction may be a trigger for apoptosis in cortical neurons.YUAN Yan BIAN Jian Chun LIU Xue Zhong ZHANG Ying SUN Ya LIU Zong Ping 2012Biomedical and Environmental Sciences2012,25,2:10
5Cadmium Activates Reactive Oxygen Species-dependent AKT/mT OR and Mitochondrial Apoptotic Pathways in Neuronal Cells显示文摘Objective To examine the role of Cd-induced reactive oxygen species(ROS)generation in the apoptosis of neuronal cells.Methods Neuronal cells(primary rat cerebral cortical neurons and PC12 cells)were incubated with or without Cd post-pretreatment with rapamycin(Rap)or N-acetyl-L-cysteine(NAC).Cell viability was determined by MTT assay,apoptosis was examined using flow cytometry and fluorescence microscopy,and the activation of phosphoinositide 3'-kinase/protein kinase B(Akt)/mammalian target of rapamycin(m TOR)and mitochondrial apoptotic pathways were measured by western blotting or immunofluorescence assays.Results Cd-induced activation of Akt/m TOR signaling,including Akt,m TOR,p70 S6 kinase(p70 S6K),and eukaryotic initiation factor 4E binding protein 1(4E-BP1).Rap,an m TOR inhibitor and NAC,a ROS scavenger,blocked Cd-induced activation of Akt/m TOR signaling and apoptosis of neuronal cells.Furthermore,NAC blocked the decrease of B-cell lymphoma 2/Bcl-2 associated X protein(Bcl-2/Bax)ratio,release of cytochrome c,cleavage of caspase-3 and poly(ADP-ribose)polymerase(PARP),and nuclear translocation of apoptosis-inducing factor(AIF)and endonuclease G(Endo G).Conclusion Cd-induced ROS generation activates Akt/m TOR and mitochondrial pathways,leading to apoptosis of neuronal cells.Our findings suggest that m TOR inhibitors or antioxidants have potential for preventing Cd-induced neurodegenerative diseases.YUAN Yan WANG Yi HU Fei Fei JIANG Chen Yang ZHANG Ya Jing YANG Jin Long ZHAO Shi Wen GU Jian Hong LIU Xue Zhong BIAN Jian Chun LIU Zong Ping 2016Biomedical and Environmental Sciences2016,29,2:8

7Leaf photosynthesis and yield components of mung bean under fully open-air elevated[CO_2]显示文摘Mung bean(Vigna radiata L.) has the potential to establish symbiosis with rhizobia,and symbiotic association of soil micro flora may facilitate the photosynthesis and plant growth response to elevated[CO_2].Mung bean was grown at either ambient CO_2 400 μmol mol^(-1) or[CO_2]((550+17) μmol mol^(-1)) under free air carbon dioxide enrichment(FACE) experimental facility in North China.Elevated[CO_2]increased net photosynthetic rate(P_n),water use efficiency(WUE) and the non-photochemical quenching(NPQ) of upper most fully-expanded leaves,but decreased stomatal conductance(G_s),intrinsic efficiency of PSII(F_v '/F_m'),quantum yield of PSII(φ_(PSll)) and proportion of open PSII reaction centers(q_p).At elevated[CO_2],the decrease of F_v'/F_m',φ_(PSII),q_p at the bloom stage were smaller than that at the pod stage.On the other hand,P_n was increased at elevated[CO_2]by 18.7 and 7.4%at full bloom(R2) and pod maturity stages(R4),respectively.From these findings,we concluded that as a legume despite greater nutrient supply to the carbon assimilation at elevated[CO_2],photosynthetic capacity of mung bean was still suppressed under elevated[CO_2]particularly at pod maturity stage but plant biomass and yield was increased by 11.6 and 14.2%,respectively.Further,these findings suggest that even under higher nutrient acquisition systems such as legumes,nutrient assimilation does not match carbon assimilation under elevated[CO_2]and leads photosynthesis down-regulation to elevated[CO_2].GAO Ji HAN Xue Saman Seneweera LI Ping ZONG Yu-zheng DONG Qi LIN Er-da HAO Xing-yu 2015Journal of Integrative Agriculture2015,14,5:6
8Blockade of HCN2 Channels Provides Neuroprotection Against Ischemic Injury via Accelerating Autophagic Degradation in Hippocampal Neurons显示文摘In the central nervous system,hyperpolarizationactivated cyclic nucleotide-gated(HCN)channels are essential to maintain normal neuronal function.Recent studies have shown that HCN channels may be involved in the pathological process of ischemic brain injury,but the mechanisms remain unclear.Autophagy is activated in cerebral ischemia,but its role in cell death/survival remains controversial.In this study,our results showed that the HCN channel blocker ZD7288 remarkably decreased the percentage of apoptotic neurons and corrected the excessive autophagy induced by oxygen-glucose deprivation followed by reperfusion(OGD/R)in hippocampal HT22 neurons.Furthermore,in the OGD/R group,p-mTOR,p-ULK1(Ser757),and p62 were significantly decreased,while p-ULK1(Ser317),atg5,and beclin1 were remarkably increased.ZD7288 did not change the expression of p-ULK1(Ser757),ULK1(Ser317),p62,Beclin1,and atg5,which are involved in regulating autophagosome formation.Besides,we found that OGD/R induced a significant increase in Cathepsin D expression,but not LAMP-1.Treatment with ZD7288 at 10μmol/L in the OGD/R group did not change the expression of cathepsin D and LAMP-1.However,chloroquine(CQ),which decreases autophagosome-lysosome fusion,eliminated the correction of excessive autophagy and neuroprotection by ZD7288.Besides,shRNA knockdown of HCN2 channels significantly reduced the accumulation of LC3-Ⅱand increased neuron survival in the OGD/R and transient global cerebral ischemia(TGCI)models,and CQ also eliminated the effects of HCN2-shRNA.Furthermore,we found that the percentage of LC3-positive puncta that co-localized with LAMP-1-positive lysosomes decreased in Con-shRNAtransfected HT22 neurons exposed to OGD/R or CQ.In HCN2-shRNA-transfected HT22 neurons,the percentage of LC3-positive puncta that co-localized with LAMP-1-positive lysosomes increased under OGD/R;however,the percentage was significantly decreased by the addition of CQ to HCN2-shRNA-transfected HT22 neurons.The present results demonstrated that blockade of HCN2 channels provides neuroprotection against OGD/R and TGCI by accelerating autophagic degradation attributable to the promotion of autophagosome and lysosome fusion.Cheng Chen Li Liu Ya-Qiao Shu Ping Jing Yun Lu Xiao-Xue Zhang Xian-Gang Zong Lian-Jun Guo Chang-Jun Li 2020Neuroscience Bulletin2020,36,8:6
9Autophagy Attenuates MnCl2-induced Apoptosis in Human Bronchial Epithelial Cells显示文摘Objective To investigate the role of autophagy in MnC l2-induced apoptosis in human bronchial epithelial 16 HBE cells.Methods Cell proliferation was measured by MTT assay.Mitochondrial membrane potential(MMP) and apoptosis were measured by flow cytometry.Autophagic vacuoles were detected by fluorescence microscopy.Cellular levels of apoptosis and autophagy-related proteins were measured by western blotting.Results 16 HBE cell proliferation was inhibited by Mn Cl2 in a dose-and time-dependent manner.Mn Cl2-induced 16 HBE cell growth inhibition was related to MMP depolarization prior to the induction of apoptosis.Our data revealed that Mn Cl2-induced apoptosis in 16 HBE cells was mediated by decreased expression of Bcl-2 and increased levels of cleaved caspase-3.It was observed that when we exposed 16 HBE cells to MnCl_2 in a dose-dependent manner,the formation of autophagic vacuoles and the levels of LC-3B-II were elevated.RNA interference of LC3 B in these Mn Cl2-exposed cells demonstrated that MMP loss and apoptosis were enhanced.Additionally,the pan-caspase inhibitor Z-VAD-FMK increased the cellular levels of Bcl-2 and decreased apoptosis,but did not affect the cellular levels of LC3 B in Mn Cl2-treated 16 HBE cells.Conclusion Mn Cl2 dose-and time-dependently inhibits 16 HBE cell proliferation and induces MMP loss and apoptosis.Autophagy acts in a protective role against Mn Cl2-induced apoptosis in 16 HBE cells.YUAN Zhun YING Xian Ping ZHONG Wei Jian TIAN Shi Min WANG Yu JIA Yong Rui CHEN Wen FU Juan Ling ZHAO Peng ZHOU Zong Can 2016Biomedical and Environmental Sciences2016,29,7:5
10Efficacy evaluation of imatinib treatment in patients with gastrointestinal stromal tumors:A meta-analysis显示文摘AIM:To perform a meta-analysis to derive a more precise estimation of imatinib treatment for different genotypes of gastrointestinal stromal tumors(GIST).METHODS:Studies were identified by searching PubMed and Embase.Inclusive criteria were patients with exon 9-mutant,exon 11-mutant or wide type(WT) GIST,receiving chemotherapy of imatinib for clinical trial,and efficacy evaluation was cumulative response (CR)including complete response and partial response.The odds ratios(OR)for CR in stem cell factor receptor (KIT)mutation patients vs WT genotype patients,KIT exon 11-mutant genotype patients vs KIT exon 9-mutant genotype patients and KIT exon 9-mutant genotype patients vs WT genotype patients were calculated with 95%confidence interval(CI)for each study as an estimation of the efficacy of imatinib.RESULTS:Five studies including 927 patients were involved in this meta-analysis.The overall OR(KIT group vs WT group)was 3.34(95%CI:2.30-4.86,P <0.00001,P heterogeneity=0.04).The overall OR in KIT exon 11 group vs KIT exon 9 group was 3.29(95%CI: 2.17-5.00,P<0.00001,P heterogeneity=0.33).The overall OR in KIT exon 9 group vs WT group was 1.23(95% CI:0.73-2.10,P=0.44,P heterogeneity=0.42).CONCLUSION:Most patients with different genotypes of GIST and KIT exon 11-mutant will benefit from the individualized treatment of imatinib.Ping Chen Liang Zong Wei Zhao Lei Shi 2010World Journal of Gastroenterology2010,16,33:5
11An Adaptive Weighted Differential Game Guidance Law显示文摘For intercepting modern high maneuverable targets, a novel adaptive weighted differential game guidance law based on the game theory of mixed strategy is proposed, combining two guidance laws which are derived from the perfect and imperfect information pattern, respectively. The weights vary according to the estimated error of the target's acceleration, the guidance law is generated by directly using the estimation of target's acceleration when the estimated error is small, and a differential game guidance law with adaptive penalty coefficient is implemented when the estimated error is large. The adaptive penalty coefficients are not constants and they can be adjusted with current target maneuverability. 13Hepatoprotective ability of tetramethylpyrazine produced by Bacillus amyloliquefaciens显示文摘Tetramethylpyrazine(TTMP),an important aroma compound,was produced by Bacillus amyloliquefaciens XJB-104 with distillers’grains as raw material.The yield of TTMP under optimized fermentation procedure was 3176.52 mg/L.TTMP in the fermentation broth was purified and the purity(>99%)was measured by gas chromatography-mass spectrometry.Furthermore,the hepatoprotective activity of TTMP in ethanol-water system was evaluated by biochemical indicators of liver injury,parameters of antioxidant defense system and inflammatory response,and liver histopathological assessment in mice.Ethanol treatment increased serum levels of alanine aminotransferase,aspartate aminotransferase,alkaline phos-phatase and lactate dehydrogenase,suggesting liver damage.Both high and low dose of TTMP significantly decreased levels of these indicators.Furthermore,the reduction in the activities or concentrations of superoxide dismutase,catalase,reduced glutathione and malondialdehyde in liver tissue caused by ethanol was significantly alleviated by TTMP.Increased inflammatory cytokines including transcription factor,tumor necrosis factor,interleukin-1beta,interleukin,macrophage chemoattractant protein,inducible nitric oxide synthase and cyclooxygenase were also suppressed by TTMP treatment.It is concluded that TTMP in ethanol-water system has potential liver-protective activity,especially when ethanol is consumed at low doses for short periods of time.Wenqing Zhang Guanru Si Zhiming Rao Shuai Zong Jinglei Li Xian Zhang Chuanqiang Gao Zhou Ping Ming Ye 2021Systems Microbiology and Biomanufacturing2021,1,2:4
14Correlation between Waist Circumference and Carotid Intima-Media Thickness in Women from Shanghai, China显示文摘Objective To evaluate whether waist circumference (WC) ≥85 cm is related to asymptomatic preclinical atherosclerosis in women from Shanghai, China. Methods A total of 2365 females aged ≥20 years recruited from 4 communities underwent physical examination and carotid artery scanning. Their carotid intima-media thickness (C-IMT) was measured. Results The C-IMT was significantly higher in overweight or obese women with their BMI ≥25.0 kg/m2 (P<0.01) and in those with their WC ≥85 cm than in those with their WC <85 cm (P<0.01). Spearman and partial correlation analysis showed that the C-IMT was significantly correlated with WC which was independent of menopausal status. The C-IMT significantly increased with the increasing WC and reached to a platform in about 85 cm. An increment tendency was found in the subgroup with its WC < 85 cm (P<0.01) while no significant tendency was found in the subgroup with its WC≥85 cm (P=0.07). Multiple stepwise regression analysis showed that the WC was an independent risk factor for C-IMT. In logistic regression model, the odd ratio of WC ≥80 cm, ≥80 cm and <85 cm and ≥85 cm for evaluating the risk of C-IMT elevation was 1.632, 1.501, and 1.878, respectively. 15Onodera's Prognostic Nutritional Index is a novel and useful prognostic marker for gastrointestinal stromal tumors显示文摘BACKGROUND Immunoinflammatory markers such as the peripheral blood neutrophil-tolymphocyte ratio(NLR)and the platelet-to-lymphocyte ratio(PLR)have gained considerable attention as prognostic markers in gastrointestinal stromal tumors(GISTs).AIM To assess the prognostic value of Onodera’s Prognostic Nutritional Index(OPNI)for GISTs.METHODS All patients who had undergone surgical resection for a primary,localized GIST from 2009 to 2016 at our cancer center were initially and retrospectively identified.Recurrence-free survival(RFS)was calculated by the Kaplan-Meier method and compared by the log-rank test.We used multivariate Cox proportional hazard regression models to identify associations with outcome variables.RESULTS A total of 235 GISTs were identified and included for analysis under our inclusion criteria.Univariate and multivariate analyses both identified the OPNI as an independent prognostic marker,and the OPNI was associated with the primary site,tumor size,mitotic index,tumor rupture,necrosis,and modified NIH risk classification.Low OPNI(<51.30;hazard ratio=5.852;95% confidence interval:1.072–31.964;P=0.0414)was associated with worse RFS.The 2-and 5-year RFS rates of the patients with a low OPNI were 92.83% and 76.22%,respectively,whereas 100% and 98.41% were achieved by the patients with a high OPNI.CONCLUSION The preoperative OPNI is a novel and useful prognostic marker for GISTs.Hao Wang Ying-Ying Xu Jun You Wen-Qing Hu Shao-Feng Wang Ping Chen Fan Yang Lei Shi Wei Zhao Liang Zong 2021World Journal of Gastrointestinal Surgery2021,13,10:4

17Pouch Roux-en-Yvs No Pouch Roux-en-Y following total gastrectomy:a meta-analysis based on 12 studies显示文摘After a total resection of the stomach,the continuity of the gastrointestinal tract can be restored either by Rouxen-Y esophagojejunostomy with or without a pouch.There is still no consensus on the best reconstruction technique.The aim of this report was to derive a more precise estimation of Roux-en-Y esophagojejunostomy with a pouch compared with Roux-en-Y esophagojejunostomy without a pouch.Studies were identified by PubMed and Embase searches,and the inclusion criteria were randomized controlled trials (RCTs) comparing reconstruction techniques between Roux-en-Y with and without a pouch.A total of 12 studies including 1,018 patients were included.The meta-analysis shows that pouch Roux-en-Y does not significantly increase total postoperative complications,anastomotic leakage or mortality.Importantly,there is no significant difference in 5-year survival rates between the two groups.Patients with Roux-en-Y esophagojejunostomy complained significantly less of reflux symptoms and dumping syndrome,and had significantly less severe reflux esophagitis.Quality of life was significantly improved in patients with Roux-en-Y esophagojejunostomy with a pouch compared with patients who received Roux-en-Y reconstruction without a pouch.The results indicate the need for Roux-en-Y esophagojejunostomy with a pouch is a gastric substitute after total gastrectomy by comparison with Roux-en-Y esophagojejunostomy without a pouch.Liang Zong Ping Chen Yinbing Chen Guohao Shi 2011The Journal of Biomedical Research2011,25,2:3
18Giant solitary fibrous tumor arising from greater omentum显示文摘Extrathoracic solitary fibrous tumors(SFTs) have been described at almost every anatomic location of human body,but reports of SFT in the abdominal cavity are rare.We herein present a rare case of SFT originating from greater omentum.Computed tomography revealed a 15.8 cm × 21.0 cm solid mass located at superior aspect of stomach.Open laparotomy confirmed its mesenchymal origin.Microscopically,its tissue was composed of non-organized and spindle-shaped cells exhibiting atypical nuclei,which were divided up by branching vessel and collagen bundles.Immunohistochemical staining showed that this tumor was negative for CD117,CD99,CD68,cytokeratin,calretinin,desmin,epithelial membrane antigen,F8 and S-100,but positive for CD34,bcl-2,α-smooth muscle actin and vimentin.The patient presented no evidence of recurrence during follow-up.SFT arising from abdominal cavity can be diagnosed by histological findings and immunohistochemical markers,especially for CD34 and bcl-2 positive cases.Liang Zong Ping Chen Guang-Yao Wang Qun-Shan Zhu 2012World Journal of Gastroenterology2012,18,44:3

20Death decoy receptor overexpression and increased malignancy risk in colorectal cancer显示文摘AIM:To evaluate human epidermal growth factor receptor 2(HER2)and death decoy receptor(DcR3)as colorectal cancer prognostic indicators.METHODS:Colorectal carcinoma specimens from 300patients were analyzed by immunohistochemistry to detect the staining patterns of HER2 and DcR3.Classification of HER2 staining was carried out using the United States Food and Drug Administration semi-quantitative scoring system,with scores of 0 or 1+indicating a tumor-negative(normal expression)status and scores of 2+and 3+indicating a tumor-positive(overexpression)status.Classification of DcR3 was carried out by quantitating the percentage of positive cells within the stained section,with<10%indicating a tumor-negative status and≥10%indicating a tumor-positive status.Correlation of the HER2 and DcR3 staining status with clinicopathological parameters[age,sex,tumor size,differentiation,and the tumor,node,metastasis(pTNM)classification]and survival was statistically assessed.RESULTS:Tumor-positive status for HER2 and DcR3was found in 18.33%and 58.33%of the 300 colorectal carcinoma specimens,respectively.HER2 tumorpositive status showed a significant correlation with tumor size(P=0.003)but not with other clinicopathological parameters.DcR3 tumor-positive status showed a significant correlation with tumor differentiation(P<0.001),pTNM stage(P<0.001),and lymph node metastasis(P<0.001).However,correlation coefficient analysis did not indicate that a statistically significant correlation exists between tumor-positive status for the HER2 and DcR3 overexpression(P=0.236).Patients with specimens classified as DcR3-overexpressing had a significantly worse overall survival(OS)rate than those without DcR3 overexpression(median OS:42.11vs 61.21 mo;HR=50.27,95%CI:44.90-55.64,P<0.001).HER2 overexpression had no significant impact on median OS(35.10 mo vs 45.25 mo;HR=44.40,95%CI:39.32-49.48,P=0.344).However,patients with specimens classified as both HER2-and DcR3-overexpressing had a significantly poorer median OS than those with only HER2 overexpression(31.80 mo vs 52.20 mo;HR=35.10,95%CI:22.04-48.16,P=0.006).CONCLUSION:HER2 overexpression is not an independent prognostic marker of colorectal cancer,but DcR3 overexpression is highly correlated with lymph node metastasis and poor OS.Liang Zong Ping Chen Da-Xin Wang 2014World Journal of Gastroenterology2014,20,15:3
