| 2 | Long-term Traffic Volume Prediction Based on K-means Gaussian Interval Type-2 Fuzzy Sets显示文摘This paper uses Gaussian interval type-2 fuzzy se theory on historical traffic volume data processing to obtain a 24-hour prediction of traffic volume with high precision. A K-means clustering method is used in this paper to get 5 minutes traffic volume variation as input data for the Gaussian interval type-2 fuzzy sets which can reflect the distribution of historical traffic volume in one statistical period. Moreover, the cluster with the largest collection of data obtained by K-means clustering method is calculated to get the key parameters of type-2 fuzzy sets, mean and standard deviation of the Gaussian membership function.Using the range of data as the input of Gaussian interval type-2 fuzzy sets leads to the range of traffic volume forecasting output with the ability of describing the possible range of the traffic volume as well as the traffic volume prediction data with high accuracy. The simulation results show that the average relative error is reduced to 8% based on the combined K-means Gaussian interval type-2 fuzzy sets forecasting method. | 3 | Molecular characterization and m RNA expression of ribosomal protein L8 in Rana nigromaculata during development and under exposure to hormones显示文摘Like Xenopus laevis, some species of the Rana genus are also used to study endocrine disrupting chemicals(EDCs). Although ribosomal protein L8(rpl8) is the most-used reference gene for analyzing gene expression by quantitative reverse transcription polymerase chain reaction in Rana, its suitability as the reference gene has never been validated in any species of the Rana genus. We characterized rpl8 c DNA in Rana nigromaculata, a promising native species in East Asia for assaying endocrine disrupting effects. We found that the rpl8 c DNA consisted of919 bp and encoded 257 amino acids, exhibiting high identities of amino acid sequence with known rpl8 in other Rana species. Then, we examined the stability of m RNA expression during development. Compared with elongation factor 1 alpha 1, another common housekeeping gene, neither stage-specific nor tissue-specific expression of the rpl8 gene was found in all tissues examined(brain, liver, intestine, tail, testis and ovary) during R. nigromaculata development. Finally, we investigated rpl8 expression under exposure to hormones. | 4 | Characterization of glucose-6-phosphate dehydrogenase deficiency and identification of a novel haplotype 487G>A/IVS5-612(G>C) in the Achang population of southwestern China显示文摘The prevalence of glucose-6-phosphate dehydrogenase (G6PD) deficiency and its gene mutations were studied in the Achang population from Lianghe County in Southwestern China. We found that 7.31% (19 of 260) males and 4.35% (10 of 230) females had G6PD deficiency. The molecular analysis of G6PD gene exons 2―13 was performed by a PCR-DHPLC-Sequencing or PCR-Sequencing. Sixteen inde-pendent subjects with G6PD Mahidol (487G>A) and the new polymorphism IVS5-612 (G>C), which combined into a novel haplotype, were identified accounting for 84.2% (16/19). And 100% Achang G6PD Mahidol were linked to the IVS5-612 C. The percentage of G6PD Mahidol in the Achang group is close to that in the Myanmar population (91.3% 73/80), which implies that there are some gene flows between Achang and Myanmar populations. Interestingly, G6PD Canton (1376G>T) and G6PD Kaiping (1388G>A), which were the most common G6PD variants from other ethnic groups in China, were not found in this Achang group, suggesting that there are different G6PD mutation profiles in the Achang group and other ethnic groups in China. | 5 | Optimizing Mechanical Properties of AlCoCrFeNiTi_x High-Entropy Alloys by Tailoring Microstructures显示文摘The effects of Ti addtions and the heat treatment on the mechanical properties of AlCoCrFeNiTix (x = 0, 0.2, 0.3, 0.4 and 0.5) high-entropy alloys (HEAs) were studied. The results show that the dendrite phase with a body-centered-cubic (bcc) structure transforms into the interdendrite phase with a new bcc structure. With the increase of the Ti contents and heat-treatment temperature, the average hardness and yield strengths are greatly improved, and the highest hardness and yielding strength are 583 HV and 2.07 GPa, respectively in the investigated HEA system. The as-cast and annealed HEAs exhibit excellent mechanical properties, combining with high yielding strength and plasticity. In view of the absence of research in vibration analysis of viscoelastic plate with varying thickness, this study devotes to investigate the dynamic behaviors of axially moving viscoelastic plate with varying thickness. Based on the thin plate theory and the two-dimensional viscoelastic differential constitutive relation, the differential equation of motion of the axially moving viscoelastic rectangular plate is derived, the plate constituted by Kelvin-Voigt model has linearly varying thickness in the y-direction. The dimensionless complex frequencies of axially moving viscoelastic plate with four edges simply supported are calculated by the differential quadrature method, curves of real parts and imaginary parts of the first three-order dimensionless complex frequencies versus dimensionless moving speed are obtained, the effects of the aspect ratio, thickness ratio, the dimensionless moving speed and delay time on the dynamic behaviors of the axially moving viscoelastic rectangular plate with varying thickness are analyzed. | 9 | A screening assay for thyroid hormone signaling disruption based on thyroid hormone-response gene expression analysis in the frog Pelophylax nigromaculatus显示文摘Amphibian metamorphosis provides a wonderful model to study the thyroid hormone(TH)signaling disrupting activity of environmental chemicals, with Xenopus laevis as the most commonly used species. This study aimed to establish a rapid and sensitive screening assay based on TH-response gene expression analysis using Pelophylax nigromaculatus, a native frog species distributed widely in East Asia, especially in China. To achieve this, five candidate TH-response genes that were sensitive to T3 induction were chosen as molecular markers, and T3 induction was determined as 0.2 nmol/L T3 exposure for 48 hr. The developed assay can detect the agonistic activity of T3 with a lowest observed effective concentration of 0.001 nmol/L and EC50 at around 0.118–1.229 nmol/L, exhibiting comparable or higher sensitivity than previously reported assays. We further validated the efficiency of the developed assay by detecting the TH signaling disrupting activity of tetrabromobisphenol A(TBBPA), a known TH signaling disruptor. In accordance with previous reports, we found a weak TH agonistic activity for TBBPA in the absence of T3,whereas a TH antagonistic activity was found for TBBPA at higher concentrations in the presence of T3, showing that the P. nigromaculatus assay is effective for detecting TH signaling disrupting activity. Importantly, we observed non-monotonic dose-dependent disrupting activity of TBBPA in the presence of T3, which is difficult to detect with in vitro reporter gene assays. | 10 | Advanced optimization of gas turbine aero-engine transient performance using linkage-learning genetic algorithm:PartⅡ,optimization in flight mission and controller gains correlation development显示文摘PartⅠhas illustrated the procedures to apply the Linkage Learning Genetic Algorithm(LLGA)in Gas Turbine Engine(GTE)controller gains tuning and generated the optimization results for runway conditions from idle to takeoff.However,the total pressure and temperature of the engine inlet vary as the changing of altitude and Mach number,which would lead to the variation in fuel flow supply regulation.As a result,the optimized gains in runway might not be suitable for other flight conditions.In order to maintain the optimal control performance,the GTE controller gains should be adjusted according to the flight conditions.This paper extends the application of the LLGA method to other flight conditions and then simulates a complete flight mission with different gains and weather condition configurations.For this purpose,the control parameters in the Simulink model of the GTE controller are first corrected by the weather condition in altitude.Then,a typical flight mission is defined and divided into different flight segments based on the altitude and Mach number configuration.One representative point is selected from each segment as the datum point for optimization process.After this step,the LLGA method is used to find the best gains combinations for different flight conditions and the differences in optimization effects for different flight conditions are analyzed subsequently.The simulation results show that the optimization effect of the control performance of each flight condition is dependent on the value of(θδ)~(1/2)and the optimal K_(pla)in some flight conditions is approximately equal to p hd times of the Kplavalue in sea level standard condition.Finally,the complete flight mission is simulated with different gains and weather condition configurations.The simulation results show that the engine performance has been greatly improved after optimization by LLGA in the transient state and the high altitude conditions.In other steady states,the optimization effect is not very obvious. | Yinfeng LIU Soheil JAFARI Theoklis NIKOLAIDIS | 2021 | Chinese Journal of Aeronautics2021,34,4: | 3 |
| 11 | Molecular characterization and developmental expression patterns of thyroid hormone receptors(TRs) and their responsiveness to TR agonist and antagonist in Rana nigromaculata显示文摘Considering some advantages of Rana nigromaculata as an experimental species, we propose that this species, like Xenopus laevis, could be used to assay thyroid hormone(TH) signaling disrupting actions. To validate the utilizability of R. nigromaculata, we investigated the responsiveness of R. nigromaculata to a TH receptor(TR) agonist(T3) and antagonist(amiodarone) by analyzing expression, based on characterizing TR cDNA and developmental expression patterns. With high levels of identity with the corresponding genes in X. laevis, both TRα and TRβ in R. nigromaculata exhibited roughly similar developmental expression patterns to those of X. laevis, in spite of some species-specific differences. Both TRα and TRβ expression had greater changes in the liver and intestine than in the tail and brain during metamorphosis. T3 exposure for 2 days induced more dramatic increases of TRβ expression in stage 27 than in stage34 tadpoles but not in stage 42 tadpoles, showing that the responsiveness of R. nigromaculata to TH decreased with development and disappeared at the onset of metamorphic climax.Corresponding to greater changes of TRβ expression in the liver and intestine than in the tail and brain during metamorphosis, the liver and intestine had higher responsiveness to exogenous T3 than the tail and brain. Amiodarone inhibited T3-induced TRβ expression. | 12 | Preparation of Cordierite-mullite Ceramics for Solar Thermal Storage显示文摘We developed cordierite-mullite composite ceramic materials to package and encapsulate PCM, and presented a preparation process from raw materials of kaolin, talc and alumina. The properties and microstructre of cordierite-mullite composite ceramic were studied. Due to the strengthening effects of mullite, the sample C2(80 wt% of cordierite and 20 wt % of mullite) sintered at 1 420 ℃ possessed excellent physical properties. Determined by X-ray diffraction(XRD), scanning electron microscope(SEM) and energy dispersive spectroscopy(EDS) analysis, cuboid-shaped cordierite crystals and needle-like or long quadrilateral-prism mullite crystals with staggered patterns were found, which endowed the composites preferable mechanical strength. | 13 | Optimization of the T3-induced Xenopus metamorphosis assay for detecting thyroid hormone signaling disruption of chemicals显示文摘T3-induced Xenopus metamorphosis is an ideal model for detecting thyroid hormone(TH)signaling disruption of chemicals. To optimize the T3-induced Xenopus assay and improve its sensitivity and reproducibility, we intend to develop quantitatively morphological endpoints and choose appropriate concentrations and exposure durations for T3 induction.Xenopus laevis at stage 52 were exposed to series of concentrations of T3(0.31–2.5 nmol/L)for 6 days. By comparing morphological changes induced by T3, we propose head area,mouth width, unilateral brain width/brain length, and hindlimb length/snout-vent length as quantitative parameters for characterizing T3-induced morphological changes, with body weight as a parameter for indicating integrated changes. By analyzing time-response curves, we found that following 4-day exposure, T3-induced grossly morphological changes displayed linear concentration–response curves, with moderate morphological changes resulting from 1.25 nmol/L T3 exposure. When using grossly morphological endpoints to detect TH signaling disruption, we propose 4 days as exposure duration of T3, with concentrations close to 1.25 nmol/L as induction concentrations. However, it is appropriate to examine morphological and molecular changes of the intestine on day 2 due to their early response to T3.
