1Protein trans-splicing based dual-vector delivery of the coagulation factor Ⅷ gene显示文摘A dual-vector system was explored for the delivery of the coagulation factor VIII gene,using intein-mediated protein trans-splicing as a means to produce intact functional factor VIII post-translationally.A pair of eukaryotic expression vectors,expressing Ssp DnaB intein-fused heavy and light chain genes of B-domain deleted factor VIII (BDD-FVIII),was constructed.With transient co-transfection of the two vectors into 293 and COS-7 cells,the culture supernatants contained (137±23) and (109±22) ng mL–1 spliced BDD-FVIII antigen with an activity of (1.05±0.16) and (0.79±0.23) IU mL–1 for 293 and COS-7 cells,respectively.The spliced BDD-FVIII was also detected in supernatants from a mixture of cells transfected with inteinfused heavy and light chain genes.The spliced BDD-FVIII protein bands from cell lysates were visualized by Western blotting.The data demonstrated that intein could be used to transfer the split factor VIII gene and provided valuable information on factor VIII gene delivery by dual-adeno-associated virus in hemophilia A gene therapy.ZHU FuXiang,LIU ZeLong,CHI XiaoYan & QU HuiGe Life Science College of Ludong University,Yantai 264025,China 2010Science China(Life Sciences)2010,53,6:27
2Lie bialgebras of generalized Witt type显示文摘In this paper, all Lie bialgebra structures on the Lie algebras of generalized Witt type are considered. It is proved that, for any Lie algebra W of generalized Witt type, all Lie bialgebras on W are the coboundary triangular Lie bialgebras. 3Variations in carbon isotope ratios of C_3 plants and distribution of C_4 plants along an altitudinal transect on the eastern slope of Mount Gongga显示文摘Variations in carbon isotopic ratios (δ 13C) of C3 plants and distribution of C4 plants were investigated along an altitudinal transect on the eastern slope of Mount Gongga, and the environmental effects on them were discussed. It is shown that plants with C4 photosynthetic pathway mainly occur at altitudes below 2100 m a.s.l., suggesting that the low summer temperature is responsible for the distributional pattern. In addition, δ 13C of C3 plants increases with elevation at the region above 2000 m a.s.l. with the characteristics of humid climate, and the increase rate in δ 13C for C3 plants is about 1.3‰ per kilometer. 4An analysis of 680 cases of cholangiocarcinoma from 8 hospitals显示文摘BACKGROUND: The outcome of patients with cholangio-carcinoma is poor. To evaluate the experience in the diagnosis and surgical treatment of cholangiocarcinoma, we investigated the status quo of diagnosis and treatment of cholangiocarcinoma in China. METHOD: The clinical data of 680 patients with cholangiocarcinoma treated at 8 hospitals from 1995 to 2001 were retrospectively analyzed with SPSS software package. RESULTS: The incidence of the tumor was the highest in the age group of 60-65 years. Meanwhile,the incidence was higher in aged men than in aged women, with a male to female ratio of 1.39:1. Proximal cholangiocarcinoma was the commonest ( 41.6% ) and distant cholangiocarcinoma the second(28.7% ) in the 680 patients. B-mode ultrasonogra-phy for cholangiocarcinoma was performed in 80.3% of the patients. Non-traumatic examinations such as computed tomography (CT), magnetic resonance image (MRI) and magnetic resonance cholangiopancreatography ( MRCP ) were more widely used than that of traumatic examinations such as percutaneous transhepatic cholangiography (PTC) and endoscopic retrograde cholangiopancreatography ( ER-CP). The low- and middle-differentiation cancer of the proximal bile duct accounted for about 50%. Most of the patients suffered from late-stage cholangiocarcinoma. The resection rate of the tumor was low, and the rate of radical operation was only 21.6% (147/680). CONCLUSIONS: Cholangiocarcinoma is common in the aged men. Its diagnosis and treatment have been improved, but little. Most patients are diagnosed as having late-stage cholangiocarcinoma at the time of outpatient clinic, and the rate of radical operation is low. 6Run off-on-out method and models for soil infiltrability on hill-slope under rainfall conditions显示文摘The soil infiltrability of hill-slope is important to such studies and practices as hydrological process, crop water supply, irrigation practices, and soil erosion. A new method for measuring soil infiltrability on hill-slope under rainfall condition with run off-on-out was advanced. Based on water (mass) balance, the mathematic models for soil infiltrability estimated from the advances of runoff on soil surface and the water running out of the slope were derived. Experiments of 2 cases were con-ducted. Case I was done under a rainfall intensity of 20 mm/h, at a slope gradient of about 0° with a runoff/on length (area) ratio of 1 : 1. Case II was under a rainfall intensity of 60 mm/h and a slope of 20° with a runoff/on length (area) ratio of 1 : 1. Double ring method was also used to measure the infiltrability for comparison purposes. The experiments were done with soil moisture of 10%. Required data were collected from laboratory experiments. The infiltrability curves were computed from the experimental data. The results indicate that the method can well conceptually represent the transient infiltrability process, with capability to simulate the very high initial soil infiltrability. The rationalities of the method and the models were validated. The errors of the method for the two cases were 1.82%/1.39% and 4.49%/3.529% (Experimental/Model) respectively, as estimated by comparing the rainfall amount with the infiltrated volume, to demonstrate the accuracy of the method. The transient and steady infiltrability measured with double ring was much lower than those with this new method, due to water supply limit and soil aggregates breaking down at initial infiltration stage. The method can overcome the short backs of the traditional sprinkler method and double ring method for soil infil-traility. 7CFAR assessment of covariance matrix estimators for non-Gaussian clutter显示文摘In the non-Gaussian clutter modeled as independent and identically distributed spherically invariant random vectors, three estimators of sample covariance matrix (SCM), normalized sample covariance matrix (NSCM) and the corresponding recursive estimator (NSCM-RE) are analyzed. Based on the uniform theorem, three corresponding adaptive normalized matched filters (ANMF) are evaluated from the standpoint of constant false alarm rate (CFAR) property. The theoretical results demonstrate that the SCM-ANMF is only CFAR to the normalized clutter covariance matrix (NCCM); the NSCM-ANMF is only CFAR to the clutter power level; and the NSCM-RE-ANMF with finite number of iterations is still not CFAR to the NCCM. To ensure CFAR property of ANMF, an adaptive estimator (AE) is devised. Moreover, with AE as the initialization matrix for the iterations, the AE-RE is proposed. With finite number of iterations, the corresponding AE-REANMF guarantees CFAR property to both of the NCCM and the clutter power level. 9The fluxes and controlling factors of N_2O and CH_4 emissions from freshwater marsh in Northeast China显示文摘Nitrous oxide (N2O) and methane (CH4) emissions were measured using a static chamber method in two adjacent plots of freshwater marsh predominated by Calamagrostis angustifolia, one is seasonal waterlogged (SW) and the other without surface water accumulation (NW), in Sanjiang Plain wetland (47°35′N, 133°31′E), northeast China, during 2002-2004. The diurnal and seasonal flux variations of both gases were significantly correlated with 5-cm-soil temperature. The NW marsh is a source of N2O and sink of CH4, while the SW marsh is sink of N2O and source of CH4. Remarkably, we observed a N2O emission peak under Eh of +300 to +100 mV, and CH4 emission peak under Eh of +300 to +400 mV, which indicate additional sources of N2O and CH4 for the freeze-thaw induced emission peaks of N2O and CH4 observed between late July and early August. 11Stochastic resonance in surface catalytic oxidation of carbon monoxide induced by colored noise显示文摘The dynamical behavior of surface catalytic oxidation reaction of Pt(110)/CO+O2 modulated by colored noise, under the condition of specific temperature, has been investigated when the partial pressure of CO gas is near the supercritical Hopf bifurcation point. By computer simulation the oscillation and stochastic resonance induced by colored noise are observed. The influences of the intensity and correlation time of colored noise on stochastic resonance are discussed. 12The synthesis of astaxanthin esters,independent of the formation of cysts,highly correlates with the synthesis of fatty acids in Haematococcus pluvialis显示文摘The compositions and contents of astaxanthin esters and fatty acids in four types of Haematococcus pluvialis cells were studied by HPLC and GC-MS. Results showed that the synthesis and accumulation of astaxanthin was independent of the formation of cysts, but was highly correlated with the synthesis and accumulation of fatty acids, though it is an well known phenomenon that the accumulation of astaxanthin is usually accompanied by the formation of cyst. The red cysts contain more than 30% of fatty acids, with 81% of the unsaturated fatty acids. 16Responses of distribution pattern of desert riparian forests to hydrologic process in Ejina oasis显示文摘By using the theories and methods of landscape ecology and the technology of GIS and RS, a study has been carried out on the responses of distribution pattern of desert riparian forest to hydrologic process on the basis of the hydrologic data from 1990 to 2000 and the TM image of 2001 year. The results showed that: (1) there appears an even distribution pattern for the relative forest area in oasis, however, the degenerated forest diaplays an increasing tendency from west to east; (2) the desert riparian forest in Ejina is in completely degenerated process at the patch scale; (3) the number of patch is influenced not only by hydrologic process,but also by agricultural activity such as cultivation. The severe deterioration of the degraded vegetation in whole oasis initiates from lower reaches, and gradually impels to upstream; the fragmentation of landscape in the terminal site is more obvious, which is influenced by river shape and decreasing flux of water. It is found that the influence of surface hydrologic process to the ground hydrologic process of desert riparian forest in Ejina oasis is little for the recent ten years. The relative area of the degenerated forest increased with increasing ground water depth in the direction of parallel to river channel. 19The response of the early developmental stages of Laminaria japonica to enhanced ultraviolet-B radiation显示文摘The responses of the early development of Laminaria japonica collected from Kiaochow Bay in China to enhanced ultraviolet-B radiation (UV-B, 280—320 nm) were studied in the laboratory. The low UV-B ra-diations (11.7—23.4 J·m-2·d-1) had no significant effects on zoospores attachment, but when the UV-B dose > 35.1 J·m-2·d-1 the attachment decreased significantly compared with the control. Germination of embryospores was >93% under the low (11.7—35.1 J·m-2·d-1) doses, and in the range of 78.5%—88.5% under the high (46.8—70.2 J·m-2·d-1) UV-B doses, indicating a significant radiation effect. Under the higher UV-B exposure (35.1—70.2 J·m-2·d-1), all of the few gametophytes formed from embryospores died 120 h post-release. After exposure to the low UV-B radiation (11.7—23.4 J·m-2·d-1), the formation of sporophytes decreased and the female gametophyte clones increased compared with the control. However, the sex ratio and the relative growth of female gametophytes/sporophytes had not signifi-cantly changed. 20Flow Characteristics of Double-Cruciform Parachute at Inflating and Inflated Conditions显示文摘The fluid-structure interaction (FSI) between the canopy and flow field on the inflating and inflated conditions is investigated based on the arbitrary Lagrange-Euler (ALE) method,in both a single-and double-cruciform parachute systems.The projection area of canopy is calculated in the inflation process.The flow field characteristics and the interaction between canopies are analyzed.Results showed that,with free stream velocity of 50m/s,overinflation phenomenon would not occur during the inflation process of the double-cruciform-parachute system,because the collision and extrusion of the two canopies during inflation obstructed the oscillation of the inner gores.Concurrently,compared with the single-cruciform parachute,the vortex motion in the wake of double-cruciform-parachute is more intense.Thus the double-cruciform parachute system oscillated at a velocity of 50 m/s with an angle of less than 6.8°.By comparison,the oscillation angle of the single-cruciform parachute was within 3.5° at the velocity of 50m/s.The results are consistent with those of the wind tunnel test.Fang Ming Sun Jianhong Zhang Tong Hou Bin Zhang Yantai 2018Transactions of Nanjing University of Aeronautics and Astronautics2018,35,6:3
