| 1 | Towards precision medicine:advances in 5-hydroxymethylcytosine cancer biomarker discovery in liquid biopsy显示文摘Robust and clinically convenient biomarkers for cancer diagnosis,early detection,and prognosis have great potential to improve patient survival and are the key to precision medicine.The advent of next-generation sequencing technologies enables a more sensitive and comprehensive profiling of genetic and epigenetic information in tumor-derived materials.Researchers are now able to monitor the dynamics of tumorigenesis in new dimensions,such as using circulating cell-free DNA(cfDNA)and tumor DNA(ctDNA).Mutation-based assays in liquid biopsy cannot always provide consistent results across studies due partly to intra-and inter-tumoral heterogeneity as well as technical limitations.In contrast,epigenetic analysis of patient-derived cfDNA is a promising alternative,especially for early detection and disease surveillance,because epigenetic modifications are tissue-specific and reflect the dynamic process of cancer progression.Therefore,cfDNA-based epigenetic assays are emerging to be a highly sensitive,minimally invasive tool for cancer diagnosis and prognosis with great potential in future precise care of cancer patients.The major obstacle for applying epigenetic analysis of cfDNA,however,has been the lack of enabling techniques with high sensitivity and technical robustness.In this review,we summarized the advances in epigenome-wide profiling of 5-hydroxymethyl-cytosine(5hmC)in cfDNA,focusing on the detection approaches and potential role as biomarkers in different cancer types. | Chang Zeng Emily Kunce Stroup Zhou Zhang Brian C-HChiu Wei Zhang | 2019 | Cancer Communications2019，39，1： | 7 |
| 2 | National Public Health Surveillance in China: Implications for Public Health in China and the United States显示文摘Study Objective:Throughout the world, political developments have brought new demands to communities to prevent and intervene in the incidence of infectious and noninfectious conditions.Historically, these developments have required new and more effective public health surveillance ac tivities. This report describes public health surveillance practice in the People’s Republic of China,making comparisons with selected aspects of surveillance in the United States with respect to collec tion, analysis, diasemination, and use of data. Main results: In both the People’s Republic of China and the United States, political change has affected health, and multiple surveillance system are used in public health practice. Surveillance of acute infectious diseases based on the reporting of legally notifiable diseases and the use of vital records for surveillance have the longest established history in both countries. In both countries, da ta from the surveillance systems have been used to affect public health policy.Conclusions: In comparing surveillance practices in there countries, we find similarities in con ditions reported and in the dissemination of the data. At the same time, legal, aseial, cultural, and economic differences between the nations have affected the practice of surveillance in analysis and evaluation. | 3 | G-Quadruplex(G4) Motifs in the Maize(Zea mays L.) Genome Are Enriched at Specific Locations in Thousands of Genes Coupled to Energy Status,Hypoxia,Low Sugar,and Nutrient Deprivation显示文摘The G-quadruplex(G4) elements comprise a class of nucleic acid structures formed by stacking of guanine base quartets in a quadruple helix.This G4 DNA can form within or across single-stranded DNA molecules and is mutually exclusive with duplex B-form DNA.The reversibility and structural diversity of G4 s make them highly versatile genetic structures,as demonstrated by their roles in various functions including telomere metabolism,genome maintenance,immunoglobulin gene diversification,transcription,and translation.Sequence motifs capable of forming G4 DNA are typically located in telomere repeat DNA and other non-telomeric genomic loci.To investigate their potential roles in a large-genome model plant species,we computationally identified 149,988 non-telomeric G4 motifs in maize(Zea mays L.,B73 AGPv2),29%of which were in non-repetitive genomic regions.G4 motif hotspots exhibited non-random enrichment in genes at two locations on the antisense strand,one in the 5'UTR and the other at the 5'end of the first intron.Several genic G4 motifs were shown to adopt sequence-specific and potassium-dependent G4 DNA structures in vitro.The G4 motifs were prevalent in key regulatory genes associated with hypoxia(group VII ERFs),oxidative stress(DJ-1/GATase1),and energy status(AMPK/SnRK) pathways.They also showed statistical enrichment for genes in metabolic pathways that function in glycolysis,sugar degradation,inositol metabolism,and base excision repair.Collectively,the maize G4 motifs may represent conditional regulatory elements that can aid in energy status gene responses.Such a network of elements could provide a mechanistic basis for linking energy status signals to gene regulation in maize,a model genetic system and major world crop species for feed,food,and fuel. | Carson M.Andorf Mykhailo Kopylov Drena Dobbs Karen E.Koch M.Elizabeth Stroupe Carolyn J.Lawrence Hank W.Bass | 2014 | Journal of Genetics and Genomics2014，41，12： | 2 |
