| 1 | Hypothetical mode of action of earthworm extract with hepatoprotective and antioxidant properties显示文摘The hepatoprotective potential of earthworm extract (EE) (Lampito mauritii, Kinberg) was evaluated against paracetamol-induced liver injury in Wistar albino rat, in comparison with silymarin, the standard hepatoprotective drug. We observed a reduction in liver antioxidants, such as glutathione (GSH), superoxide dismutase (SOD), glutathione peroxidase (GPx), and catalase (CAT) and in serum total protein, and an increase in serum alkaline phosphatase (ALP), serum aspertate aminotranferase (AST), serum alanine aminotranferase (ALT), bilirubin and liver thiobarbituric acid reactive substances (TBARS) due to liver injury in the paracetamol-administered rats (2 g/kg). On the contrary, increased activities of liver GSH, SOD, GPx, CAT and serum total protein level, and decrease in the contents of serum ALP, AST, ALT, bilirubin and liver TBARS were observed in rats administered with different doses of EE (100, 200 and 300 mg/kg), which are similar to the activities of hepato-protective drug silymarin (150 mg/kg). The mode of action of EE as evidenced by the above parameters may suggest that EE, on the one hand, prevents the formation of the reactive oxygen groups, or scavenges these groups, thereby preventing the damage on the hepatic cells, and, on the other hand, modulates the genes responsible for synthesis of antioxidant enzymes such as GPx, CAT and SOD in liver tissue and decreases the serum enzymatic activities such as ALP, AST and ALT. | Mariappan BALAMURUGAN Kasi PARTHASARATHI Lalpet Souri RANGANATHAN Edwin L. | 2 | Evaluation of heavy metal contamination hazards in nuisance dust particles, in Kurdistan Province, western Iran显示文摘The effects of natural andgeochemical factors depending on heavy metal contamination in nuisance dust particles were evaluated. The nuisance dust particles were sampled using passive deposit gauge method for one year from April2010 to March2011 and the obtained samples were measured for the total contents and the contamination levels of Fe, Mn, Cu and As usinggeo-accumulation index (Igeo ), enrichment factor (EF) and the integrated pollution index (IPI). The results showed that, the contamination levels of Fe and Mn based on Igeo values, were uncontaminated (Igeo < 0) (variations of the Igeo index was from -3.11 to -1.751 for Fe, from -0.630 to -1.925 for Mn), while the values of Cu and As were demonstrated to have moderate contamination based on Igeo values (variations of Igeo index was from -1.125 to 0.848 for Cu, and from -2.002 to 1.249 for As). The analysis of EF also revealed minor to moderate enrichment for Mn (1.215-4.214), minor to moderately severe enrichment for Cu (2.791-6.484), and As (1.370-8.462), respectively. The variation of the IPI index also showed low to moderate level of heavy metal pollution in nuisance dust particulates (0.511-1.829). | 4 | A validated HPLC method for the determination of donepezil in human plasma after derivatization with 9-fluorenylmethyl chloroformate显示文摘A new HPLC method has been developed for determining donepezil in human plasma. To find the optimum conditions, a derivatization reaction was performed in different media, and the reaction product was identified by NMR and GC-MS after a semi-preparative HPLC separation. Under optimized conditions, donepezil was derivatized by 9-fluorenylmethyl chloroformate in chloroform and carbonate buffer at pH 9.5 in the presence of NaI after solid-phase extraction from a plasma sample. The reaction product was quantified on a reversed-phase TRACER EXCEL ODS-A, 5 μm column using a mixture of acetonitrile–10 mM acetate buffer(pH 6.0)–THF(60:35:5, v/v/v) as the mobile phase with fluorescence detection at 264 nm(ex) and 313 nm(em). Fluoxetine was used as the internal standard. The total run-time of the analysis was about 10 min, and a clean chromatogram was obtained. The developed method was linear over the range of 1–100 ng/mL in 500 μL of plasma samples(r2>0.998). The intra-day and inter-day precision values were in the range of 2.6%–11.6%.
