1TGF-β/BMP signaling and other molecular events: regulation of osteoblastogenesis and bone formation显示文摘Transforming growth factor-beta(TGF-β)/bone morphogenetic protein(BMP) plays a fundamental role in the regulation of bone organogenesis through the activation of receptor serine/threonine kinases. Perturbations of TGF-β/BMP activity are almost invariably linked to a wide variety of clinical outcomes, i.e., skeletal, extra skeletal anomalies, autoimmune, cancer, and cardiovascular diseases. Phosphorylation of TGF-β(I/II) or BMP receptors activates intracellular downstream Smads, the transducer of TGF-β/BMP signals. This signaling is modulated by various factors and pathways, including transcription factor Runx2. The signaling network in skeletal development and bone formation is overwhelmingly complex and highly time and space specific.Additive, positive, negative, or synergistic effects are observed when TGF-β/BMP interacts with the pathways of MAPK, Wnt, Hedgehog(Hh), Notch, Akt/m TOR, and mi RNA to regulate the effects of BMP-induced signaling in bone dynamics. Accumulating evidence indicates that Runx2 is the key integrator, whereas Hh is a possible modulator, mi RNAs are regulators, and b-catenin is a mediator/regulator within the extensive intracellular network. This review focuses on the activation of BMP signaling and interaction with other regulatory components and pathways highlighting the molecular mechanisms regarding TGF-β/BMP function and regulation that could allow understanding the complexity of bone tissue dynamics.Md Shaifur Rahman Naznin Akhtar Hossen Mohammad Jamil Rajat Suvra Banik Sikder M Asaduzzaman 2015Bone Research2015,3,1:53
2Pharmacologic inducers of the uric acid exporter ABCG2 as potential drugs for treatment of gouty arthritis显示文摘Uric acid is the end product of purine catabolism and its plasma levels are maintained below its maximum solubility in water(6–7 mg/dl).The plasma levels are tightly regulated as the balance between the rate of production and the rate of excretion, the latter occurring in urine(kidney),bile(liver)and feces(intestinal tract).Reabsorption in kidney is also an important component of this process. Both excretion and reabsorption are mediated by specific transporters.Disruption of the balance between production and excretion leads to hyperuricemia, which increases the risk of uric acid crystallization as monosodium urate with subsequent deposition of the crystals in joints causing gouty arthritis.Loss-of-function mutations in the transporters that mediate uric acid excretion are associated with gout. The ATP-Binding Cassette exporter ABCG2 is important in uric acid excretion at all three sites:kidney(urine),liver(bile),and intestine(feces).Mutations in this transporter cause gout and these mutations occur at significant prevalence in general population. However, mutations that are most prevalent result only in partial loss of transport function. Therefore,if the expression of these partially defective transporters could be induced, the increased number of the transporter molecules would compensate for the mutation-associated decrease in transport function and hence increase uric acid excretion. As such, pharmacologic agents with ability to induce the expression of ABCG2 represent potentially a novel class of drugs for treatment of gouty arthritis.Bojana Ristic Mohd Omar Faruk Sikder Yangzom D.Bhutia Vadivel Ganapathy 2020Asian Journal of Pharmaceutical Sciences2020,15,2:13
3Screening and evaluation of reliable traits of upland cotton(Gossypium hirsutum L.)genotypes for salt tolerance at the seedling growth stage显示文摘Background:Salt stress significantly inhibits the growth, development, and productivity of cotton because of osmotic, ionic, and oxidative stresses. Therefore, the screening and development of salt tolerant cotton cultivars is a key issue towards sustainable agriculture. This study subjected 11 upland cotton genotypes at the seedling growth stage to five different salt concentrations and evaluated their salt tolerance and reliable traits. Results:Several morpho-physiological traits were measured after 10 days of salinity treatment and the salt tolerance performance varied significantly among the tested cotton genotypes. The optimal Na Cl concentration for the evaluation of salt tolerance was 200 mmol·L-1.Membership function value and salt tolerance index were used to identify the most consistent salt tolerance traits.Leaf relative water content and photosynthesis were identified as reliable indicators for salt tolerance at the seedling stage. All considered traits related to salt tolerance indices were significantly and positively correlated with each other except for malondialdehyde.Cluster heat map analysis based on the morpho-physiological salt tolerance-indices clearly discriminated the 11 cotton genotypes into three different salt tolerance clusters.Cluster I represented the salt-tolerant genotypes(Z9807,Z0228,and Z7526)whereas clusters II(Z0710,Z7514,Z1910,and Z7516)and III(Z0102,Z7780,Z9648,and Z9612)represented moderately salttolerant and salt-sensitive genotypes, respectively. Conclusions:A hydroponic screening system was established.Leaf relative water content and photosynthesis were identified as two reliable traits that adequately represented the salt tolerance of cotton genotypes at the seedling growth stage.Furthermore, three salt-tolerant genotypes were identified, which might be used as genetic resources for the salt-tolerance breeding of cotton.SIKDER Ripon Kumar WANG Xiangru JIN Dingsha ZHANG Hengheng GUI Huiping DONG Qiang PANG Nianchang ZHANG Xiling SONG Meizhen 2020Journal of Cotton Research2020,3,2:5
4Characterization, prevalence and antibiogram study of Staphylococcus aureus in poultry显示文摘Objective: To reveal the presence of methicillin resistant Staphylococcus aureus(S. aureus)(MRSA) in poultry samples and to determine the antibiogram pattern against five antibiotics. Methods: Samples from different poultry farm of Chittagong city, Bangladesh were examined for S. aureus by different biochemical tests and confirmed as MRSA by identifying the presence of mec A gene using PCR. Antibiotic resistance pattern in S. aureus was determined by antibiotic disk diffusion method. Results: In this study, a total of 60 samples(30 from nasal swabs and 30 from cloacal swabs) were used, of which 54 were confirmed as S. aureus by different biochemical tests. Among these, 12 were confirmed as MRSA by detecting mec A gene using PCR.During antibiogram study, both nasal and cloacal samples showed the highest resistance against penicillin-G and the lowest resistance was observed against neomycin. Conclusions: Based on the present study, it can be said that different antibiotics are used extensively in poultry that leads to MRSA and is alarming for human health.Yeasmeen Ali Md.Ashraful Islam Nazmul Hasan Muzahid Mohd.Omar Faruk Sikder Md.Amzad Hossain Lolo Wal Marzan 2017Asian Pacific Journal of Tropical Biomedicine2017,7,3:2
