| 3 | Clinicopathologic factors and molecular markers related to lymph node metastasis in early gastric cancer显示文摘AIM: To analyze predictive factors for lymph node metastasis in early gastric cancer.METHODS: We analyzed 1104 patients with early gastric cancer(EGC) who underwent a gastrectomy with lymph-node dissection from May 2003 through July 2011. The clinicopathologic factors and molecular markers were assessed as predictors for lymph node metastasis. Molecular markers such as microsatellite instability, human mut L homolog 1, p53, epidermal growth factor receptor(EGFR) and human epidermal growth factor receptor 2(HER2) were included. The χ2 test and logistic regression analysis were used to determine clinicopathologic parameters.RESULTS: Lymph node metastasis was observed in 104(9.4%) of 1104 patients. Among 104 cases of lymph node positive patients, 24 patients(3.8%) were mucosal cancers and 80 patients(16.7%) were submucosal. According to histologic evaluation, the number of lymph node metastasis found was 4(1.7%) for well differentiated tubular adenocarcinoma, 45(11.3%) for moderately differentiated tubular adenocarcinoma, 36(14.8%) for poorly differentiated tubular adenocarcinoma, and 19(8.4%) for signet ring cell carcinoma. Of 690 EGC cases, 77 cases(11.2%) showed EGFR overexpression. HER2 overexpression was present in 110 cases(27.1%) of 406 EGC patients. | 4 | Photodynamic therapy prolongs metal stent patency in patients with unresectable hilar cholangiocarcinoma显示文摘AIM:To evaluate the effect of photodynamic therapy (PDT) on metal stent patency in patients with unresectable hilar cholangiocarcinoma (CC). METHODS:This was a retrospective analysis of patients with hilar CC referred to our institution from December, 1999 to January, 2011. Out of 232 patients, thirty-three patients with unresectable hilar CC were treated. Eighteen patients in the PDT group were treated with uncovered metal stents after one session of PDT. Fifteen patients in the control group were treated with metal stents alone. Porfimer sodium (2 mg/kg) was administered intravenously to PDT patients. Fortyeight hours later, PDT was administered using a diffusing fiber that was advanced across the tumor by either endoscopic retrograde cholangiopancreatography or percutaneous cholangiography. After performance of PDT, uncovered metal stents were inserted to ensure adequate decompression and bile drainage. Patient survival rates and cumulative stent patency were calculated using Kaplan-Meier analysis with the log-rank test. RESULTS:The PDT and control patients were comparable with respect to age, gender, health status, pretreatment bilirubin, and hilar CC stage. When compared to control, the PDT group was associated with significantly prolonged stent patency (median 244 ± 66 and 177 ± 45 d, respectively, P = 0.002) and longer patient survival (median 356 ± 213 and 230 ± 73 d, respectively, P = 0.006). Early complication rates were similar between the groups (PDT group 17%, control group 13%) and all patients were treated conservatively. Stent malfunctions occurred in 14 PDT patients (78%) and 12 control patients (80%). Of these 26 patients, twenty-two were treated endoscopically and four were treated with external drainage. CONCLUSION:Metal stenting after one session of PDT may be safe with acceptable complication rates. | 9 | Mitochondria-targeting drug conjugates for cytotoxic, anti-oxidizing and sensing purposes:current strategies and future perspectives显示文摘mitochondrial-targeting moieties to anticancer drugs, antioxidants and sensor molecules. Among them, the most widely applied mitochondrial targeting moiety is triphenylphosphonium(TPP), which is a delocalized cationic lipid that readily accumulates and penetrates through the mitochondrial membrane due to the highly negative mitochondrial membrane potential. Other moieties, including short peptides,dequalinium, guanidine, rhodamine, and F16, are also known to be promising mitochondrial targeting agents. Direct conjugation of mitochondrial targeting moieties to anticancer drugs, antioxidants and sensors results in increased cytotoxicity, anti-oxidizing activity and sensing activity, respectively,compared with their non-targeting counterparts, especially in drug-resistant cells. Although many mitochondria-targeted anticancer drug conjugates have been investigated in vitro and in vivo, further clinical studies are still needed. On the other hand, several mitochondria-targeting antioxidants have been analyzed in clinical phases I, II and III trials, and one conjugate has been approved for treating eye disease in Russia. | 16 | Friction stir welding of 5052 aluminum alloy plates显示文摘Friction stir welding between 5052 aluminum alloy plates with a thickness of 2 mm was performed. The tool for welding was rotated at speeds ranging from 500 to 3000 r/min under a constant traverse speed of 100 mm/min. The results show that at all tool rotation speeds, defect-free welds are successfully obtained. Especially at 1000, 2000 and 3000 r/min, the welds exhibit very smooth surface morphologies. At 500, 1000, and 2000 r/min, onion ring structure is clearly observed in the friction-stir-welded zone (SZ). In addition, the onion ring structure region becomes wider as the tool rotation speed is increased. The gain size in the SZ is smaller than that in the base metal, and is decreased with a decrease of the tool rotation speed. In all tool rotation speeds, the SZ exhibits higher average hardness than the base metal. Especially at 500 r/min, the average hardness of the SZ reaches a level about 33% greater than that of the base metal. At 500, 1 000 and 2 000 r/min, the tensile strength of the friction stir welded (FSWed) plates is similar to that of the base metal (about 204 MPa). The elongation of the FSWed plates is lower than that of the base metal (about 22%). | 18 | 缺氧/厌氧/好氧/预缺氧-MBR组合工艺对低碳氮比市政污水中营养盐去除的研究(英文)显示文摘A novel full scale modified A2O(anoxic/anaerobic/aerobic/pre-anoxic)-membrane bioreactor(MBR) plant combined with the step feed strategy was operated to improve the biological nutrient removal(BNR) from low C/N ratio municipal wastewater in Southern China. Transformation of organic carbon, nitrogen and phosphorus, and membrane fouling were investigated. Experimental results for over four months demonstrated good efficiencies for chemical oxygen demand(COD) and 4NH-N? removal, with average values higher than 84.5% and 98.1%, respectively. A relatively higher total nitrogen(TN) removal efficiency(52.1%) was also obtained at low C/N ratio of 3.82, contributed by the configuration modification(anoxic zone before anaerobic zone) and the step feed with a distribution ratio of 1︰1. Addition of sodium acetate into the anoxic zone as the external carbon source, with a theoretical amount of 31.3 mg COD per liter in influent, enhanced denitrification and the TN removal efficiency in-creased to 74.9%. Moreover, the total phosphate(TP) removal efficiency increased by 18.0%. | 20 | Early treadmill exercise increases macrophage migration inhibitory factor expression after cerebral ischemia/reperfusion显示文摘The neuroprotective function of macrophage migration inhibitory factor(MIF) in ischemic stroke was rarely evaluated.This study aimed to investigate the effects of early treadmill exercise on recovery from ischemic stroke and to determine whether these effects are associated with the expression levels of MIF and brain-derived neurotrophic factor(BDNF) in the ischemic area.A total of 40 male Sprague-Dawley rats were randomly assigned to the ischemia and exercise group [middle cerebral artery occlusion(MCAO)-Ex,n = 10),ischemia and sedentary group(MCAO-St,n = 10),sham-surgery and exercise group(Sham-Ex,n= 10),or sham-surgery and sedentary group(Sham-St,n = 10).The MCAO-Ex and MCAO-St groups were subjected to MCAO for 60 minutes,whereas the Sham-Ex and Sham-St groups were subjected to an identical operation without MCAO.Rats in the MCAO-Ex and Sham-Ex groups then ran on a treadmill for 30 minutes once a day for 5 consecutive days.After reperfusion,the hanging time tested by the wire hang test was longer and the relative fractional anisotropy determined by MRI was higher in the peri-infarct region of the MCAO-Ex group compared with the MCAO-St group.The expression levels of MIF and BDNF in the peri-infarct region were upregulated in the MCAO-Ex group.Increased MIF and BDNF levels were positively correlated with relative fractional anisotropy changes in the peri-infarct region.There was no significant difference in the levels of MIF and BDNF in the peri-infarct region between the Sham-Ex and Sham-St groups.Our study demonstrated that early exercise(initiated 48 hours after the MCAO) could improve motor and neuronal recovery after ischemic stroke.Furthermore,the increased levels of MIF and BDNF in the peri-infarct region(penumbra) may be one of the mechanisms of enhanced neurological function recovery.All experiments were approved by the Institutional Animal Care and Use Committee in Asan Medical Center in South Korea(2016-12-126). | Min Cheol Chang Chae Ri Park Seung Hwa Rhie Woo Hyun Shim Dae Yul Kim | 2019 | Neural Regeneration Research2019,14,7: | 7 |
