1Types and Characteristics of the Lower Silurian Shale Gas Reservoirs in and Around the Sichuan Basin显示文摘This study analyzed the characteristics and types of the Lower Silurian shale gas reservoirs in and around Sichuan Basin through field observations, slices, Ar-ion-beam milling, scanning electron microscopy, and x-ray diffraction analysis of 25 black shale outcrops and samples. Two main types of shale gas reservoirs were determined, i.e., fractures and pores. Fractures were classified into five categories, i.e., giant, large, medium, small, and micro, according to the features of the shale gas reservoirs, effect of fracture on gas accumulation, and fracture nature. Pore types include organic matter pores, mineral pores(mineral surface, intraparticle, interparticle, and corrosional pore), and nanofractures. The various fracture types, fracture scales, pore types, and pore sizes exert different controls over the gas storage and production capacity. Pores serve as a reservoir for gas storage and, the gas storage capacity can be determined using pores; fractures serve as pathways for gas migration, and gas production capacity can be determined using them.NIE Haikuan ZHANG Jinchuan JIANG Shengling 2015Acta Geologica Sinica(English Edition)2015,89,6:17
2Molecular Cloning and Expression Analysis of the ζ-Carotene Desaturase Gene in Chinese kale(Brassica oleracea var.alboglabra Bailey)显示文摘ζ-Carotene desaturase(ZDS)is an important enzyme in carotenoid biosynthesis.Here,the Brassica oleracea var.alboglabra ZDS(Boa ZDS)gene was cloned from Chinese kale via reverse transcription-polymerase chain reaction(RT-PCR)and deposited in Gen Bank(accession number KY662297).The Boa ZDS gene contains an open reading frame of 1 686 bp that encodes a 561-amino acid protein.Sequence analysis indicates that the ZDS protein is apparently conserved during plant evolution and is most closely related to B.oleracea var.capitata and B.rapa.The promoter sequence of the Boa ZDS gene was predicted to harbor several cis-acting elements that are related to light and phytohormone responses.Semiquantitative RT-PCR analysis showed that Boa ZDS expression varied among different developmental stages and organs.Relative ZDS expression remained stable during germination and seedling stages and rapidly increased at the mature leaf stage.The leaves showed the highest ZDS expression levels compared to the other organs.ZDS expression decreased in all flower tissues during blooming.The fused protein of Boa ZDS was obtained by prokaryotic expression.Heterologous expression of Boa ZDS in Escherichia coli confirmed that Boa ZDS encodes a functionalζ-carotene desaturase that increases β-carotene accumulation in E.coli cells harboring a β-carotene-producing plasmid.The findings of the present study provide a molecular basis for the elucidation of ZDS gene function in Chinese kale.SUN Bo ZHANG Fen XUE Shengling CHANG Jiaqi ZHENG Aihong JIANG Min MIAO Huiying WANG Qiaomei TANG Haoru 2018Horticultural Plant Journal2018,4,3:10
3A Sensitive and Specific IgM-ELISA for the Serological Diagnosis of Human Leptospirosis Using a rLipL32/1-LipL21-OmpL1/2 Fusion Protein显示文摘Objective To construct a lipL32/1‐lipL21‐OmpL1/2 fusion gene and its prokaryotic expression system,and to establish an enzyme‐linked immunosorbent assay (ELISA) using the rLipL32/1‐LipL21‐OmpL1/2 fusion antigen of Leptospira interrogans for sensitive and specific detection of IgM in the serum of patients with leptospirosis.Methods lipL32/1‐lipL21‐OmpL1/2 fusion genes were constructed using a primer‐linking PCR.The target recombinant protein antigens,rLipL32/1,rLipL21,rOmpL1/2 and rLipL32/1‐LipL21‐OmpL1/2,were expressed and the purified antigens were then immobilized to the surface of microplate wells for ELISA‐based detection ofIgM in the sera of leptospirosis patients.Results Of 493 acute leptospirosis patients,95.7% and 97.8% were positive by rLipL32/1‐LipL21‐ OmpL1/2‐IgM‐ELISA using different serum dilutions,which was higher than the rLipL32/1‐IgM‐ELISA (93.1% and 90.3%),rLipL21‐IgM‐ELISA (90.3% and 87.0%),and rOmpL1‐IgM‐ELISA (85.6% and 81.1%) (P<0.01).All IgM‐ELISAs tested negative against 56 non‐leptospirosis patients with typhoid fever,hemorrhagic fever or dengue fever.Conclusion Trigeminal fusion antigen increases ELISA sensitivity and the rLipL32/1‐LipL21‐OmpL1/2‐ IgM‐ELISA is a sensitive and specific serological diagnostic method for clinical leptospirosis.SUN AiHua WANG Yuan DU Peng WU ShengLing YAN Jie 2011Biomedical and Environmental Sciences2011,24,3:6
4Experimental and Simulation Studies on Cold Welding Sealing Process of Heat Pipes显示文摘Sealing quality strongly affects heat pipe performance, but few studies focus on the process of heat pipe sealing. Cold welding sealing technology based on a stamping process is applied for heat pipe sealing. The bonding mechanism of the cold welding sealing process(CWSP) is investigated and compared with the experimental results obtained from the bonding interface analysis. An orthogonal experiment is conducted to observe the effects of various parameters, including the sealing gap,sealing length, sealing diameter, and sealing velocity on bonding strength. A method with the utilization of saturated vapor pressure inside a copper tube is proposed to evaluate bonding strength. A corresponding finite element model is developed to investigate the effects of sealing gap and sealing velocity on plastic deformation during the cold welding process. Effects of various parameters on the bonding strength are determined and it is found that the sealing gap is the most critical factor and that the sealing velocity contributes the least effect. The best parameter combination(A_1B_3C_1D_3, with a 0.5 mm sealing gap, 6 mm sealing length, 3.8 mm sealing diameter, and 50 mm/s sealing velocity) is derived within the experimental parameters. Plastic deformation results derived from the finite element model are consistent with those from the experiment. 8Effect of immunopotentiator of the Chinese herbal medicine on the quantity of T lymphocyte in chicken显示文摘In order to study the mechanism of immunopotentiator,the quantity of T lymphocyte was observed.Total 240 1-day chikens were divided into 3 groups randomly:one contrast group and two groups being drunk immunopotentiator according to the concentration 10 mL·L^(-1) and 5 mL·L^(-1) lasting for 48 d.The number of T lymphocyte in blood was measured by E-rosette when the chikens were at 12-,24-,36-,and 48-day.The results showed that the percent of T lymphocyte in the trial group was obviously higher than that of the contrast,and the 10 mL·L^(-1) group was higher than that of the 5 mL·L^(-1) group;the quantity distribution of T lymphocyte in intestinal mucosa lymphoid tissue of 14-day,21-day chiken was surveyed in the contrast group and the 10 mL·L^(-1) group with the method of routine histology-slices and ANAE,the results showed that the quantity of T lymphocyte in the 10 mL·L^(-1) group was significantly higher than that of the contrast,which indicated that immunopotentiator increased markedly the quantity of T lymphocyte,and the 10 mL·L^(-1) group was higher than that of the 5 mL·L^(-1) group.SHI Qiumei SHEN Ping GAO Guisheng ZHAO Ruili TANG Shengling 2007Journal of Northeast Agricultural University(English Edition)2007,14,2:1
