1Heat shock induction of a 65 kDa ATP-binding proteinase in rat C6 glioma cells显示文摘The 45, 55, 65 and 100 kDa ATP-binding proteinases (ATP-BPases) of the heat-shocked (44 ℃ for 30 min, recovery for 12h) rat C6 glioma cells were purified by DEAE-ionexchange and ATP-affinity chromatography. Their molecular masses, isoelectric points (pI), pH-optima and other properties were analyzed by native proteinase gels.It was shown that the 65 kDa ATP-BPase is specifically induced by heat shock and not detectable in control cells.Its N-terminal 1-9 amino acid sequence was determined by Edman degradation, but no homologies to other proteins in the protein data bases were found. 30 and 31 kDa proteinases can be cleaved from the 45, 55 and 65 kDa proteinases to which they are linked. 2微生物解磷特性及其铅污染土壤修复应用显示文摘土壤污染会威胁食品安全与人体健康,中国土壤重金属污染形势较为严峻,其中以矿区污染尤为突出。铅是五大重点监控重金属之一,其毒性极强,危害植物、动物及人体健康。土壤中铅不能被生物降解,但可与磷酸盐形成稳定化合物进行固定,从而降低其生物有效性,降低土壤污染风险和食品安全风险。磷作为植物生长必需大量元素,通常以磷肥形式施加于土壤,大量磷肥转化为难溶性磷酸盐或有机磷,不易被植物吸收利用。解磷微生物可将难溶性有机/无机磷溶解,释放磷酸根,为植物提供磷素的同时,可络合铅生成难溶性磷酸铅沉淀,降低其生物有效性,是生物固铅修复污染土壤的有效途径。因此,该文综述了土壤微生物的解磷效率与机制、磷酸根固铅的作用机理及实际修复应用前景,并提出现存问题及未来研究方向,为提高矿区铅污染土壤生物修复效率提供了基础信息和技术参考。苏奇倩 徐其静 丁豪杰 马晟 李林 Rensing Christopher 刘雪 2020环境科学与技术2020,43,12:7
3中国和美国原始土壤中非高温发现和鉴定显示文摘近年来在非极端环境中已经发现有古菌(Archaea)的存在, 但在中国原始土壤中还未见报道。本研究的目的是调查古菌是否存在于两个分别取自中国新疆和广西的土壤及两个美国亚利桑那州南部地区的土壤中。我们分别构建了这四个原始土壤的古菌16S rDNA文库并对28个克隆的16S rDNA进行了鉴定。所有这些16S rDNA的序列都归类于古菌的泉古菌门(Crenarchaeota)。进化树分析表明, 这些泉古菌的16S rDNA属于非高温陆地环境中的泉古菌种群, 明显区别于海洋和淡水地带的泉古菌种群。这个泉古菌种群又有两个分支, 这两个分支在16S rDNA序列上和G+C含量上有明显的区别。本研究在两个中国和两个美国原始土壤中鉴定了非高温泉古菌的存在, 由此证明泉古菌的存在范围不只局限于高温等极端环境。另外, 美国原始土壤中的泉古菌只属于一个进化分支, 这说明非高温泉古菌种群的类型和土壤的地理位置及土壤特性有关。樊昊心 Derek J. 4梭梭根际促生菌(PGPR)菌肥对番茄产量、品质和土壤特性的影响显示文摘【目的】探究梭梭根际促生菌(PGPR)菌肥对番茄生长、果实产量及根际土壤特性的影响。【方法】课题组前期从梭梭根际土壤中分离得到芽孢杆菌(Bacillus sp.)WM13-24、假单胞菌(Pseudomonas sp.)M30-35和根瘤菌(Rhizobium sp.)WMN-33个菌株,并结合已有的解淀粉芽孢杆菌(Bacillus amyloliquefaciens)GB03和苜蓿中华根瘤菌(Sinorhizobium meliloti)ACCC17578制备成含有5种促生菌的复合菌肥;以芽孢杆菌WM13-24制备单株菌肥;并以LB培养基和微生物肥料巧农2号作为对照进行番茄(Solanum lycopersicum)田间试验。【结果】与LB培养基对照相比,施用复合菌肥后,番茄的产量、可溶性糖含量、可溶性蛋白含量、维生素C含量、有机酸含量、土壤过氧化氢酶活性、脲酶活性、磷酸酶活性分别增加了73.75%、28.53%、15.73%、75.42%、15.15%、15.71%、1.8倍和26.73%。【结论】PGPR菌肥均可显著改善番茄品质,提高番茄产量。与单株菌肥相比,以梭梭根际促生菌为主要菌源制备的复合菌肥显著提高了番茄产量、可溶性糖含量、可溶性蛋白含量、维生素C含量;且优化了番茄根际土壤特性。赵玲玉 索升州 赵祺 姚丹 李慧萍 Christopher Rensing 张金林 2022甘肃农业大学学报2022,57,3:5
5土壤农用地膜微生物降解研究进展显示文摘地膜可保持土壤湿度,调节土壤温度及限制杂草生长从而促进农作物增产,在现代农业生产中具有不可或缺的作用.然而,地膜主要成分聚乙烯(PE)性质稳定,难以降解,极易在农田土壤中残留并积累.此外,地膜在生产过程中添加邻苯二甲酸酯类(PAEs)作为塑化剂,该类有机物极易在土壤和水体环境中积累和迁移,且生物毒性大,对生态环境、粮食安全和人体健康构成极大威胁.聚乙烯和邻苯二甲酸酯复合污染是土壤有机污染治理的重点和难点.因此,农用地膜污染土壤修复是环境科学研究的重要课题,亦是作物生产安全和人类健康的重要保障.微生物降解的生物修复较物理化学技术具有效率高、无二次污染、成本低、环境扰动小等优点,具有广泛应用前景.由此,本文综述农用地膜使用和土壤残留现状及其生物降解的研究进展,以期为地膜污染农田土壤的生物修复提供基础信息和技术参考.丁豪杰 苏奇倩 李林 李晓锋 徐其静 Rensing Christopher 刘雪 2021中国环境科学2021,41,9:4
6降低水稻籽粒镉砷累积的研究进展显示文摘矿山开采、废弃物排放及污水灌溉等引发区域农田土壤重金属污染,并通过食物链富集、传递,严重威胁人类健康。水稻是重要主食,且极易吸收镉(Cd)和砷(As),已成为Cd、As进入人体的主要膳食来源。因此,通过调控措施降低水稻籽粒Cd、As含量,对保障水稻及其制成品的食用安全与人体健康具有重要的现实意义。概述了降低水稻Cd、As含量的调控措施,主要包括:(1)利用生物质炭、铁盐、肥料等材料钝化土壤中Cd、As,降低其生物有效性,从而降低水稻Cd、As吸收;(2)外源添加硫(S)可消除植物的膜脂过氧化胁迫,促进水稻体内植物络合素等非蛋白巯基形成,将Cd固定在水稻组织器官液泡中,减少其向籽粒的转移,降低籽粒Cd积累;(3)外源添加硅(Si),Si可与亚砷酸盐(As^(3+))形成吸收竞争,降低水稻As吸收;(4)控制稻田水分可改变根际土壤氧化还原状态,影响水稻Cd、As吸收积累,同时根系泌氧促进根表铁膜形成,调控土壤氧化还原状态促进As氧化为砷酸盐(As^(5+)),增强根表对As^(5+)的吸附固定,从而降低水稻对As的吸收量;(5)调控转运蛋白表达,敲除Mn转运蛋白基因OsNRAMP5、茎/叶细胞质转运蛋白基因OsLCT1,以及缺失Si外排转运蛋白基因Lsi2,过表达OsHMA3n、OsHMA2、ScAcr3p基因,通过促进As^(3+)外排、降低Cd/As^(3+)向木质部和籽粒转运等过程降低籽粒Cd、As积累;(6)Cd/As低累积品种的筛选与培育。通过农艺措施、基因工程、种质资源筛选等多手段结合运用,选育Cd/As低累积、高产量的水稻品种,合理规划种植模式,有效控制水稻对Cd和As的吸收累积,为中、轻度污染土壤的水稻安全生产提供现实可能。张慧娟 苏奇倩 丁豪杰 李晓锋 徐其静 Rensing Christopher 刘雪 2022生态与农村环境学报2022,38,7:2
7Secondary Cell Wall Deposition in Developing Secondary Xylem of Poplar显示文摘Although poplar is widely used for genomic and biotechnological manipulations of wood, the cellular basis of wood development in poplar has not been accurately documented at an ultrastructural level. Developing secondary xylem cells from hybrid poplar (Populus deltoides x P. trichocarpa), which were actively making secondary cell walls, were preserved with high pressure freezing/freeze substitution for light and electron microscopy. The distribution of xylans and mannans in the different cell types of developing secondary xylem were detected with immunofluorescence and immuno-gold labeling. While xylans, detected with the monoclonal antibody LM10, had a general distribution across the secondary xylem, mannans were enriched in the S2 secondary cell wall layer of fibers. To observe the cellular structures associated with secondary wall production, cryofixed fibers were examined with transmission electron microscopy during differentiation. There were abundant cortical microtubules and endomembrane activity in cells during the intense phase of secondary cell wall synthesis.
