| 1 | Dynamic changes of typeⅠ,Ⅲand N collagen synthesis and distribution of collagen-producing cells in carbon tetrachloride-induced rat liver fibrosis显示文摘AIM To find out the relationship between the gene transcription of different types ofprocollagen and the deposition of the relevant collagens in the liver tissue and to confirm the types of collagen producing cells in liverfibrogenesis.METHODS Dynamic changes of the expression of α1(Ⅰ ), α1 (Ⅲ ) and α1 (Ⅳ) procollagen mRNAand relevant collagens and the distribution ofcollagen producing cells during liver fibrogenesis of rat induced by CCl4 (20 weeks)were investigated with Northern blot analysis,in situ hybridization and immunohistochemicaltechniques.RESULTS The increased expression of α1 (Ⅲ)procollagen mRNA by Northern blot analysis was the most predominant one among the threemRNAs during fibrogenesis. However, theenhanced expression of al (Ⅳ) procollagenmRNA occurred very early while the expressionof α1 (Ⅰ) mRNA was not enhanced much until themiddle stage of the exPeriment. D6smin (Dm)positive hepatic stellate cells (HSCs) and fewmyofibroblasts (MFs) in and around the necrotic areas expressed α1 (Ⅰ), α1 (Ⅲ) and α1 (Ⅳ)procollagen mRNA signals detected by in situhybridization at the early stage of theexperiment. All the three procollagen mRNAsignals thereafter mainly localized in fibroblasts (Fbs) and MFs in fibrotic septa during the middle and late stages of fibrosis, which distributedparallel to the corresponding collagens detected by immunohistochemical study. ln addition, the endothelial cells of sinusoids and the small blood vessels within the septa also showed α1(Ⅳ) procollagen mRNA and type Ⅳ collagen expressionCONCLUSION It is considered that 'HSC-MF-Fb' effect cell system is the major cellularsource of collagen production in liver fibrosis, in which HSCs are collagen producing precursor cells in the early liver fibrogenesis, thereafter the synthesis of type Ⅰ, Ⅲ and Ⅳ collagens (Col Ⅰ, Col Ⅲ and Col Ⅳ) mainly derives fromMFs and Fbs, which play a very important role in the progress of liver fibrosis. | 2 | A study of interaction between surface water and groundwater using environmental isotope in Huaisha River basin显示文摘The surface water and groundwater are important components of water cycle, and the interaction between surface water and groundwater is the important part in water cycle research. As the effective tracers in water cycle research, environmental isotope and hydrochemistry can reveal the interrelationships between surface water and groundwater effectively. The study area is the Huaisha River basin, which is located in Huairou district, Beijing. The field surveying and sampling for spring, river and well water were finished in 2002 and 2003. The hydrogen and oxygen isotopes and water quality were measured at the laboratory. The spatial characteristics in isotope and evolution of water quality along river lines at the different area were analyzed. The altitude effect of oxygen isotope in springs was revealed, and then using this equation, theory foundation for deducing recharge source of spring was estimated. By applying the mass balance method, the annual mean groundwater recharge rate at the catchment was estimated. Based on the groundwater recharge analysis, combining the hydrogeological condition analysis, and comparing the rainfall-runoff coefficients from the 1960s to 1990s in the Huaisha River basin and those in the Chaobai River basin, part of the runoff in the Huaisha River basin is recharged outside of this basin, in other words, this basin is an un-enclosed basin. | 3 | Development of distributed time-variant gain model for nonlinear hydrological systems显示文摘In this paper, a rainfall-runoff modeling system is developed based on a nonlinear Volterra functional series and a hydrological conceptual modeling approach. Two models, i.e. the time-variant gain model (TVGM) and the distributed time-variant gain model (DTVGM) that are built on the platform of Digital Elevation Model (DEM), Remote Sensing (RS) and Unit Hydro-logical Process were proposed. The developed DTVGM model was applied to two cases in the Heihe River Basin that is located in the arid and semiarid region of northwestern China and the Chaobai River basin located in the semihumid region of northern China. The results indicate that, in addition to the classic dynamic differential approach to describe nonlinear processes in hy-drological systems, it is possible to study such complex processes through the proposed sys-tematic approach to identify prominent hydrological relations. The DTVGM, coupling the advan-tages of both nonlinear and distributed hydrological models, can simulate variant hydrological processes under different environment conditions. | 4 | A novel gene delivery system targeting cells expressing VEGF receptors显示文摘Two ligand oligopeptides GV1 and GV2 were designed according to the putative binding region of VEGF to its receptors. GV1, GV2 and endosome releasing oligopeptide HA20 were conjugated with poly-L-lysine or protamine and the resulting conjugates could interact with DNA in a noncovalent bond to form a complex. Using pSV2-β-galactosidase as a reporter gene, it has been demonstrated that exogenous gene was transferred into bovine aortic arch-derived endothelial cells (ABAE) andhuman malignant melanoma cell lines (A375) in vitro. In vivo experiments, exogenous gene was transferred into tumor vascular endothelial cells and tumor cells of subcutaneously transplanted human colon cancer LOVO, human malignant melanoma A375 and human hepatoma graft in nude mice. This system could also target gene to intrahepatically transplanted human hepatoma injected via portal vein in nude mice. | 6 | Proteomic analysis of mitochondria reveals a metabolic switch from fatty acid oxidation to glycolysis in the failing heart显示文摘This work characterizes the mitochondrial proteomic profile in the failing heart and elucidates the molecular basis of mitochondria in heart failure. Heart failure was induced in rats by myocardial infarction, and mitochondria were isolated from hearts by differential centrifugation. Using two-dimensional gel electrophoresis and matrix-assisted laser desorption/ionization-time of flight mass spectrometry, a system biology approach was employed to investigate differences in mitochondrial proteins between normal and failing hearts. Mass spectrometry identified 27 proteins differentially expressed that involved in energy metabolism. Among those, the up-regulated proteins included tricarboxylic acid cycle enzymes and pyruvate dehydrogenase complex subunits while the down-regulated proteins were involved in fatty acid oxidation and the OXPHOS complex. | 7 | A study of scale effect on specific sediment yield in the Loess Plateau,China显示文摘Based on data from 148 hydrometric stations in the Yellow River Basin, an analysis of regional scale relationship, or the relationship between specific sediment yield and drainage basin area, has been undertaken in the study area of the Loess Plateau. For different regions, scale relationship in log-log ordinate can be fitted by two types of lines: straight and parabola, and for each line, a function was fitted using regression analysis. The different scale relationships have been explained in terms of the difference in surface material distribution and landforms. To offset the scale-induced influence, calcu-lation has been done based on the fitted functions, in order to adjust the data of specific sediment yield to a common standard area. Based on the scaled data, a map of specific sediment yield was con-structed using Kriging interpolation. For comparison, a map based on the un-scaled data of specific sediment yield was also constructed using the same method. The two maps show that the basic pattern of specific sediment yield was basically the same. The severely eroded areas (Ys >10000 t km-2a-1) were at the same locations from Hekouzhen to Longmen in the middle Yellow River Basin. | 8 | Hyperconcentrated flows as influenced by coupled wind-water processes显示文摘Using data from more than 40 rivers in the middle Yellow River basin, a study has been made of the influence of coupled wind-water processes on hyperconcentrated flows. A simple “vehicle” model has been proposed to describe hyperconcentrated flows. The liquid phase of two-phase flows is a “vehicle”, in which coarse sediment particles are carried as solid-phase. The formation and characteristics of hyperconcentrated flows are closely related with the forma-tion and characteristics of this liquid-phase and solid-phase. Surface materials and geomorphic agents of the middle Yellow River basin form some patterns of combination, which have deep influence on the formation and characteristics of liquid- and solid-phases of hyperconcentrated flows. The combination of high percentages of relatively coarse material with low percentages of fine material appears in the area predominated by the wind process, where the supply of rela-tively coarse sediment is sufficient, but the supply of relatively coarse sediment is not. The com-bination of low percentages of relatively coarse material with high percentages of fine material appears in the area predominated by the water process, where the supply of fine sediment is sufficient, but the supply of fine sediment is not. In the area predominated by coupled wind-water processes appears the combination of medium percentages of coarse and fine materials, and thus both coarse and fine sediments are in relatively sufficient supply. The manner in which the mean annual sediment concentrations of liquid- and solid-phases vary with total suspended sediment concentration is different. With the increased total suspended sediment concentration, mean annual sediment concentration of liquid-phase increased to a limit and then remained constant; however, mean annual sediment concentrations of solid-phase in-creased continuously. Thus, the magnitude of total suspended sediment concentration depends on the supply conditions of relatively coarse sediment and the ability of the flow to carry these relatively coarse sediment particles. In the area predominated by wind process, both the liquid- and the solid-phases cannot develop well, and their concentrations are low. In the area pre-dominated by the water process, the mean annual sediment concentrations of liquid- and solid-phases are also low. Only in the area predominated by coupled wind-water processes, can the conditions most favor the development of both the liquid- and solid-phases, and then the peaks of mean annual sediment concentrations of liquid- and solid-phases appear. Low values of suspended sediment concentrations appear in the areas predominated by the wind process or by the water process, a fact indicating that the predominating wind process or water process does not favor the development of hyperconcentrated flows. Considering flood forecasting’s uncertainty and updating demand, a hybrid hydrological model, namely Differential Hy- drological Grey Model with self-memory function (DHGM-SM), is developed. The model has two fold features. One is to establish a self-memorization equation linked with DHGM, that could extract useful information from past data series and realize updating of hydrological dynamic process. The other is that this model has higher efficiency relative to original hydrological model without self-memory func- tion. This approach was applied to river flow forecasting of two representative basins in Tunxi of South China and Daqinggou of North China. | 10 | The promoter analysis of the human C17orf25 gene, a novel chromosome 17pl3.3 gene显示文摘The human C17orf25 gene (Accession No. AF177342) is one of thirteen genes cloned from a region displaying a high score of loss of heterozygosity within chromosome 17p13.3 in human hepatocellular car-cinoma in China[l]. To unveil the underlying mechanisms for the transcription regulation of this gene and understand its implication to the hepatocellular carcinogenesis, we looked into the relevant aspects by both bioinformatic and experimental executions. We found: 1, The abundant expression of the C17orf25 gene was evident in all the cell lines and tissue samples tested, showing little hepatoma-selectivity; 2, Its tran-scription starts at a single site, locating at -60 from the translation initiation codon; 3, A 58 bp fragment containing the transcription start, extending from -112 to -55, represents the minimal promoter; 4, The consensus sequence within this fragment recognized by SP1 contributes predominantly to the activity of the minimal promoter; 5, The bioinformatic analysis suggests that the C17ort25 gene may encode a protein in the family of the glyoxalase. | 11 | The impact of vegetation restoration on erosion-induced sediment yield in the middle Yellow River and management prospect显示文摘According to the characteristics of water and erosion environments of different natural zones on the Loess Plateau, this paper studies changes of vegetation types, distribution boundaries of forest and grass as well as restoration capacity of vegetation in different natural zones in the middle Yellow River. The annual precipitation of 530 mm is the critical annual pre-cipitation for forest and grass distribution in the middle Yellow River. Among the zonal and azonal environmental factors affecting watershed sediment yield, the intermediate diameter D50 (mm) of suspended load and forest coverage (V, %) play the leading role. Of them the effect weight of forest coverage (V, %) on catchments sediment yield is only 3.4% less than the role of the intermediate diameter (D50, mm), they are almost the same. To effectively control soil erosion in semiarid, especially in hilly-gullied areas and make sediment transport modulus reduce to less than 6000 t/km2, it is rather difficult by merely relying on natural restoration of forest. In the process of cultivated land conversion into forest land and grassland, measures suiting local conditions should be adopted in tree species selection and artificial afforestation (grass planting) based on management with biological measures for slopeland and engineering measures for hilly-gullied areas, so that watershed forest coverage in key counties can reach at least over 30%.Compared with the standard period of precipitation prior to the 1960s, with the decrease of an-nual precipitation at various periods, plant productivities decline to different degrees under natural conditions. The main reason accountable for the low survival rate of new seedlings and grass over years is due to precipitation decrease. | 12 | Coupling the hydrological and ecological process to implement the sustainable water resources management in Hanjiang River Basin显示文摘Research on the combination of hydrological variation and ecological demands plays an important role in water availability assessment and sustainable management on basin scale. An integrative frame, consisting of hydrological regime modelling, ecological water demands estimation and renewable resources management, is given in this paper in order to support the implementation of the sustainable water resources management. The suggested integrative frame has been used to study the integrated water resources management in southern Hanjiang River Basin which is the water source of South-toNorth Water Transportation Project in China. SWAT (soil and water assessment tool) model was used to simulate the monthly averaging streamflow and the amounts of ecological water demands in stream were also estimated in order to evaluate the available water resources for the local users and the transportation project. Then we formed the developing scenarioes by coupling the available water resources, the recovering rate of water resources in natural water cycle, the local water use rate and the amounts of the water transported to North, and used the Scheafer model to analyze their evolving trajectories. Eventually, the sustainable water resources management measures were assembled by the comprehensive evaluation of the scenarioes. | 13 | Ecological hydraulic radius model to calculate instream flow requirements for transporting sediment in the western water transfer region显示文摘Transporting sediment is a natural function of the river. To maintain the normal evolution of the river certain amount of water flow is required, which is called the instream flow requirements for transporting sediment (IFRTS). We defined the permitting flow velocity by the conception of IFRTS, and also put forward the ecological hydraulic radius model (EHRM) to estimate IFRTS. The calculating process of EHRM is explained by the example of Daofu Hydrological Station on Xianshui branch of Yalong River in the west line first-stage construction of South-North Water Transfer Project. The result shows that the IFRTS occupied 29.7%―59.5% of annual mean discharge in flood season, the average of IFRTS was about 100.2 m3/s during 1966―1987, it is close to the IFRTS 90 m3/s calculated by IFRTS conception. | 15 | A distributed hydrological model with its application to the Jinghe watershed in the Yellow River Basin显示文摘For the purpose of water resources management in the Yellow River Basin with highly spatial difference, a daily distributed hydrological model was proposed, of which the determination of spatially-distributed parameters and model inputs processing were performed by means of GIS/RS. In the model, the computation of runoff yield was based on the topography index method and flow routing was modeled by Maskingum method. The operation of the model is followed by means of “command structure” technique based upon the topography of river network. A case study using the model was conducted for the Jinghe watershed, which locates at the middle Yellow River Basin. | 17 | Translational pancreatic cancer research:a comparative study on patient-derived xenograft models显示文摘AIM To assess the viability of orthotopic and heterotopic patient-derived pancreatic cancer xenografts implanted into nude mice.METHODS This study presents a prospective experimental analytical follow-up of the development of tumours in mice upon implantation of human pancreatic adenocarcinoma samples. Specimens were obtained surgically from patients with a pathological diagnosis of pancreatic adenocarcinoma. Tumour samples from pancreatic cancer patients were transplanted into nude mice in three different locations(intraperitoneal, subcutaneous and pancreatic). Histological analysis(haematoxylin-eosin and Masson's trichrome staining) and immunohistochemical assessment of apoptosis(TUNEL), proliferation(Ki-67), angiogenesis(CD31) and fibrogenesis(α-SMA) were performed. When a tumour xenograft reached the target size, it was reimplanted in a new nude mouse. Three sequential tumour xenograft generations were generated(F1, F2 and F3).RESULTS The overall tumour engraftment rate was 61.1%. The subcutaneous model was most effective in terms of tissue growth(69.9%), followed by intraperitoneal(57.6%) and pancreatic(55%) models. Tumour development was faster in the subcutaneous model(17.7 ± 2.6 wk) compared with the pancreatic(23.1 ± 2.3 wk) and intraperitoneal(25.0 ± 2.7 wk) models(P = 0.064). There was a progressive increase in the tumour engraftment rate over successive generations for all three models(F1 28.1% vs F2 71.4% vs F3 80.9%, P < 0.001). There were no significant differences in tumour xenograft differentiation and cell proliferation between human samples and the three experimental models among the sequential generations of tumour xenografts. However, a progressive decrease in fibrosis, fibrogenesis, tumour vascularisation and apoptosis was observed in the three experimental models compared with the human samples.
