4Molecular mechanism of action of anti-tumor necrosis factor antibodies in inflammatory bowel diseases显示文摘Anti-tumor necrosis factor(TNF) antibodies are successfully used in the therapy of inflammatory bowel diseases(IBD). However, the molecular mechanism of action of these agents is still a matter of debate. Apart from neutralization of TNF, influence on the intestinal barrier function, induction of apoptosis in mucosal immune cells, formation of regulatory macrophages as well as other immune modulating properties have been discussed as central features. Nevertheless, clinically effective anti-TNF antibodies were shown to differ in their mode-of-action in vivo and in vitro. Furthermore, the anti-TNF agent etanercept is effective in the treatment of rheumatoid arthritis but failed to induce clinical response in Crohn's disease patients, suggesting different contributions of TNF in the pathogenesis of these inflammatory diseases. In the following, we will review different aspects regarding the mechanism of action of anti-TNF agents in general and analyze comparatively different effects of each antiTNF agent such as TNF neutralization, modulation of the immune system, reverse signaling and induction of apoptosis. We discuss the relevance of the membranebound form of TNF compared to the soluble form for the immunopathogenesis of IBD. 5In vitro antioxidant activities of ethanol extract from Enhalus acoroides (L.F.) Royle显示文摘Objective:To search for new sources of safe and inexpensive antioxidants,the leaf,root and rhizome of Enhalus acoroides(E.acoroides) were screened for the first time for their antioxidant activities.Methods:Samples of leaf,root and rhizome from E.acoroides were tested for total phenolic content,proanthocyanidins and in vitro antioxidant activity in terms of total antioxidant assay,DPPH assay and FRAP assay.Results:The leaf sample was found to have high levels of phenolic(0.323±0.028 mg TAE/g) and proanthocyanidins(0.570 0±0.000 3 mg TAE/g) when compared to root and rhizome.The leaf samples exhibited higher total antioxidant activity (11.770±0.026 mg Ascorbic acid equivalent/g),higher percentage of DPPH radical scavenging activity(25.76±0.04) and higher reducing power(18.060±0.073) in terms of mg GAE/g.In addition, there was a significant correlation between total phenolic content and total antioxidant activity (R^2=0.923),DPPH assay(R^2=0.509).FRAP assay(R^2=0.994).Similarly,significant correlation was found between the proanthocyanidin and total antioxidant activity(R^2=0.977),DPPH assay (R^2=0.464) and the FRAP assay(R^2=0.998).Conclusions:These results suggested that E.acoroides have strong antioxidant potential.Further study is necessary for isolation and characterization of the active antioxidant agents,which can be used to treat various oxidative stress-related diseases.Rengasamy Ragupathi Raja Kannan Rajasekaran Arumugam Perumal Anantharaman 2010Asian Pacific Journal of Tropical Medicine2010,3,11:15
6Phytochemical and anti-bacterial activity of epidermal glands extract of Christella parasitica (L.) H. Lev.显示文摘Objective:To study the morphology,biochemistry and bioactivity of the epidermal glands of the glandular morphotype of Christella parasitica(C.parasitica)(L.) H.Lev.Methods: Morphological studies on epidermal glands were earned out by using light microscope and scanning electron microscope.To prepare the extract,the shade-dried fronds of glandular morphotype were soaked in acetone.For antibacterial studies paper disc method was followed by using various pathogenic bacteria.Results:Detailed micromorphological,phytochemical and bioactivity studies on a medicinal fern C.parasitica(L.) H.Lev.showed its inlraspecific variation in antibacterial activity.The presence or absence of the epidermal glands was the key factor for antibacterial activity in the morphovariants of this species.The epidermal glands were orange-coloured,stalked and elongated ones of about 84.2μm×45μm,and distributed on the undersurface of cosla,coslules and veins in croziers,young and mature leaves.Frequency of glands varied from 15/cm on costa in mature leaves to 140/cm on costules in croziers.The acetone extract of the glands showed antibacterial activities and also toxic effect against mosquito larvae and tadpoles of frog.Preliminary phytochemical analysis and HPLC studies of the gland extract showed the presence of various kinds of terpenoids,alkaloids,tannins,saponins and flavonoids in it.Conclusions:The present study shows that epidermal glands of the glandular morphotype of C. parasitica(L.) Systematic prediction of structural integrity of critical structures such ascombustion chambers,pressure vessels,nuclear reactor components,boilers etc.,ensures the human safety,environmental protection,and the economical considerations.The present work aims at prediction of fatigue behaviour of symmetric structures like pressure vessels in the presence of common welding defects such as lack of fusion( LOF),lack of penetration( LOP) and porosity.A ring type specimen which replicates the stress pattern in thepressure vessel is considered for the study of severity of weld imperfections. 8Isolation and characterization of potential antibiotic producing actinomycetes from water and sediments of Lake Tana,Ethiopia显示文摘Objective:To isolate,evaluate and characterize potential antibiotic producing actinomycetes from water and sediments of Lake Tana,Ethiopia.Methods:A total of 31 strains of actinomycetes were isolated and tested against Gram positive and Gram negative bacterial strains by primary screening.In the primary screening.11 promising isolates were identified and subjected to solid state and submerged state fermentation methods to produce crude extracts.The fermented biomass was extracted by organic solvent extraction method and tested against bacterial strains by disc and agar well diffusion methods.The isolates were characterized by using morphological,physiological and biochemical methods.Results:The result obtained from agar well diffusion method was belter than disc diffusion method.The crude extract showed higher inhibition zone against Gram positive bacteria than Cram negative bacteria.One-way analysis of variance confirmed most of the crude extracts were statistically significant at 95%confidence interval.The minimum inhibitor)concentration and minimum bactericidal concentration of crude extracts were 1.65 mg/mL and 3.30 mg/mL against Staphylococcus aureus,and 1.84 mg/mL and 3.80 mg/mL against Escherichia coli respectively.The growth of aerial and substrate mycelium varied in different culture media used.Most of the isolates were able to hydrolysis starch and urea:able to survive at 5%concentration of sodium chloride:optimum temperature for their growth was 30°C.Conclusions:The results of the present study revealed that freshwater actinomycetes of Luke Tana appear to have immense potential as a source ol antibacterial compounds.Gebreselema Gebreyohannes Feleke Moges Samuel Sahil Nagappan Raja 2013Asian Pacific Journal of Tropical Biomedicine2013,3,6:12
9Inverted internal limiting membrane flap technique for very large macular hole显示文摘AIM: To assess the anatomical and visual outcome of idiopathic macular holes greater than 1000 μm using the inverted internal limiting membrane flap technique.METHODS: This retrospective case series included 5 eyes of 5 patients with idiopathic macular hole with base diameter greater than 1000 μm who underwent inverted internal limiting membrane flap technique along with standard 23 G pars plans vitrectomy with posterior hyaloid detachment and fluid gas exchange with 12%-14% perfluoropropane(C3F8). Preoperative and postoperative visual acuity and spectral domain optical coherence tomography images were evaluated. The main outcome measures were visual outcome and macular hole closure.RESULTS: Mean age was 63.2±8.4y with all 5 subjects being females. Mean duration of symptoms was 11±14 mo with a mean postoperative follow up of 13.2 ±13 mo. The mean base diameter of the macular holes was 1420±84.8 μm(1280-1480 μm). Type 1 closure was achieved in four out of five patients, while one patient had type 2 closure using the inverted internal limiting membrane(ILM) flap technique. Median baseline BCVA was 0.79 log MAR(Snellen's equivalent 20/120) and median final BCVA0.6 log MAR(Snellen's equivalent 20/80) with mean visual improvement of approximately three lines improvement. 10Role of interventional radiology in the management of acute gastrointestinal bleeding显示文摘Acute gastrointestinal bleeding(GIB) can lead to significant morbidity and mortality without appropriate treatment. There are numerous causes of acute GIB including but not limited to infection, vascular anomalies, inflammatory diseases, trauma, and malignancy. The diagnostic and therapeutic approach of GIB depends on its location, severity, and etiology. The role of interventional radiology becomes vital in patients whose GIB remains resistant to medical and endoscopic treatment. Radiology offers diagnostic imaging studies and endovascular therapeutic interventions that can be performed promptly and effectively with successful outcomes. Computed tomography angiography and nuclear scintigraphy can localize the source of bleeding and provide essential information for the interventional radiologist to guide therapeutic management with endovascular angiography and transcatheter embolization. 12Structure-based Comparative Analysis and Prediction of N-linked Glycosylation Sites in Evolutionarily Distant Eukaryotes显示文摘The asparagine-X-serine/threonine (NXS/T) motif, where X is any amino acid except proline, is the consensus motif for N-linked glycosylation. Significant numbers of high-resolution crystal structures of glycosylated proteins allow us to carry out structural analysis of the N-linked glycosylation sites (NGS). Our analysis shows that there is enough structural information from diverse glycoproteins to allow the development of rules which can be used to predict NGS. A Python-based tool was developed to investigate asparagines implicated in N-glycosylation in five species: Homo sapiens, Mus musculus, Drosophila melanogaster, Arabidopsis thaliana and Saccharomyces cerevisiae. Our analysis shows that 78% of all asparagines of NXS/T motif involved in N-glycosylation are localized in the loop/turn conformation in the human proteome. Similar distribution was revealed for all the other species examined. Comparative analysis of the occurrence of NXS/T motifs not known to be glycosylated and their reverse sequence (S/TXN) shows a similar distribution across the secondary structural elements, indicating that the NXS/T motif in itself is not biologically relevant. Based on our analysis, we have defined rules to determine NGS. Using machine learning methods based on these rules we can predict with 93% accuracy if a particular site will be glycosylated. If structural information is not available the tool uses structural prediction results resulting in 74% accuracy. The tool was used to identify glycosylation sites in 108 human proteins with structures and 2247 proteins without structures that have acquired NXS/T site/s due to non-synonymous variation. 13Assessment of heavy metal pollution from the sediment of Tupilipalem Coast,southeast coast of India显示文摘Sediment from twelve stations was sampled from the Tupilipalem Coast, southeast coast of India, and the presence of a set of heavy metals was established including iron(Fe), manganese(Mn), chromium(Cr),copper(Cu), nickel(Ni), lead(Pb), zinc(Zn) and cadmium(Cd). The heavy metals were assessed by factor analysis, the results of which showed positive and/or negative correlations among Fe, Mn, Cr, Cu, Ni, Pb,Zn, and Cd. Factor analysis also indicated that heavy metals in the sediments of the study area have different natural and anthropogenic sources. Similarly, a sediment pollution assessment was done using the Geoaccumulation Index(Igeo), Enrichment Factor(EF), and Pollution Load Index(PLI). The Geoaccumulation Index indicated that the surface sediment of the Tupilipalem Coast was extremely contaminated with Fe, Mn, Cr, Cu, Ni, Pb, and Zn. The calculation of enrichment factors showed a significant enrichment with respect to Pb, Zn, and Cd and a moderate enrichment with Cr, Cu, and Ni. The falling trend of average contents' enrichment factors is Cd > Pb > Zn > Cu > Cr > Ni > Mn > Fe. The PLI values of the Cd show higher(> 1) values due to the influence of distinct external sources like agricultural runoff, industrial activities, and other anthropogenic inputs. Ninety two percent of heavy metals under study showed the highest concentrations at station TP-5 where the Buckingham Canal and other agricultural and aquacultural effluents connect with the Bay of Bengal. 14经口内镜下肌切开术治疗贲门失弛缓症的临床价值分析显示文摘目的本研究旨在探讨经口内镜下肌切开术(POEM)治疗贲门失弛缓症(AC)的短期及中期疗效、分析相关并发症情况。方法回顾性分析兰州大学第二医院2012年1月至2018年4月消化内镜中心行POEM治疗的59例患者,观察临床疗效及并发症,对比分析AC治疗前后临床症状评分系统(Eckardt)的变化。结果59例患者中,病程4~380个月,手术成功率为100.0%,随访时间6~89个月。术前Eckardt评分(6.45±1.63)分,术后1月评分(0.52±0.56)分(P<0.05),术后6月Eckardt评分(0.54±0.59)分(P<0.05)。术后12月随访47例,Eckardt评分(0.61±0.70)分;术后体重增加:(9.92±5.97)kg,术后出现气体相关并发症6例(10.16%),穿孔1例(1.69%),均成功保守治疗;胃食管反流23例(38.9%)。结论POEM治疗AC安全有效,短期疗效显著,中期疗效良好。冯洁 Raja Waqar Ali 齐晔 王燕 郝晋雍 高丽萍 黄晓俊 2019现代消化及介入诊疗2019,24,11:7
15Efficacy of peroral endoscopic myotomy vs other achalasia treatments in improving esophageal function显示文摘AIM: To assess and compare the esophageal function after peroral endoscopic myotomy(POEM) vs other conventional treatments in achalasia.METHODS: Chart review of all achalasia patients who underwent POEM, laparoscopic Heller myotomy(LHM) or pneumatic dilation(PD) at our institution between January 2012 and March 2015 was performed. Patient demographics, type of achalasia, prior treatments, pre- and post-treatment timed barium swallow(TBE) and high-resolution esophageal manometry(HREM) findings were compared between the three treatment groups. Patients who had both pre- and 2 mo posttreatment TBE or HREM were included in the final analysis. TBE parameters compared were barium column height, width and volume of barium remaining at 1 and 5 min. HREM parameters compared were basal lower esophageal sphincter(LES) pressures and LES-integrated relaxation pressures(IRP). Data are presented as mean ± SD, median [25th, 75 th percentiles] or frequency(percent). Analysis of variance, KruskalWallis test, Pearsons χ~2 test and Fishers Exact tests were used for analysis.RESULTS: A total of 200 achalasia patients were included of which 36 underwent POEM, 22 underwent PD and 142 underwent LHM. POEM patients were older(55.4 ± 16.8 years vs 46.5 ± 15.7 years, P = 0.013) and had higher BMI than LHM(29.1 ± 5.9 kg/m^2 vs 26 ± 5.1 kg/m^2, P = 0.012). More number of patients in POEM and PD groups had undergone prior treatments compared to LHM group(72.2% vs 68.2% vs 44.3% respectively, P = 0.003). At 2 mo post-treatment, all TBE parameters including barium column height, width and volume remaining at 1 and 5 min improved significantly in all three treatment groups(P = 0.01 to P < 0.001) except the column height at 1 min in PD group(P = 0.11). At 2 mo post-treatment, there was significant improvement in basal LES pressure and LES-IRP in both LHM(40.5 mm Hg vs 14.5 mm Hg and 24 mm Hg vs 7.1 mm Hg respectively, P < 0.001) and POEM groups(38.7 mm Hg vs 11.4 mm Hg and 23.6 mm Hg vs 6.6 mm Hg respectively, P < 0.001). However, when the efficacy of three treatments were compared to each other in terms of improvement in TBE or HREM parameters at 2 mo, there was no significant difference(P > 0.05).CONCLUSION: POEM, PD and LHM were all effective in improving esophageal function in achalasia at shortterm. 16Seroepidemiology of bluetongue disease in small ruminants of northeast of Iran显示文摘Objective:To estimate the prevalence and distribution of bluetongue vims antibody in sheep and goats in 25 townships of Khorasan Razavi.Bluetongue is an infectious,non-contagious,arthropod born viral disease of ruminants and has been reported from most of the tropical and subtropical regions of the world.Methods:A total number of 1034 serum samples from sheep and goats were collected and transmitted to Serological Laboratory of Veterinary Council of Khorasan Razavi.Serums were screened for the presence of group-specific bluetongue virus antibody using competitive Enzyme Linked Immuno Sorbent Assay(c-ELISA).Kesults:The seropositivitv of sheep and goats for bluetongue was found to be 89.2%.The highest prevalence rate was seen in Taybad.Khalil-abad and Torbat-jam(100%)and the least prevalence rate was seen in Jovein(55%).Conclusions:The results showed that the majority of animals in the north-east of Iran are infected with bluetongue vims.High correlation between abortion history and seroposivity emphasize the economical importance of bluetongue virus in the sheep herds of the region.Vahid Najarnezhad Mahin Rajae 2013Asian Pacific Journal of Tropical Biomedicine2013,3,6:7
17Antibacterial potential of three seagrasses against human pathogens显示文摘Objective:To evaluate the antibacterial activity of Halophila stipulacea(H.stipulacea), Cymodocea serrulata(C.serrulata) and Halodule pinifolia(H.pinifolia) against seven human bacterial pathogens.Methods:The antibacterial activities of the extracts on the various test organisms using disc diffusion method and Minimum Inhibitory Concentraction(MIC).Results: Methanol and chloroform extracts of all the three seagrasses were active against all the tested pathogens,whereas the hexane extract of seagrasses was not active against Staphylococcus aureus (S.aureus).Antibacterial activity of three seagrass screened,was in the order of H.pinifolia > H.stipulacea.> C.serrulata.Conclusions:This antibacterial studies can further investigated on seagrasses for purification of bioactive substance and its possible utility in disease control.Rengasamy Ragupathi Raja Kannan Rajasekaran Arumugam Perumal Anantharaman 2010Asian Pacific Journal of Tropical Medicine2010,3,11:7
18Antibacterial activity of selected ethnomedicinal plants from South India显示文摘Objective:To screen the antimicrobial potential of three ethnomedicinal plants Chassalia curviflora Thw.(C.curviflora),Cyclea peltata Hook.F.& Thomson(C.peltata) and Euphorbia hirta L(E.hirta) used in folk medicines in Aarukani hills Kani tribe,Tamil Nadu,India against human bacterial pathogens.Methods:Antibacterial efficacy was performed by disc diffusion method against the pathogens viz.,Escherichia coli(E.coli)(ATCC 35218),Staphylococcus aureus (S.aureus)(ATCC 6538),Salmonella typhi(S.typhi)(MTCC 733),Proteus vulgaris(P.vulgaris), Proteus mirabilis(P.mirabilis) and Streptococcus pyogenes(S.pyogenes) and incubated for 24 h at 37°C.Results:The maximum degree of antibacterial activity was observed in C.peltata followed by C.curviflora.While E.hirta showed comparatively low degree of antibacterial activity.The methanolic extract of C.peltata showed the antibacterial activity against three pathogens viz.,S.pyogenes,P.vulgaris and E.coli with the inhibition zones 12 mm,10 mm and 9 mm,respectively,hexane extracts of C.peltata also showed the antibacterial activity against two selected pathogens viz.,P.vulgaris and P.mirabilis with 15 mm and 12 mm of inhibition zones.All the three different concentrations(025,0.50 & 0.75 mg/mL) of methanolic extract of C. peltata show the inhibitory effect on the three susceptible bacteria S.pyogenes,P.vulgaris and E. coli with the maximum inhibition in the highest concentration(0.75 mg/mL).The methanolic and hexane extracts of C.curviflora exhibited the antibacterial activity against only one bacterium each i.e.P.vulgaris and S.typhi with the maximum zone of inhibition 13 and 11 mm respectively. The methanolic and hexane extracts of E.hirta exhibited the antibacterial activity against only one bacterium Le.S.pyogenes with the maximum zone of inhibition 13 and 11 mm respectively. 20An ethnobotanical survey of medicinal plants used by ethnic people in West and South district of Tripura, India显示文摘An ethno-medicinal investigation was conducted to highlights the traditional knowledge of medicinal plants being used by the tribe in West and South district of Tripura. This paper provides information about the different uses of plants used in their primary health care system. Tripura is a small north-eastern state of India and also a part of both Himalayan and Indo-Burma biodiversity region. It is a goldmine of medicinal plants and use of different plants in tribal traditional heath care systems has long history. Nineteen different tribes in Tripura, depend on natural resources at a great extent. This paper documented 113 medicinal plant species from 56 families along with their botanical name, local name, family name, habit, medicinal parts used, and traditional usage of application. The dominant families are Euphorbiaceae (7 species), Apocynaceae (6 species), Fabaceae and Rubiaceae (5 species each), Caesalpiniaceae, Asteraceae, Liliaceae and Verbenaceae (4 species each), Combretaceae, Labiatae, Malvaceae, Rutaceae and Zingiberaceae (3 species each). Tribes of Tripura have rich traditional knowledge on plant based medicine.
