2中间偃麦草的GISH分析显示文摘以染色体组为 Ee Ee 的二倍体长穗偃麦草 (Thinopyrum elongatum,2 n=2 x=1 4)、染色体组为 Eb Eb的二倍体比萨偃麦草 (Th.bessarabicum,2 n=2 x=1 4)、染色体组为 St St St St的四倍体拟鹅冠草 (Pseudoroegneiria strigosa,2 n=4x=2 8)的总基因组 DNA为探针 ,对中间偃麦草 (Th.intermedium)进行 GISH分析。结果表明 ,中间偃麦草是由 2个亲缘关系较近的染色体组、1个亲缘关系较远的染色体组构成 ;中间偃麦草所含的亲缘关系较近的染色体组分别与二倍体长穗偃麦草染色体组 Ee、比萨偃麦草染色体组 Eb、以及 1个亲缘关系较远的染色体组与拟鹅冠草染色体组 St 基本相似 ,但不完全一样。因而 ,中间偃麦草的染色体组用Eet Eet Ebt Ebt St吉万全 薛秀庄 王秋英 王长有 FEDAK George PATROSKI Rene 刘广田 2001西北植物学报2001,21,3:29

6Spatial patterns of plant diversity and communities in Alpine ecosystems of the Hengduan Mountains, northwest Yunnan, China显示文摘Aims Conduct a quantitative,but rapid,regional-level assessment of the alpine flora across northwest Yunnan(NWY)to provide a broadbased understanding of local and regional patterns of the composition,diversity and health of alpine ecosystems across NWY.Methods A stratified random sampling design was employed to select sites across the different mountain ranges of NWY.Vegetation was sampled by stratifying each site by the three major alpine vegetation community types:meadow,dwarf shrub and scree.Two 50-m transects were randomly located within each community type at each sampling site with 101-m^(2) subplots systematically placed along each transect.Environmental variables were recorded at each transect.Multivariate analyses were used to classify the major plant community assemblages and link community patterns to environmental and habitat variables.Important Findings Forb species richness varied from 19 to 105 species per site(21 sites total)with an average of 59 species per site(60 m^(2) sampled per site).Most species were patchily distributed with narrow distributions and/or small population sizes;over half the species occurred at only one or two sites.Distinct species assemblages were identified in the meadow vegetation that was strongly aggregated by geographic location suggesting the presence of distinct phytogeographic zones of the meadow alpine flora.Elevation and geographic location were the dominant environmental gradients underlying the variations in species composition.Jaccard’s coefficient of similarity averaged only 10%among sites indicating there was little similarity in the alpine flora across the region.The alpine vegetation is highly heterogeneous across the complex landscape of the Hengduan Mountains of NWY.Conservation strategies need to take into account the large geographic differences in the flora to maximize protection of biodiversity.Ruth Sherman Renee Mullen Li Haomin Fang Zhendong Wang Yi 2008Journal of Plant Ecology2008,1,2:20
7遗传搭车与方差分析在水稻定向选择群体的抗旱性位点分析中的初步应用显示文摘水稻抗旱性属于复杂性状,近年的遗传研究日益关注胁迫下的产量相关位点。选用来自同一抗旱性较好的供体亲本(BG300),具有两种遗传背景(IR64和特青)和两种选择条件(低地和旱地)的4个水稻高代回交(BC2)抗旱性定向选择导入系群体。通过SSR分子标记,检测群体中供体等位基因的导入频率,分析了选择条件和遗传背景对供体等位基因导入频率的影响。在此基础上,利用基于遗传搭车原理的卡方检验对等位基因导入频率的偏离进行检测,同时结合表型-基因型方差分析(ANOVA)的方法,对上述群体进行了和抗旱性相关位点的QTL分析。共检测到显著位点24个,其中卡方检验发现20个,方差分析检测到8个。有4个被这两种方法都检测到,很可能是与胁迫下的产量直接相关的位点。同时也对非胁迫下的产量位点进行了检测,发现上述抗旱性位点与非胁迫下产量的关系不大。还就选择条件和遗传背景的效应等对目标性状QTL定位的影响进行了讨论。研究结果将为进一步的水稻抗旱性分子育种研究提供有用的信息和材料。郑天清 徐建龙 傅彬英 高用明 Satish VERUKA Renee LAFITTE 翟虎渠 万建民 朱苓华 黎志康 2007作物学报2007,33,5:19
8Steps to consider in the approach and management of critically ill patient with spontaneous intracerebral hemorrhage显示文摘Spontaneous intracerebral hemorrhage is a type of stroke associated with poor outcomes. Mortality is elevated, especially in the acute phase. From a pathophysiological point of view the bleeding must traverse different stages dominated by the possibility of re-bleeding, edema, intracranial hypertension, inflammation and neurotoxicity due to blood degradation products, mainly hemoglobin and thrombin. Neurological deterioration and death are common in early hours, so it is a true neurologicalneurosurgical emergency. Time is brain so that action should be taken fast and accurately. The most significant prognostic factors are level of consciousness, location, volume and ventricular extension of the bleeding. Nihilism and early withdrawal of active therapy undoubtedly influence the final result. Although there are no proven therapeutic measures, treatment should be individualized and guided preferably by pathophysiology. The multidisciplinary teamwork is essential. Results of recently completed studies have birth to promising new strategies. For correct management it's important to establish an orderly and systematic strategy based on clinical stabilization, evaluation and establishment of prognosis, avoiding secondary insults and adoption of specific individualized therapies, including hemostatic therapy and intensive control of elevated blood pressure. 11Osteopontin increases hepatocellular carcinoma cell growth in a CD44 dependant manner显示文摘AIM:To investigate the role of osteopontin(OPN) and its splice variants in the proliferation of hepatocellular carcinoma(HCC).METHODS:The expression of OPN variants in HCC cell lines as well as HCC tissue samples and nontumour tissue was studied using polymerase chain reaction.OPN variant cDNAs were cloned into a mammalian expression vector allowing both transient expression and the production of stable OPN expressing cell lines.OPN expression was studied in these cells using Western blotting,immunofluoresnce and enzyme linked immunosorbent assay.A CD44 blocking antibody and siRNA targeting of CD44 were used to examine the role of this receptor in the OPN stimulated cell growth observed in culture.Huh-7 cells stably expressing either OPN-A,-B or-C were injected subcutaneously into the flanks of nude mice to observe in vivo tumour growth.Expression of OPN mRNA and protein in these tumours was examined using reverse transcriptionpolymerase chain reaction and immunohistochemistry.RESULTS:OPN is expressed in HCC in 3 forms,the full length OPN-A and 2 splice variants OPN-B and-C.OPN variant expression was noted in HCC tissue as well as cognate surrounding cirrhotic liver tissue.Expression of these OPN variants in the HCC derived cell line Huh-7 resulted in secretion of OPN into the culture medium.Transfer of OPN conditioned media to na ve Huh-7 and HepG2 cells resulted in significant cell growth suggesting that all OPN variants can modulate cell proliferation in a paracrine manner.Furthermore the OPN mediated increase in cellular proliferation was dependent on CD44 as only CD44 positive cell lines responded to OPN conditioned media while siRNA knockdown of CD44 blocked the proliferative effect.OPN expression also increased the proliferation of Huh-7 cells in a subcutaneous nude mouse tumour model,with Huh-7 cells expressing OPN-A showing the greatest proliferative effect.CONCLUSION:This study demonstrates that OPN plays a significant role in the proliferation of HCC through interaction with the cell surface receptor CD44.Modulation of this interaction could represent a novel strategy for the control of HCC.Renee J Phillips Karla J Helbig Kylie H Van der Hoek Devanshi Seth Michael R Beard 2012World Journal of Gastroenterology2012,18,26:12

The determination of P in impregnated cotton and yarn (90 % cotton 10 % polyester) has been investigated as an example with the instrument of AAS vario 6, Analytik Jena AG. The RSD for 5 duplicates was between 2.3%~7.4%, comparable with liquid sampling graphite furnace AAS analysis. 13Cyclooxygenase-2 polymorphisms and the risk of esophageal adeno-or squamous cell carcinoma显示文摘AIM:To determine whether-1195 A→G and/or-765 G→C polymorphisms in Cyclooxygenase-2(COX-2 ) may have a risk modifying effect on the development of esophageal carcinoma in a Dutch Caucasian population.METHODS:Two study groups were recruited, 252 patients with esophageal carcinoma and 240 healthy controls, matched for race, age, gender and recruiting area.DNA was isolated from whole blood and used for genotyping.PCR products were digested with restriction enzymes and products were analyzed by agarose gel electrophoresis.Odds ratios(OR) and 95% confldence intervals(CI) were estimated.RESULTS:The distribution of the-1195 A→G polymorphism was signif icantly different in esophageal cancer patients compared to controls.The-1195 GG genotype resulted in a higher risk of developing esophageal adenocarcinoma(OR = 3.85, 95% CI:1.45-10.3) compared with the-1195 AA genotype as a reference.The-765 G→C genotype distribution was not different between the two groups.The GG/ GG haplotype was present more often in esophageal adenocarcinoma patients than in controls(OR = 3.45, 95% CI:1.24-9.58;with AG/AG as a reference).The same trends were observed in patients with squamous cell carcinomas, however, the results did not reach statistical signif icance.CONCLUSION:Presence of the COX-2-1195 GG genotype and of the GG/GG haplotype may result in a higher risk of developing esophageal carcinoma.Jón O Kristinsson Paul van Westerveld Rene HM te Morsche Hennie MJ Roelofs T Wobbes Ben JM Witteman Adriaan CITL Tan Martijn GH van Oijen Jan BMJ Jansen Wilbert HM Peters 2009World Journal of Gastroenterology2009,15,28:11
14The role of the androgen receptor in prostate development and benign prostatic hyperplasia:A review显示文摘Benign prostatic hyperplasia(BPH)is a benign enlargement of the prostate in which incidence increases linearly with age,beginning at about 50 years old.BPH is a significant source of morbidity in aging men by causing lower urinary tract symptoms and acute urinary retention.Unfortunately,the etiology of BPH incidence and progression is not clear.This review highlights the role of the androgen receptor(AR)in prostate development and the evidence for its involvement in BPH.The AR is essential for normal prostate development,and individuals with defective AR signaling,such as after castration,do not experience prostate enlargement with age.Furthermore,decreasing dihydrotestosterone availability through therapeutic targeting with 5a-reductase inhibitors diminishes AR activity and results in reduced prostate size and symptoms in some BPH patients.While there is some evidence that AR expression is elevated in certain cellular compartments,how exactly AR is involved in BPH progression has yet to be elucidated.It is possible that AR signaling within stromal cells alters intercellular signaling and a“reawakening”of the embryonic mesenchyme,loss of epithelial AR leads to changes in paracrine signaling interactions,and/or chronic inflammation aids in stromal or epithelial proliferation evident in BPH.Unfortunately,a subset of patients fails to respond to current medical approaches,forcing surgical treatment even though age or associated co-morbidities make surgery less attractive.Fundamentally,new therapeutic approaches to treat BPH are not currently forthcoming,so a more complete molecular understanding of BPH etiology is necessary to identify new treatment options.Renee E.Vickman Omar E.Franco Daniel C.Moline Donald J.Vander Griend Praveen Thumbikat Simon W.Hayward 2020Asian Journal of Urology2020,7,3:11
15Magnetic resonance imaging in assessment of stress urinary incontinence in women: Parameters differentiating urethral hypermobility and intrinsic sphincter deficiency显示文摘AIM: To define the magnetic resonance imaging(MRI) parameters differentiating urethral hypermobility(UH) and intrinsic sphincter deficiency(ISD) in women with stress urinary incontinence(SUI).METHODS: The static and dynamic MR images of 21 patients with SUI were correlated to urodynamic(UD) findings and compared to those of 10 continent controls. For the assessment of the urethra and integrity of the urethral support structures, we applied the highresolution endocavitary MRI, such as intraurethral MRI, endovaginal or endorectal MRI. For the functional imaging of the urethral support, we performed dynamic MRI with the pelvic phased array coil. We assessed the following MRI parameters in both the patient and thevolunteer groups:(1) urethral angle;(2) bladder neck descent;(3) status of the periurethral ligaments,(4) vaginal shape;(5) urethral sphincter integrity, length and muscle thickness at mid urethra;(6) bladder neck funneling;(7) status of the puborectalis muscle;(8) pubo-vaginal distance. UDs parameters were assessed in the patient study group as follows:(1) urethral mobility angle on Q-tip test;(2) Valsalva leak point pressure(VLPP) measured at 250 cc bladder volume; and(3) maximum urethral closure pressure(MUCP). The UH type of SUI was defined with the Q-tip test angle over 30 degrees, and VLPP pressure over 60 cm H2 O. The ISD incontinence was defined with MUCP pressure below 20 cm H2 O, and VLPP pressure less or equal to 60 cm H2 O. We considered the associations between the MRI and clinical data and UDs using a variety of statistical tools to include linear regression, multivariate logistic regression and receiver operating characteristic(ROC) analysis. All statistical analyses were performed using STATA version 9.0(Stata Corp LP, College Station, TX).RESULTS: In the incontinent group, 52% have history of vaginal delivery trauma as compared to none in control group(P < 0.001). There was no difference between the continent volunteers and incontinent patients in body habitus as assessed by the body mass index. Pubovaginal distance and periurethral ligament disruption are significantly associated with incontinence; periurethral ligament symmetricity reduces the odds of incontinence by 87%. Bladder neck funneling and length of the suprapubic urethral sphincter are significantly associated with the type of incontinence on UDs; funneling reduced the odds of pure UH by almost 95%; increasing suprapubic urethral sphincter length at rest is highly associated with UH. Both MRI variables result in a predictive model for UDs diagnosis(area under the ROC = 0.944). 19Intensivist-based deep sedation using propofol for pediatric outpatient flexible bronchoscopy显示文摘AIM To evaluate the safety and efficacy of sedating pediatric patients for outpatient flexible bronchoscopy.METHODS A retrospective chart review was conducted for all children, age 17 years or under who underwent flexible bronchoscopy under deep sedation in an outpatient hospital-based setting. Two sedation regimens were used; propofol only or ketamine prior to propofol. Patients were divided into three age groups; infants(less than 12 mo), toddlers(1-3 years) and children(4-17 years). Demographics, indication for bronchoscopy, sedative dosing, sedation and recovery time and adverse events were reviewed.RESULTS Of the total 458 bronchoscopies performed, propofol only regimen was used in 337(74%) while propofol and ketamine was used in 121(26%). About 99% of the procedures were successfully completed. Children in the propofol + ketamine group tend to be youngerand have lower weight compared to the propofol only group. Adverse events including transient hypoxemia and hypotension occurred in 8% and 24% respectively. Median procedure time was 10 min while the median discharge time was 35 min. There were no differences in the indication of the procedure, propofol dose, procedure or recovery time in either sedative regimen. When compared to other age groups, infants had a higher incidence of hypoxemia.CONCLUSION Children can be effectively sedated for outpatient flexible bronchoscopy with high rate of success.
