Elegant biochemical work identified the DNA fragmentation factor (DFF) as a major apoptotic endonuclease for DNA fragmentation in vitro. Genetic studies in mice support the importance of DFF in DNA fragmentation and possibly in apoptosis in vivo. Recent work also suggests the existence of additional endonucleases for DNA degradation. | 4 | Fast multipole boundary element analysis of 2D viscoelastic composites with imperfect interfaces显示文摘A fast multipole boundary element method(FMBEM)is developed for the analysis of 2D linear viscoelastic composites with imperfect viscoelastic interfaces.The transformed fast multipole formulations are established using the time domain method. | 5 | Mitochondrial DNA A1555G mutation screening using a testing kit method and its significance in preventing aminoglycoside-related hearing loss显示文摘To report a new screening method for mitochondrial DNA 1555A→G mutation and the results of genotype analysis in 19 maternal inherited deafness pedigrees. Method Five hundred and forty-six non-syndromic neuro-sensory hearing loss patients were tested for 1555A→G mutation using a new compact testing kit, which allows clear distinction between wild type and 1555 A→G mutated mtDNAs. Results Nineteen subjects among the 546 patients (3.48%) were found to carry mtDNA A1555G mutation. The results were confirmed by sequencing in an ABI 3100 Avant sequencer. Conclusions Maternal inherited deafness families are a frequently seen in outpatient group. | 6 | Effects of Soil Fertility and Atmospheric CO_2 Enrichment on Leaf, Stem and Root Dark Respiration of Populus tremuloides显示文摘An open-top chamber experiment was conducted at the University of Michigan Biological Station near Pellston, Michigan, USA, to study the effects of soil fertility and CO2 on leaf, stem and root dark respiration (Rd) of Populus tremuloides. Overall, area-based day-time leaf Rd (Rda) was significantly greater at elevated than at ambient CO2 in high-fertility soil, but not in low-fertility soil. Mass-based leaf Rd (Rdm) was overall greater for high- than for low-fertility soil grown trees at elected, but not at ambient CO2. Nighttime leaf Rd. and Rdm were unthected by soil fertility or CO2, nor was stem Rda, which ranged from 1.0 to 1.4 μmol m-2 s-1 in the spring and 3.5 to 4.5 μmol m-2 s-1 in the summer. Root Rda. was significantly higher in high- than in low-fertility soil, but was unaffected by CO2. Since biomass production of P. tremuloides will be significantly greater at elevated CO2 while specific Rd will either increase or remain unchanged, we predict that carbon loss to the atmosphere through respiration from this ecologically important species would increase at higher CO2. The model is capable of simulating the thermal, deformation and microstructure evolution in the piercing, elongating and reducing/sizing and the austenite decomposition in the mill annealing and cooling operations. Finite difference schemes are employed to model cooling, reducing/sizing and stretch reducing, and finite-element schemes are employed to simulate piercing and elongating. The model predicts the thermal history, deformation, rolling load, torque, recrystallization and grain growth in hot tube rolling, austenite decomposition in cooling or annealing, and the final structure-properties. In this paper mathematical models which are employed to describe the thermal, deformation and microstructure evolution along with the modeling results are presented. | 10 | Mitochondrial DNA Mutations Associated with Aminoglycoside Ototoxicity显示文摘The mitochondrial 12S rRNA has been shown to be the hot spot for mutations associated with both aminoglycoside-induced and non-syndromic hearing loss. Of all the mutations, the homoplasmic A1555G and C1494T mutations at a highly conserved decoding region in the 12S rRNA have been associated with aminoglycoside-induced and non-syndromic hearing loss in many families worldwide. The A1555G or C1494T mutation is expected to form novel 1494C-G1555 or 1494U-A1555 base-pair at the highly conserved A-site of 12S rRNA. These transitions make the secondary structure of this RNA more closely resemble the corresponding region of bacterial 16S rRNA. Thus, the new U-A or G-C pair in 12S rRNA created by the C1494T or A1555G transition facilitates the binding of aminoglycosides, thereby accounting for the fact that the exposure to aminoglycosides can induce or worsen hearing loss in individuals carrying these mutations. Furthermore, the growth defect and impairment of mitochondrial translation were observed in cell lines carrying the A1555G or C1494T mutation in the presence of high concentration of aminoglycosides. In addition, nuclear modifier genes and mitochondrial haplotypes modulate the phenotypic manifestation of the A1555G and C1494T mutations. These observations provide the direct genetic and biochemical evidences that the A1555G or C1494T mutation is a pathogenic mtDNA mutation associated with aminoglycoside-induced and nonsyndromic hearing loss. | 12 | Widely predicting specific protein functions based on protein-protein interaction data and gene expression profile显示文摘GESTs (gene expression similarity and taxonomy similarity), a gene functional prediction approach previously proposed by us, is based on gene expression similarity and concept similarity of functional classes defined in Gene Ontology (GO). In this paper, we extend this method to protein-protein interac-tion data by introducing several methods to filter the neighbors in protein interaction networks for a protein of unknown function(s). Unlike other conventional methods, the proposed approach automati-cally selects the most appropriate functional classes as specific as possible during the learning proc-ess, and calls on genes annotated to nearby classes to support the predictions to some small-sized specific classes in GO. Based on the yeast protein-protein interaction information from MIPS and a dataset of gene expression profiles, we assess the performances of our approach for predicting protein functions to “biology process” by three measures particularly designed for functional classes organ-ized in GO. Results show that our method is powerful for widely predicting gene functions with very specific functional terms. | 13 | Antisense Oligodeoxynucleotide Inhibits Expression of Re-com binantPorcine Follicle-Stim ulating Horm one Receptor显示文摘To assess the role of follicle stimulating hormone receptor(FSHR) gene expression in regulating expression of FSHR protein in the plasma membrane, the effects of a porcine FSHR cDNA antisense oligodeoxynucleotide (ODN) on FSHR mRNA levels and 125 I FSH binding were determined in Chinese hamster ovary cells expression recombinant porcine FSHR (pFSHR CHO cells). An 18 mer phosphorothioate endcapped antisense ODN that corresponded to the region surrounding the translation initiation codon of the porcine FSHR cDNA was synthesized. An 18 mer nonsense sequence of identical nucleotide composition, which had little homology to known DNA sequences, was synthesized for use as a control. pFSHR CHO cells were cultured in 24 well plates (10 5 cells/well) in the absence or presence of 1 20 μmol/L antisense or nonsense ODN for 24 h and then assayed for porcine FSHR mRNA, using quantitative reverse transcription and competitive polymerase chain reaction, and for 125 I FSH binding activity. Treatment with 10 μmol/L antisense ODN caused a paradoxical increase in porcine FSHR mRNA from 0.89±0.06 to 1.64±0.08 ng/mg total RNA ( P <0.05). Transfection with lipofectamine and 0.33 μmol/L antisense ODN caused an increase in porcine mRNA from 0.95±0.08 to 1.53±0.07 ng/mg total RAN. This was probably due to upregulation of mRNA synthesis resulting from inhibition of porcine FSHR protein translation. The nonsense ODN had no effect on porcine FSHR mRNA. Antisense, but not nonsense, ODN (10 μmol/L) inhibited membrane binding of 125 I FSH by 13.6± 0.8 % ( P <0.05) in 24 h. Treatment of cells with antisense ODN (10 μmol/L) for 48 h resulted in a 76±1.5 % ( P <0.05) inhibition of 125 I FSH binding. In contrast, transfection with lipofectamine and 0.33 μmol/L antisense ODN at 0 h caused a 76.1±1.3 % ( P <0.05) reduction in binding within 24 h. Binding had returned to 52.3±2.3 % ( P < 0.05) of normal by 48 h. These results indicate that an antisense ODN corresponding to the region of the translation start site of the porcine FSHR cDNA is an effective specific inhibitor of porcine FSHR synthesis and that inhibition of receptor synthesis causes a decrease in functional membrane bound FSHR. | ZHU Changhong 1, Mark D. DEFORMTM was originally designed for metal forming applications. While its FEM engine employs the updated Lagrangian method, DEFORMTM includes an automated remeshing procedure so as to continue the simulation when the mesh is severely distorted. Due to its unique remeshing capability, the use of the code has been extended to other manufacturing processes, such as welding, machining, heat treatment and glass forming. The heat treatment enhancement includes the phase transformation kinetics. To demonstrate the methodology, applications of the program to various manufacturing processes such as ingot breakdown, extrusion, forging and heat treatment, are presented in this paper. | WT.
