| 1 | Clinical application of specific antibody against glypican-3 for hepatocellular carcinoma diagnosis显示文摘Glypican-3(GPC3) is a promising tumor marker for hepatocellular carcinoma(HCC) diagnosis with high sensitivity and specificity.The aim of this study was to establish an immunohistochemical detection method for GPC3 using the 7D11 monoclonal antibody(7D11 mAb) and evaluate its application for HCC diagnosis.The feasibility of the 7D11 mAb was evaluated by immunohistochemistry performed on adjacent normal liver and intrahepatic cholangiocarcinoma(ICC) samples,Furthermore,the serum GPC3 levels were evaluated in 40 HCC patients,7 ICC patients and 50 healthy donors.The results showed that GPC3 was expressed in 85% of HCC tissues(34/40),but was undetectable in ICC tissues and adjacent normal tissues.GPC3 was significantly increased in the serum of HCC patients(17/40,42.5%) but was undetectable in the serum of ICC patients(0/7,0%) and healthy donors(0/50,0%).This prospective study evaluated the clinical usefulness of 7D11 mAb for GPC3 detection in HCC patients.In conclusion,the use of 7D11 mAb might be good for GPC3 large-scale applications for clinical diagnosis of HCC. | YU JuanPing MA Qiang ZHANG Bin MA RuiJuan XU XiGuang LI MingSong XU WeiWen LI Ming | 2013 | Science China(Life Sciences)2013,56,3: | 18 |
| 2 | SERK Receptor-like Kinases Control Division Patterns of Vascular Precursors and Ground Tissue Stem Cells during Embryo Development in Arabidopsis显示文摘During embryo development, the vascular precursors and ground tissue stem cells divide to renew them-selves and produce the vascular tissue, endodermal cells, and cortical cells. However, the molecular mech-anisms regulating division of these stem cells have remained largely elusive. In this study, we show that loss of function of SOMATIC EMBRYOGENESIS RECEPTOR-UKE KINASE (SERK) genes results in aberrant em-bryo development. Fewer cortical, endodermal, and vascular cells are generated in the embryos of serk1 serk2bak1 triple mutants. WUSCHEL-RELATED HOMBOBOX5 (WOXS) is ectopically expressed in vascular cells of serkl serk2 bak1 embryos. The first transverse division of vascular precursors in mid-globular em-bryos and second asymmetric division of ground tissue stem cells in early-heart embryos are abnormally altered to a longitudinal division. The embryo defects can be partially rescued by constitutively activated mitogen-activated protein kinase (MAPK) kinase kinase YODA (YDA) and MAPK kinase MKK5. | 3 | Astronomical time scale of the Turonian constrained by multiple paleoclimate proxies显示文摘One of the clocks that record the Earth history is(quasi-) periodic astronomical cycles.These cycles influence the climate that can be ultimately stored in sedimentary rocks.By cracking these(quasi-) periodic sedimentation signals,high resolution astronomical time scale(ATS) can be obtained.Paleoclimate proxies are widely used to extract astronomical cycles.However different proxies may respond differently to astronomical signals and nonastronomical noises including tectonics,diagenesis,and measurement error among others.Astronomical time scale constructed based on a single proxy where its signal-to-noise ratio is low may have uncertainty that is difficult to evaluate but can be revealed by utilizing other proxies.Here,we test eight astronomical age models using two astrochro no logical methods from four paleoclimate proxies(i.e.,color reflection L~* and b~*,natural gamma radiation,and bulk density) from the Turonian to the Coniacian of the Cretaceous Period at the Demerara Rise in the equatorial Atlantic.The two astrochronological methods are time calibration using long eccentricity bandpass filtering(E1 bandpass) and tracking the long eccentricity from evolutive harmonic analysis(tracking EHA).The statistical mean and standard deviation of four age models from the four proxies are calculated to construct one integrated age model with age uncertainty in each method.Results demonstrate that extracting astronomical signals from multiple paleoclimate proxies is a valid method to estimate age model uncertainties.Anchored at the Cenomanian/Turonian boundary with an age of 93.9 ± 0.15 Ma from biostratigraphy,the ages for CC11/CC12(calcareous nannofossil zones),Turonian/Coniacian(CC12/CC13),CC13/CC14,and Coniacian/Santonian boundaries are 91.25±0.20 Ma,89.87±0.20 Ma,86.36±0.33 Ma,and 86.03±0.32 Ma in E1 bandpass method,compared with 91.17±0.36 Ma,89.74±0.38 Ma,86.13±1.31 Ma,and 85.80±1.33 Ma respectively in tracking EHA method.These results are consistent with previous studies within error and provide a reliable estimation of uncertainties of the ages. | Chao Ma Mingsong Li | 2020 | Geoscience Frontiers2020,11,4: | 5 |
| 4 | Biting force and tongue muscle strength as useful indicators for eating and swallowing capability assessment among elderly patients显示文摘Weakened capability of eating and swallowing is common among elderly population,affecting negatively on their health and well-being.This work measured occlusal force and tongue muscle strength of elderly patients and then assessed whether correlations existed between oral physiological properties and the eating and swallowing capability.A total of 26 elderly patients with potential swallow disorder were recruited in this work.Participants’denture status and daily diet texture were determined by a pre-questionnaire analysis.Their swallowing capability was assessed and rated by water drinking test.Maximum tongue pressure,incisor occlusal force and hand gripping force were measured for each subject.It was observed that participating subjects have on average a significantly lower biting force and maximum tongue pressure than that of normal adults.The swallowing grades assessed by water drinking test showed significant correlations with both the biting force(r=−0.498,P<0.05)and the tongue muscle strength(r=−0.544,P<0.05).The biting force of dysphagia participants correlates significantly with dental conditions(P<0.05).The observed correlation between tongue pressure and diet texture indicates that tongue muscle strength could be used as a reliable indicator of the eating and swallowing capability for elderly people. | Xinmiao Wang Gangying Zheng Mingsong Su Yanqiu Chen Hua Xie Weijia Han Qing Yang Jianqin Sun Jianshe Chen | 2019 | Food Science and Human Wellness2019,8,2: | 4 |

| 6 | Climate change response to astronomical forcing during the Oligocene-Miocene transition in the equatorial Atlantic(ODP Site 926)显示文摘The Oligocene-Miocene transition period was characterized by a decrease in global CO2 levels, expansion of polar ice sheet, fall in global sea-level, etc. However, the reasons for, and mechanisms of, this global, extreme-cold climate change event(Mi-1) still remain controversial. Our samples from the core of the Ocean Drilling Program(ODP) Leg 154, Site 926, located in the equatorial Atlantic, mainly consist of light-gray, nannofossil chalk with foraminifers interbedded with greenish-gray, clayey, nannofossil chalk sediments. Color variation from light-gray layers(up to 80% carbonate content) to dark layers(60% carbonate content) was observed to occur cyclically at the meter scale. Therefore, we chose color reflectance lightness(L*) data as the paleoclimate proxy on which to perform cyclostratigraphic analysis because it could reflect carbonate content changes. Based on the recognition of the 405 kyr long eccentricity and 40 kyr obliquity cycles of the L* series, we tuned the series to establish an absolute astronomical time scale using the published age of the Oligocene-Miocene boundary(OMB) as the anchor for an absolute age control point. The power spectra of the tuned L* series showed that the long eccentricity signals became significantly weak, while the obliquity signals became strong, from the Late Oligocene to the Early Miocene. The 405 kyr long eccentricity minimum coincided with the 1.2 Myr obliquity node at the OMB, and similar convergences might be closely related to other extreme-cold events in Earth’s history. | 8 | The staphylococcal nuclease prevents biofilm formation in Staphylococcus aureus and other biofilm-forming bacteria显示文摘The staphylococcal nuclease, encoded by the nuc1 gene, is an important virulence factor of Staphylococcus aureus. However, the physiological role of the nuclease has not been fully characterized. The current study observed that biofilm development could be prevented in staphylococcal nuclease-producing strains of S. aureus; however, when the nuc1 gene was knocked out, the ability to form a biofilm significantly increased. Scanning electron and confocal scanning laser microscopy were used to evaluate the role of the nuc1 gene in biofilm formation. Moreover, the nuc1 gene product, staphylococcal nuclease, and recombinant NUC1 protein were found to have a visible effect on other biofilm-forming bacteria, such as Pseudomonas aeruginosa, Actinobacillus pleuropneumoniae, and Haemophilus parasuis. The current study showed a direct relationship between staphylococcal nuclease production and the prevention of biofilm development. The findings from this study underscore the important role of staphylococcal nuclease activity to prevent biofilm formation in S. aureus. | 9 | The CRL3^(BTBD9) E3 ubiquitin ligase complex targets TNFAIP1 for degradation to suppress cancer cell migration显示文摘Tumor necrosis factor alpha-induced protein 1(TNFAIP1)modulates a plethora of important biological processes,including tumorigenesis and cancer cell migration.However,the regulatory mechanism of TNFAIP1 degradation remains largely elusive.In the present study,with a label-free quantitative proteomic approach,TNFAIP1 was identified as a novel ubiquitin target of the Cullin-RING E3 ubiquitin ligase(CRL)complex.More importantly,Cul3-ROC1(CRL3),a subfamily of CRLs,was identified to specifically interact with TNFAIP1 and promote its polyubiquitination and degradation.Mechanistically,BTBD9,a specific adaptor component of CRL3 complex,was further defined to bind and promote the ubiquitination and degradation of TNFAIP1 in cells.As such,downregulation of BTBD9 promoted lung cancer cell migration by upregulating the expression of TNFAIP1,whereas TNFAIP1 deletion abrogated this effect.Finally,bioinformatics and clinical sample analyses revealed that BTBD9 was downregulated while TNFAIP1 was overexpressed in human lung cancer,which was associated with poor overall survival of patients.Taken together,these findings reveal a previously unrecognized mechanism by which the CRL3^(BTBD9) ubiquitin ligase controls TNFAIP1 degradation to regulate cancer cell migration. | Lihui Li Wenjuan Zhang Yue Liu Xiaojun Liu Lili Cai Jihui Kang Yunjing Zhang Wenlian Chen Changsheng Dong Yanmei Zhang Mingsong Wang Wenyi Wei Lijun Jia | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 2 |

| 18 | Recrystallization nucleation under close-setδphase in a nickel-based superalloy during annealing显示文摘Annealing is one of the most efficient heat treatment to refine the grain microstructure in metal alloys.Prior to the annealing,an aging treatment can be implemented to precipitate the second phases for more nucleation sites in deformed mixed grains during subsequent recrystallization.However,the underlying mechanism behind the second phases and recrystallization has not yet been fully understood.Taking a typical deformed nickel(Ni)-based(GH4169)superalloy as an exapmle,this research aims to clairfy the connections between the recrystallization and delta(δ)phase(Ni_(3)Nb) during annealing.All deformed samples were also treated by aging and then annealing.The results show that the microstructure and component ofδphase dominated the recrystallization nucleation in the Ni-based alloy.Moreover,there was the presence of theδphase interval limiting nucleation(PILN)andδphase protecting nucleation(PPN),when a high content of close-set phase was precipitated from the matrix.The PILN determined the primary condition for the recrystallization nucleation that happended at the interval ofδphase larger than the critical nucleation size.The peak nucleation occurred in the PPN,where the recrystallization nucleus were free of the penetration by the adjacent grains,before the nucleation incubation time was reached.Thus,a new feasible JNtheory was proposed to determine the maximum domain of recrystallization nucleation under close-set precipitated phases.All the possible nucleation sites were assumed under PPN in the JNtheory.As such,the original microstructure can be expected to design for the annealing via the aging process.A quantitative evaluation of recrystallization nucleation can also achieved in the Ni-based alloys during aging. | Guanqiang Wang Mingsong Chen Yongcheng Lin Hongbin Li Yuqiang Jiang Yanyong Ma Chengxu Peng Jinliang Cai Quan Chen | 2022 | Journal of Materials Science & Technology2022,,20: | 1 |
