| 5 | Mitochondrial protection by low doses of insulin-like growth factor-Ⅰin experimental cirrhosis显示文摘AIM: To characterize the mitochondrial dysfunction in experimental cirrhosis and to study whether insulin-like growth factor-Ⅰ(IGF-Ⅰ) therapy (4 wk) is able to induce beneficial effects on damaged mitochondria leading to cellular protection. METHODS: Wistar rats were divided into three groups: Control group, untreated cirrhotic rats and cirrhotic rats treated with IGF-Ⅰtreatment (2 μg/100 g bw/d). Mitochondrial function was analyzed by flow cytometry in isolated hepatic mitochondria, caspase 3 activation was assessed by Western blot and apoptosis by TUNEL in the three experimental groups. RESULTS: Untreated cirrhotic rats showed a mitochon- drial dysfunction characterized by a significant reduction of mitochondrial membrane potential (in status 4 and 3); an increase of intramitochondrial reactive oxigen species (ROS) generation and a significant reduction of ATPase activity. IGF-Ⅰtherapy normalized mitochondrial func-tion by increasing the membrane potential and ATPase activity and reducing the intramitochondrial free radical production. Activity of the electron transport complexes Ⅰand Ⅲ was increased in both cirrhotic groups. In addition, untreated cirrhotic rats showed an increase of caspase 3 activation and apoptosis. | 6 | Effect of Y_2O_3 stabilized ZrO_2 coating with tri-model structure on bi-layered thermally grown oxide evolution in nano thermal barrier coating systems at elevated temperatures显示文摘Bi-layered thermally grown oxide(TGO) layer plays a major role in the spallation of Y2 O3 stabilized ZrO2(YSZ) layer form the bond coat in the thermal barrier coating(TBC) systems during oxidation. On the other hand, bi-layered TGO formation and growth in the TBC systems with nanostructured YSZ have not been deeply investigated during cyclic oxidation. Hence, Inconel 738/NiCrAlY/normal YSZ and Inconel 738/NiCrAlY/nano YSZ systems were pre-oxidized at 1000 °C and then subjected to cyclic oxidation at 1150 °C. According to microstructural observations, nanostructured YSZ layer over the bond coat should have less micro-cracks and pinholes, due to the compactness of the nanostructure and the presence of nano zones that resulted in lower O infiltration into the nanothermal barrier coating system, formation of thinner and nearly continuous mono-layered thermally grown oxide on the bond coat during pre-oxidation, lower spinels formation at the Al2 O3 /YSZ interface and finally, reduction of bi-layered thermally grown oxide thickness during cyclic oxidation. | 9 | Investigation of three steps of hot corrosion process in Y_2O_3 stabilized ZrO_2 coatings including nano zones显示文摘Phase transformation of tetragonal ZrO2 to monoclinic phase and also increment of bond coat oxidation kinetic(TGO thickening) can substantially restrict the life time of thermal barrier coating systems(TBCs). So, nanostructured and conventional Y2O3 stabilized ZrO2 coatings were evaluated in fused V2O5-Na2SO4 salts during thermal exposure in air. Microstructural characterization showed lower hot corrosion products(monoclinic zirconia, YVO4 crystals) formation and reduction of TGO thickness in thermal barrier coating system consisting of nanostructured Y2O3 stabilized ZrO2(YSZ) top coat. It was found that inhomogeneities, pores and micro-cracks played a principal role in the molten salts infiltration into the YSZ coating during three steps of hot corrosion process. | 20 | Genetic barriers in transplantation medicine显示文摘The successful of transplantation is determined by the shared human leukocyte antigens(HLAs) and ABO blood group antigens between donor and recipient. In recent years, killer cell receptor [i.e., killer cell immunoglobulinlike receptor(KIR)] and major histocompatibility complex(MHC) class I chain-related gene molecule(i.e., MICA) were also reported as important determinants of transplant compatibility. At present, several different genotyping techniques(e.g., sequence specific primer and sequence based typing) can be used to characterize blood group, HLA, MICA and KIR and loci. These molecular techniques have several advantages because they do not depend on the availability of anti-sera, cellular expression and have greater specificity and accuracy compared with the antibody-antigen based typing. Nonetheless, these molecular techniques have limited capability to capture increasing number of markers which have been demonstrated to determine donor and recipient compatibility. It is now possible to genotype multiple markers and to the extent of a complete sequencing of the human genome using next generation sequencer(NGS). This high throughput genotyping platform has been tested for HLA, and it is expected that NGS will be used to simultaneously genotype a large number of clinically relevant transplantation genes in near future. This is not far from reality due to the bioinformatics support given by the immunogenetics community and the rigorous improvement in NGS methodology.
