1LncRNA PVT1 promotes ovarian cancer progression by silencing miR-214显示文摘Objective: Emerging evidence indicates that long non-coding RNAs(lnc RNAs) are critical in carcinogenesis and progression of ovarian cancer. This study aimed to explore the functions and molecular mechanisms of plasmacytoma variant translocation I(PVT1) in ovarian cancer Methods: PVT1 and miR-214 were detected by qRT-PCR assays in ovarian cancer tissues and cells. The cell proliferation,migration, and invasion abilities were detected by cell functional experiments, respectively. Western blot assay was performed to detect epithelial-mesenchymal transition(EMT) markers. MiR-214 expression regulated by PVT1 was studied by RNA immunoprecipitation(RIP) and chromatin immunoprecipitation(Ch IP) assays.Results: The expression of PVT1 was up-regulated in ovarian cancer tissues and cell lines. Elevated PVT1 expression was associated with advanced stage and indicated poor prognosis for ovarian cancer patients. The knockdown of PVT1 impaired SKOV3 cell proliferation, migration, and invasion in vitro. The promotion of ovarian cancer progression by PVT1 involved in regulation of the epithelial-mesenchymal transition process and PVT1 interaction with EZH2 represses miR-214 expression in ovarian cancer cells.Conclusions: PVT1 plays an important role in ovarian cancer tumorigenesis, which might be as a novel diagnostic marker and therapeutic target for ovarian cancer.Ying Chen Hui Du Lewen Bao Wenxin Liu 2018Cancer Biology & Medicine2018,15,3:18
2Metal-dependent anaerobic methane oxidation in marine sediment:insights from marine settings and other systems显示文摘Anaerobic oxidation of methane(AOM) plays a crucial role in controlling global methane emission. This is a microbial process that relies on the reduction of external electron acceptors such as sulfate, nitrate/nitrite, and transient metal ions. In marine settings, the dominant electron acceptor for AOM is sulfate, while other known electron acceptors are transient metal ions such as iron and manganese oxides. Despite the AOM process coupled with sulfate reduction being relatively well characterized,researches on metal-dependent AOM process are few, and no microorganism has to date been identified as being responsible for this reaction in natural marine environments. In this review, geochemical evidences of metal-dependent AOM from sediment cores in various marine environments are summarized. Studies have showed that iron and manganese are reduced in accordance with methane oxidation in seeps or diffusive profiles below the methanogenesis zone. The potential biochemical basis and mechanisms for metal-dependent AOM processes are here presented and discussed. 3Application of GA-SVM in classification of surrounding rock based on model reliability examination显示文摘In order to improve the discrimination precision of support vector machine(SVM) in classification of surrounding rock, a Genetic Algorithm(GA) was used to optimize SVM parameters in the solution space.The idea of examination of model reliability was introduced to check the reliability of the SVM parameters,obtained by genetic algorithms.In the process of model reliability,a trend examination method is presented,which checks the reliability of the model via the influence trend of impact factors on the object of evaluation and their evaluation level.Trend examination methods are universal,showing new ideas in model reliability examination and can be used in any problems of examination of reliability of models,based on previous experience.We established a GA-SVM based reliability model of a classification the surrounding rock and applied it to a practical engineering situation.The result shows that the improved SVM has a high capability for generalization and prediction accuracy in classification of surrounding rock.QIU Daohong LI Shucai ZHANG Lewen XUE Yiguo 2010Mining Science and Technology2010,20,3:6
