1Association between MDM2-SNP309 and hepatocellular carcinoma in Taiwan Residents population显示文摘AIM:To investigate the risk association and compare the onset age of hepatocellular carcinoma(HCC) patients in Taiwan with different genotypes of MDM2- SNP309. METHODS:We analyzed MDM2-SNP309 genotypes from 58 patients with HCC and 138 cancer-free healthy controls consecutively.Genotyping of MDM2-SNP309 was conducted by restriction fragment length polymor- phism assay. RESULTS:The proportion of homozygous MDM2- SNP309 genotype(G/G)in cases and cancer-free healthy controls was similar(17.2%vs 16.7%).Multi-variate analysis showed that the risk of G/G genotypeof MDM2-SNP309 vs wild-type T/T genotype in patients with HCC was not significant(OR=1.265,95% CI=0.074-21.77)after adjustment for sex,hepatitis B or C virus infection,age,and cardiovascular disease/ diabetes.Nevertheless,there was a trend that GG genotype of MDM2-SNP309 might increase the risk in HCC patients infected with hepatitis virus(OR=2.568, 95%CI=0.054-121.69).Besides,the homozygous MDM2-SNP309 genotype did not exhibit a significantly earlier age of onset for HCC. 3Sitagliptin protects rat kidneys from acute ischemia- reperfusion injury via upregulation of GLP-1 and GLP-1 receptors显示文摘Aim: Sitagliptin, an oral glucose-lowering agent, has been found to produce cardiovascular protection possibly via anti-inflammatory and anti-atherosclerotic activities of glucagon-like peptide-1 receptor (GLP-1). The aim of this study was to investigate whether sitagliptin protected the kidney function from acute ischemia-reperfusion (IR) injury in rats. Methods: Adult male SD rats were categorized into 4 groups: sham control, IR injury, IR+sitagliptin (300 mg/kg) and IR+sitagliptin (600 mg/kg). Acute renal IR injury of both kidneys was induced by clamping the renal pedicles for I h. The drug was orally administered at 1, 24 and 48 h after acute IR. Blood samples and 24-h urine were collected before and at 72 h after acute IR. 4Apoptotic Effect of Cisplatin and Cordycepin on OC3 Human Oral Cancer Cells显示文摘Objective:To evaluate apoptotic effects of cisplatin and cordycepin as single agent or in combination with cytotoxicity in oral cancer cells.Methods:The influences of cisplatin(2.5μg/mL)and/or cordycepin treatment(10 or 100μmol/L)to human OC3 oral cancer cell line were investigated by morphological observation for cell death appearance,methylthiazoletetrazolium(MTT)assay for cell viability,flow cytometry assay for cell apoptosis,and Western blotting for apoptotic protein expressions.Results:Data demonstrated that co-administration of cisplatin(2.5μg/mL)and cordycepin(10 or 100μmol/L)resulted in the enhancement of OC3 cell apoptosis compared to cisplatin or cordycepin alone treatment(24 h),respectively(P<0.05).In flow cytometry assay,percentage of cells arrested at subG1 phase with co-treatment of cordycepin and cisplatin(30%)was significantly higher than cisplatin(5%)or cordycepin(12%)alone group(P<0.05),confirming a synergistically apoptotic effect of cordycepin and cisplatin.In cellular mechanism study,co-treatment of cordycepin and cisplatin induced more stress-activated protein kinase/Jun terminal kinase(JNK),the expressions of caspase-7,and the cleavage of poly ADP-ribose polymerase(PARP)as compared to cisplatin or cordycepin alone treatment(P<0.05).Conclusion:Cisplatin and cordycepin possess synergistically apoptotic effect through the activation of JNK/caspase-7/PARP pathway in human OC3 oral cancer cell line.CHEN Ying-hui HAO Lyh-Jyh HUNG Chih-peng CHEN Jung-wei LEU Sew-fen HUANG Bu-miin 2014Chinese Journal of Integrative Medicine2014,20,8:3

63D bioprinting of stem cells and polymer/bioactive glass composite scaffolds for bone tissue engineering显示文摘A major limitation of using synthetic scaffolds in tissue engineering applications is insufficient angiogenesis in scaffold interior.Bioactive borate glasses have been shown to promote angiogenesis.There is a need to investigate the biofabrication of polymer composites by incorporating borate glass to increase the angiogenic capacity of the fabricated scaffolds.In this study,we investigated the bioprinting of human adipose stem cells(ASCs)with a polycaprolactone(PCL)/bioactive borate glass composite.Borate glass at the concentration of 10 to 50 weight%,was added to a mixture of PCL and organic solvent to make an extrudable paste.ASCs suspended in Matrigel were ejected as droplets using a second syringe.Scaffolds measuring 10×10×1 mm^3 in overall dimensions with pore sizes ranging from 100–300μm were fabricated.Degradation of the scaffolds in cell culture medium showed a controlled release of bioactive glass for up to two weeks.The viability of ASCs printed on the scaffold was investigated during the same time period.This 3D bioprinting method shows a high potential to create a bioactive,highly angiogenic three-dimensional environment required for complex and dynamic interactions that govern the cell’s behavior in vivo.Caroline Murphy Krishna Kolan Wenbin Li Julie Semon Delbert Day Ming Leu 2017International Journal of Bioprinting2017,3,1:3
