The cells were treated byselective extraction methOd and prepared for whole mount electron microscopy observation.The samples were examined respectively withscanning and transmission electron microscope.RESULTS The nuclear matrix filaments andintermediate filaments in MGC80-3 cells wererelatively few and scattered, not welldistributed and arranged irregularly. The nuclear lamina was ununiformly thick and compact,connected to the nuclear matrix filaments and intermediate filaments relaxedly. However, thetwo kinds of filaments were abundant and welldistributed, different in slender and thick form and interweaved into a regular network in the cells induced by 10-6 mol/ L RA. The nuclear matrix filaments and intermediate filaments were connected closely by the thin and compact fiber-Iike lamina, and interlaced into a regularnetwork throughout the whole cell region.CONCLUSION The NM-IF system in MGc80-3cells had undergone a restorational changesimilar to those of normaI cells after RAinducement. | 7 | Intrahepatic biliary cystic neoplasms:Surgical results of 9 patients and literature review显示文摘AIM: To investigate the eligible management of the cystic neplasms of the liver. METHODS: The charts of 9 patients who underwent surgery for intrahepatic biliary cystic liver neoplasms between 2003 and 2008 were reviewed retrospectively. Informed consent was obtained from the patients and approval was obtained from the designated review board of the institution. RESULTS: All patients were female with a median (range) age of 49 (27-60 years). The most frequent symptom was abdominal pain in 6 of the patients. Four patients had undergone previous laparotomy (with other diagnoses) which resulted in incomplete surgery or recurrences. Liver resection (n = 6) or enucleation (n = 3) was performed. The final diagnosis was intrahepaticbiliary cystadenoma in 8 patients and cystadenocarcinoma in 1 patient. All symptoms resolved after surgery. There has been no recurrence during a median (range) 31 (7-72) mo of follow up. CONCLUSION: In spite of the improvement in imaging modalities and increasing recognition of biliary cystadenoma and cystadenocarcinoma, accurate preoperative diagnosis may be difficult. | 9 | Effects of resveratrol in experimental and clinical non-alcoholic fatty liver disease显示文摘The prevalence of obesity and related conditions like non-alcoholic fatty liver disease(NAFLD) is increasing worldwide and therapeutic options are limited.Alternative treatment options are therefore intensively sought after.An interesting candidate is the natural polyphenol resveratrol(RSV) that activates adenosinmonophosphate-activated protein kinase(AMPK) and silent information regulation-2 homolog 1(SIRT1).In addition,RSV has known anti-oxidant and anti-inflammatory effects.Here,we review the current evidence for RSVmediated effects on NAFLD and address the different aspects of NAFLD and non-alcoholic steatohepatitis(NASH) pathogenesis with respect to free fatty acid(FFA) flux from adipose tissue,hepatic de novo lipogenesis,inadequate FFA β-oxidation and additional intra- and extrahepatic inflammatory and oxidant hits.We review the in vivo evidence from animal studies and clinical trials.The abundance of animal studies reports a decrease in hepatic triglyceride accumulation,liver weight and a general improvement in histological fatty liver changes,along with a reduction in circulating insulin,glucose and lipid levels.Some studies document AMPK or SIRT1 activation,and modulation of relevant markers of hepatic lipogenesis,inflammation and oxidation status.However,AMPK/SIRT1-independent actions are also likely.Clinical trials are scarce and have primarily been performed with a focus on overweight/obese participants without a focus on NAFLD/NASH and histological liver changes.Future clinical studies with appropriate design are needed to clarify the true impact of RSV treatment in NAFLD/NASH patients. | Sara Heebll Karen Louise Thomsen Steen B Pedersen Hendrik Vilstrup Jacob George Henning Grnbk | 2014 | World Journal of Hepatology2014，6，4： | 11 |
| 10 | Associations of sense of coherence with psychological distress and quality of life in inflammatory bowel disease显示文摘AIM:To investigate the relationship between sense of coherence,psychological distress and health related quality of life in inflammatory bowel disease(IBD).METHODS:This cross-sectional study enrolled a consecutive sample of 147 IBD(aged 45.1 ± 14.1 years; 57.1% female) patients recruited from a tertiary gastroenterology service.Sixty-four participants met diagnostic criteria for Crohn's disease,while eightythree patients had ulcerative colitis.Socio-demographic data(education,age,race,gender,gross monthly income and marital status),disease-related variables(illness activity,relapse rate in past 2 years,history of surgery and time since diagnosis),sense of coherence(Antonovsky's SOC scale),psychological distress symptoms(Hospital Anxiety and Depression Scale) and health-related quality of life(HRQo L; WHOQOLBref) were assessed.Hierarchical multiple regression analyses were performed to identify factors that are independently associated with psychological distress and HRQo L in patients with IBD and to provide indications for possible moderating or mediating effects.In addition,formal moderation and mediation analyses(Sobel tests) were performed to confirm potential moderators/mediators of the relationship between SOC,psychological distress symptoms and HRQoL.RESULTS:Lower SOC scores(std beta=-0.504; P < 0.001),female gender(std beta = 0.176; P = 0.021) and White race(std beta = 0.164; P = 0.033) were independently associated with higher levels of depressive symptoms,while lower levels of SOC(std beta =-0.438; P < 0.001) and higher relapse rate(std beta = 0.161; P = 0.033) were independently associated with more severe anxiety symptoms.A significant interaction between time since diagnosis and SOC was found with regard to the severity of depressive or anxiety symptoms,as the interaction term(time since diagnosis X SOC) had beta coefficients of-0.191(P = 0.009) and-0.172(P = 0.026),respectively.Lower levels of anxiety symptoms(std beta =-0.369; P < 0.001),higher levels of SOC(std beta = 0.231; P = 0.016) and non-White race(std beta =-0.229; P = 0.006),i.e.,mixed-race,which represented the reference category,were independently associated with higher levels of overall HRQoL.Anxiety symptoms were the most potent independent correlate of most aspects of HRQoL.In addition,anxiety mediated the association between SOC and satisfaction with health,as well as its relationship with physical,mental,and social relations HRQo L.Depressive symptoms also mediated the association between SOC and mental HRQoL.CONCLUSION:Our data indicated that SOC is an important construct,as it influences psychological distress and has significant albeit indirect effects on several HRQoL domains in IBD. | Thiago H Freitas Elias Andreoulakis Gilberto S Alves Hesley LL Miranda Lúcia LBC Braga Thomas Hyphantis André F Carvalho | 2015 | World Journal of Gastroenterology2015，21，21： | 10 |
| 11 | Functional roles and clinical values of insulin-like growth factor-binding protein-5 in different types of cancers显示文摘Insulin-like growth factor-bindin g proteins (IGFBPs) are critical regulators of the mitogenic activity of insulin-like growth factors (IGFs). IGFBP5, one of these IGFBPs, has special structural features, including a nuclear transport domain, heparin-binding motif, and IGF/extracellular matrix/acid-labile subunit-binding sites. Furthermore, IGFBP5 has several functional effects on carcinogenesis and even normal cell processes, such as cell growth, death, motility, and tissue remodeling. These biological effects are sometimes related with IGF (IGF-dependent effects) and sometimes not (IGF-independent effects). The functional role of IGFBP5 is most likely determined in a cell-type and tissue-type specific manner but also depends on cell context, especially in terms of the diversity of interacting proteins and the potential for nuclear localization. Clinical findings show that IGFBP5 has the potential to be a useful clinical biomarker for predicting response to therapy and clinical outcome of cancer patients. | 13 | Efficacies of β-L-D4A against Hepatitis B virus in 2.2.15 cells显示文摘AIM:To investigate the antiviral effect of beta-L-enantiomer of 2',3'-didehydro-2',3'-dideoxyadenosine (β-L-D4A) on 2.2.15 cells transfected with the hepatitis B virus (HBV) genome. METHODS:Lamivudine (3TC) as a positive control. Then, HBV DNA in treated 2.2.15 cells and the Hepatitis B surface antigen (HBsAg) in the culture supernatants were detected to determine the inhibitory effect of β-L-D4A. At the same time, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) was used to detect the survival ratio of 2.2.15 cells. RESULTS:β-L-D4A has a dose-dependent inhibitory effect on HBV DNA replication;this effect was apparent when the concentration was above 1 mol/L. When β-L-D4A was at the highest concentration, 100 mol/L, the HBsAg inhibition ratio was above 50%. The Therapeutic index (TI) of β-L-D4A was above 2.1. | 15 | PD-L1 and PD-L2 expression correlated genes in non-small-cell lung cancer显示文摘Background:Programmed cell death ligand-1(PD-L1)and ligand-2(PD-L2)interaction with programmed cell death protein-1(PD-1)represent an immune-inhibiting checkpoint mediating immune evasion and is,accordingly,an important target for blockade-based immunotherapy in cancer.In non-small-cell lung cancer(NSCLC),improved understanding of PD-1 checkpoint blockade-responsive biology and identification of biomarkers for prediction of a clinical response to immunotherapy is warranted.Thus,in the present study,we systematically described PD-L1 and PD-L2 expression correlated genes in NSCLC.Methods:We performed comparative retrospective analyses to identify PD-L1 and PD-L2 mRNA expression corre-lated genes in NSCLC.For this,we examined available datasets from the cancer cell line encyclopedia(CCLE)project lung non-small-cell(Lung_NSC)and the cancer genome atlas(TCGA)projects lung adenocarcinoma(LUAD)and squamous cell carcinoma(LUSC).Results:Analysis of the CCLE dataset Lung_NSC identified expression correlation between PD-L1 and PD-L2.Moreo-ver,we identified expression correlation between 489 genes and PD-L1,191 genes and PD-L2,and 111 genes for both.PD-L1 and PD-L2 also expression correlated in TCGA datasets LUAD and LUSC.In LUAD,we identified expression corre-lation between 257 genes and PD-L1,914 genes and PD-L2,and 211 genes for both.In LUSC,we identified expression correlation between 26 genes and PD-L1,326 genes and PD-L2,and 13 genes for both.Only a few genes expression correlated with PD-L1 and PD-L2 across the CCLE and TCGA datasets.Expression of Interferon signaling-involved genes converged in particular with the expression correlated genes for PD-L1 in Lung_NSC,for PD-L2 in LUSC,and for both PD-L1 and PD-L2 in LUAD.In LUSC,PD-L1,and to a lesser extent PD-L2,expression correlated with chromosome 9p24 localized genes,indicating a chromosome 9p24 topologically associated domain as an important driver of in particu-lar LUSC PD-L1 expression.Expression correlation analyses of the PD-L1 and PD-L2 receptors programmed cell death protein-1(PD-1),Cluster of differentiation 80(CD80),and Repulsive guidance molecule B(RGMB)showed that PD-1 and CD80 expression correlated with both PD-L1 and PD-L2 in LUAD.CD80 expression correlated with PD-L2 in LUSC.Conclusions:We present gene signatures associated with PD-L1 and PD-L2 mRNA expression in NSCLC which could possess importance in relation to understand PD-1 checkpoint blockade-responsive biology and development of gene signature based biomarkers for predicting clinical responses to immunotherapy. | Trine Vilsbøll Larsen Dianna Hussmann Anders Lade Nielsen | 2019 | Cancer Communications2019，39，1： | 7 |

Methods: In adult male BALB/c mice,testosterone implants, estradiol benzoate, vasectomy, unilateral cryptorchidism, unilateral varicocele were adminis tered/performed. The mice were followed up for different periods of time and were then sacrificed with testes incised for examination. The control mice received the vehicle or sham-operation. Results: IL-10 was present in Leydig cells of nearly every testis and IL-10 + macrophages in 39% of testes. IL-6 was found in the testes of intact adult mice, mice treated with testosterone for 70 days, cryptorchid testes and sham-operated testes.
