117β-雌二醇对雄性金鱼卵黄原蛋白的诱导作用显示文摘采用腹腔注射17β-雌二醇的方法诱导雄性金鱼卵黄原蛋白产生,注射浓度为0.05mg·g-1BW,诱导2周后取尾静脉血,离心分离血浆,进行血浆常规聚丙烯酰胺凝胶电泳,通过对卵黄原蛋白特性基团磷、脂和糖蛋白的染色,确定了卵黄原蛋白在电泳图谱上的位置,开发了一种简便、高效的定性卵黄原蛋白的聚丙烯酰胺凝胶电泳法。电泳结果表明,在0.05mg·g-1BW的注射浓度下,2周后17β-雌二醇诱导了雄性金鱼卵黄原蛋白产生,并通过ELISA检测卵黄原蛋白的平均含量为690.2ng·mL-1,与对照组雄性金鱼平均含量为10.7ng·mL-1的差异极显著(P<0.01),比雌性对照组检出量285.5ng·mL-1高1倍多;17β-雌二醇诱导组雄鱼血浆钙离子和血总蛋白含量明显增加。邴欣 汝少国 Isoda Hiroko 姜明 王蔚 王诗红 2004水产学报2004,28,3:24
2Growth and Eco-Physiological Performance of Cotton Under Water Stress Conditions显示文摘A cotton cultivar Xinluzao 8 was grown under four levels of water stress treatments (normal irrigation, slight, mild and severe water stress) from the initial reproductive growth stage in Shihezi, Xinjiang, China, in 2002, to evaluate the growth and eco-physiological performances. Under water stress conditions, the transpiration ability decreased while the leaf temperature increased. Although the relative leaf water content decreased as water stress increased, the differences among the treatments were small, indicating that cotton has high ability in maintaining water in leaf. The stomatal density increased as water stress increased, while the maximum stomatal aperture reduced only in the severest stressed plants. The time of the maximum stomatal aperture was delayed in the mild and severe stressed plants. When severe stress occurred, the stomata were kept open until the transpiration decreased to nearly zero, suggesting that the stomata might not be the main factor in adjusting transpiration in cotton. Cotton plant has high adaptation ability to water stress conditions because of decrease in both stomatal conductance and hydraulic conductance from soil-to-leaf pathway. The actual quantum yield of photosystem Ⅱ(PS Ⅱ) decreased under water stress conditions, while the maximum quantum yield of PS Ⅱ did not vary among treatments, suggesting that PS Ⅱ would not be damaged by water stress. 3采用Caco-2和PC12细胞检测有机磷农药的细胞毒性显示文摘建立了采用哺乳动物细胞系Caco-2和PC12细胞检测有机磷农药细胞毒性的新方法。1 0μmol L甲基对硫磷、敌敌畏、氧乐果、辛硫磷处理经过单层培养的Caco-2细胞,随着农药处理时间的延长,Caco-2细胞跨上皮细胞电阻抗值逐渐降低,处理60min时,辛硫磷和甲基对硫磷对Caco-2细胞具有明显的透过性,约是正常组的1 5倍,4种农药对Caco-2细胞透过性由大到小的顺序为:甲基对硫磷,辛硫磷,敌敌畏,氧乐果;0 01μmol L的甲基对硫磷和辛硫磷使PC12细胞乙酰胆碱酯酶活性迅速降低,随着处理浓度的增加酶活性降低不明显;而小于1 0μmol L的氧乐果和敌敌畏呈现渐进降低的趋势,当浓度大于1 0μmol L时酶活性降低比较显著。汝少国 ISODA Hiroko 邴欣 2003环境科学研究2003,16,2:6
4Assessment of Photochemical Reflectance Index as a Tool for Evaluation of Chlorophyll Fluorescence Parameters in Cotton and Peanut Cultivars Under Water Stress Condition显示文摘The relationships between photochemical reflectance index (PRI) and the chlorophyll fluorescence parameters were examined to assess suitability of PRI as a remote-sensing tool for the chlorophyll fluorescence parameters. A greenhouse experiment was conducted using cotton and peanut crops under water stress condition. Five cotton and six peanut cultivars were grown using Andosole soil in pots maintained at two water levels; the control and water stress treatment (WS) of 100 and 50% of the daily transpiration, respectively. Higher non-photochemical quenching (NPQ) was exhibited by peanut than that of cotton by the water stress. On the other hand, the decreases of the actual quantum yield of photosystem II (?F/F′m) and PRI by the water stress in cotton were larger than those in peanut. There were positively significant correlation coefficients between PRI and ?F/F′m in cotton at noon and in the afternoon including the control and WS. The correlations of PRI with NPQ were negatively significant at noon and in the afternoon for cotton, and in the afternoon for peanut. No clear relationship was found among these parameters in the morning probably due to the diurnal increase in global solar radiation. 5Anticancer effect of linalool via cancer-specific hydroxyl radical generation in human colon cancer显示文摘AIM To investigate the anticancer mechanisms of the monoterpenoid alcohol linalool in human colon cancer cells.METHODS The cytotoxic effect of linalool on the human colon cancer cell lines and a human fibroblast cell line was examined using the WST-8 assay. The apoptosisinducing effect of linalool was measured using the terminal deoxynucleotidyl transferase d UTP nickend labeling assay and flow cytometry with Annexin V. Oxidative stress was investigated by staining for diphenyl-1-pyrenylphosphine, which is a cellular lipid peroxidation marker, and electron spin resonance spectroscopy. Sixteen SCID mice xenografted with human cancer cells were randomized into 3 groups for in vivo analysis: control and low-dose and high-dose linalool groups. The control group was administered tap water orally every 3 d. The linalool treatment groups were administered 100 or 200 μg/kg linalool solution orally for the same period. All mice were sacrificed under anesthesia 21 d after tumor inoculation, and tumors and organs were collected for immunohistochemistry using an anti-4-hydroxynonenal antibody. Tumor weights were measured and compared between groups. RESULTS Linalool induced apoptosis of cancer cells in vitro, following the cancer-specific induction of oxidative stress, which was measured based on spontaneous hydroxyl radical production and delayed lipid peroxidation. Mice in the high-dose linalool group exhibited a 55% reduction in mean xenograft tumor weight compared with mice in the control group(P < 0.05). 6Evaluation of safety for hepatectomy in a novel mouse model with nonalcoholic-steatohepatitis显示文摘AIM To investigate whether the liver resection volume in a newly developed nonalcoholic steatohepatitis(NASH) model influences surgical outcome.METHODS For establishment of a NASH model, mice were fed a high-fat diet for 4 wk, administered CCl_4 for the last 2 wk, and administered T0901317 for the last 5 d. We divided these mice into two groups: A 30% partial hepatectomy(PH) of NASH liver group and a 70% PH of NASH liver group. In addition, a 70% PH of normal liver group served as the control. Each group was evaluated for survival rate, regeneration, apoptosis, necrosis and DNA expression after PH.RESULTS In the 70% PH of NASH group, the survival rate was significantly decreased compared with that in the control and 30% PH of NASH groups(P < 0.01). 10 of 32 mice in the NASH 70% PH group died within 48 h after PH. Serum aspartate aminotransferase(AST) levels and total bilirubin(T-Bil) in the NASH 70% PH group were significantly higher than the levels in the other two groups(AST: P < 0.05, T-Bil: P < 0.01). In both PH of NASH groups, signaling proteins involved in regeneration were expressed at lower levels than those in the control group(P < 0.01). The 70% PH of NASH group also exhibited a lower number of Ki-67-positive cells and higher rates of apoptosis and necrosis than the NASH 30% PH group(P < 0.01). In addition, DNA microarray assays showed differences in gene expression associated with cell cycle arrest and apoptosis.CONCLUSION The function of the residual liver is impaired in fatty liver compared to normal liver. 7Enhanced hepatic differentiation in the subpopulation of human amniotic stem cells under 3D multicellular microenvironment显示文摘BACKGROUND To solve the problem of liver transplantation donor insufficiency,an alternative cell transplantation therapy was investigated.We focused on amniotic epithelial cells(AECs)as a cell source because,unlike induced pluripotent stem cells,they are cost-effective and non-tumorigenic.The utilization of AECs in regenerative medicine,however,is in its infancy.A general profile for AECs has not been comprehensively analyzed.Moreover,no hepatic differentiation protocol for AECs has yet been established.To this end,we independently compiled human AEC libraries,purified amniotic stem cells(ASCs),and co-cultured them with mesenchymal stem cells(MSCs)and human umbilical vein endothelial cell(HUVECs)in a 3D system which induces functional hepatic organoids.AIM To characterize AECs and generate functional hepatic organoids from ASCs and other somatic stem cells METHODS AECs,MSCs,and HUVECs were isolated from the placentae and umbilical cords of cesarean section patients.Amnion and primary AEC stemness characteristics and heterogeneity were analyzed by immunocytochemistry,Alkaline phosphatase(AP)staining,and flow cytometry.An adherent AEC subpopulation was selected and evaluated for ASC purification quality by a colony formation assay.AEC transcriptomes were compared with those for other hepatocytes cell sources by bioinformatics.The 2D and 3D culture were compared by relative gene expression using several differentiation protocols.ASCs,MSCs,and HUVECs were combined in a 3D co-culture system to generate hepatic organoids whose structure was compared with a 3D AEC sphere and whose function was elucidated by immunofluorescence imaging,periodic acid Schiff,and an indocyanine green(ICG)test.RESULTS AECs have certain stemness markers such as EPCAM,SSEA4,and E-cadherin.One AEC subpopulation was also either positive for AP staining or expressed the TRA-1-60 and TRA-1-81 stemness markers.Moreover,it could form colonies and its frequency was enhanced ten-fold in the adherent subpopulation after selective primary passage.Bioinformatics analysis of ribose nucleic acid sequencing revealed that the total AEC gene expression was distant from those of pluripotent stem cells and hepatocytes but some gene expression overlapped among these cells.TJP1,associated with epidermal growth factor receptor,and MET,associated with hepatocyte growth factor receptor,were upregulated and may be important for hepatic differentiation.In conventional flat culture,the cells turned unviable and did not readily differentiate into hepatocytes.In 3D culture,however,hepatic gene expression of the AEC sphere was elevated even under a two-step differentiation protocol.Furthermore,the organoids derived from the MSC and HUVEC co-culture showed 3D structure with polarity,hepatic-like glycogen storage,and ICG absorption/elimination.CONCLUSION Human amniotic epithelial cells are heterogeneous and certain subpopulations have high stemness.Under a 3D co-culture system,functional hepatic organoids were generated in a multicellular microenvironment.Kinji Furuya Yun-Wen Zheng Daisuke Sako Kenichi Iwasaki Dong-Xu Zheng Jian-Yun Ge Li-Ping Liu Tomoaki Furuta Kazunori Akimoto Hiroya Yagi Hiromi Hamada Hiroko Isoda Tatsuya Oda Nobuhiro Ohkohchi 2019World Journal of Stem Cells2019,11,9:2
