| 2 | 质子重离子肿瘤放疗的临床注册试验初步报告显示文摘目的探讨IONTRIS粒子放疗设备临床应用的安全性和有效性。方法2014年6月至2014年8月间共有35例肿瘤患者入组,其中男性31例,女性4例:年龄36~80岁,中位年龄69岁。头颈肿瘤10例(均为手术后复发),胸部肿瘤4例,原发性肝癌1例,腹膜后肉瘤术后复发1例.前列腺癌19例。碳离子照射剂量为2.74—10GyE/次,质子照射剂量为2—5.5GyE/次,5次/周。放疗前对每个放射野进行水箱剂量验证。结果碳离子照射22例,质子照射13例。放疗后1年,35例患者全部生存。在16例头颈部、胸部和腹盆腔肿瘤患者中,完全缓解2例,部分缓解1例,疾病稳定12例,疾病进展1例,1年无进展生存率为93.8%(15/16)。19例前列腺癌患者的1年无生化复发生存率为100%。35例患者对粒子放疗的耐受性良好,放疗期间的美国东部肿瘤协作组评分和体重无明显改变。25例(71.4%)患者发生了与粒子放疗相关的急性不良事件(AE)33例次,均为1级.在放疗1年时急性AE全部消失。有6例(17.1%)患者在放疗后1年发生后期AE6例次,全部为1级。结论采用IONTRIS设备治疗肿瘤患者安全、有效。 | 陆嘉德 叶明 郭小毛 傅深 Moyers F. | 6 | Investigation of process parameters of electrohydrodynamic jetting for 3D printed PCL fibrous scaffolds with complex geometries显示文摘Tissue engineering is a promising technology in the field of regenerative medicine with its potential to create tissues de novo.Though there has been a good progress in this field so far,there still exists the challenge of providing a 3D micro-architecture to the artificial tissue construct,to mimic the native cell or tissue environment.Both 3D printing and 3D bioprinting are looked upon as an excellent solution due to their capabilities of mimicking the native tissue architecture layer-by-layer with high precision and appreciable resolution.Electrohydrodynamic jetting(E-jetting)is one type of 3D printing,in which,a high electric voltage is applied between the extruding nozzle and the substrate in order to print highly controlled fibres.In this study,an E-jetting system was developed in-house for the purpose of 3D printing of fibrous scaffolds.The effect of various E-jetting parameters,namely the supply voltage,solution concentration,nozzle-to-substrate distance,stage(printing)speed and solution dispensing feed rate on the diameter of printed fibres were studied at the first stage.Optimized parameters were then used to print Polycaprolactone(PCL)scaffolds of highly complex geometries,i.e.,semi-lunar and spiral geometries,with the aim of demonstrating the flexibility and capability of the system to fabricate complex geometry scaffolds and biomimic the complex 3D micro-architecture of native tissue environment.The spiral geometry is expected to result in better cell migration during cell culture and tissue maturation. | Hui Wang Sanjairaj Vijayavenkataraman Yang Wu Zhen Shu Jie Sun Jerry Fuh Ying Hsi | 2016 | International Journal of Bioprinting2016,2,1: | 4 |
| 7 | Genome-wide DNA hypermethylation and homocysteine increase a risk for myopia显示文摘AIM: To test for the association between genome-wide methylation and myopia in human and mice. METHODS: Long interspersed nucleotide element 1(LINE-1) methylation levels were used to surrogate genome-wide methylation level. We first tested for the association between high myopia(<-6 D) and LINE-1 methylation in leukocytes in 220 cases and 220 control subjects. Secondly, we validated the results of LINE-1 methylation in eyes from the form deprivation myopia(FDM) mice. Furthermore,we calculated the correlation of LINE-1 methylation levels between leukocyte DNA and ocular DNA in the mice. We also tested whether dopamine can alter LINE-1 methylation levels. RESULTS: The LINE-1 methylation level was significantly higher in the myopic human subjects than controls. The upper and middle tertiles of the methylation levels increased an approximately 2-fold(P≤0.002) risk for myopia than the lower tertile. Similarly, FDM mice had high LINE-1 methylation levels in the leukocyte, retina and sclera, and furthermore the methylation levels detected from these three tissues were significantly correlated. Immunohistochemical staining revealed higher levels of homocysteine and methionine in the rodent myopic eyes than normal eyes. Dopamine treatment to the cells reduced both LINE-1 methylation and DNA methyltransferase levels. CONCLUSION: LINE-1 hypermethylation may be associated with high myopia in human and mice. | 8 | Biliopleural fistula: A rare complication of percutaneous transhepatic gallbladder drainage显示文摘A 79-year-old previously healthy man presented with acute acalculous cholecystitis with obstruction of the biliary tract. He was successfully treated with antibiotics and percutaneous transhepatic gallbladder drainage, but returned to the hospital two days after discharge with a rare complication of this technique, biliopleural fistula. A thoracostomy tube was inserted to drain the pleural effusion, and the patient’s previous antibiotics reinstated. | 9 | 离线模式下PET/CT显像验证质子放疗精准度的模型实验显示文摘目的通过对聚甲基丙烯酸甲酯(PMMA)模体行离线模式下PET/CT显像,探讨PET-边界验证方法(PET-RV)用于定量分析质子放疗精准性的可行性。方法在PMMA模体上分别给予2 Gy和4 Gy的照射剂量,对每个剂量组根据临床靶区(CTV)的不同,以2.5 cm为步进分别设计7个照射深度组(5.0、7.5、10.0、12.5、15.0、17.5、20.0 cm),共14个照射计划(14组照射野)。以能量在48~221 MeV之间的质子束照射,照射结束后10 min左右进行PET/CT显像。对PET/CT数据分别进行图像配准、平滑和约化等预处理,然后利用Raystation PET-RV软件对图像进行感兴趣区(ROI)的确定和采样线的抽提,通过生成的预测感生放射性活度与实际采集感生放射性活度采样线计算ΔR50,即2条曲线末端在最大预测感生放射性活度最大值下降一半(V50)处对应的空间位置差异。结果每个照射野的ROI均为5.0 cm×5.0 cm×2.5 cm的立方体,采样线在其内间隔3 mm均匀分布,共得到289组采样线。14组数据经分析后得到的ΔR50(深度差异)均值都处于-1~1 mm之间。结论离线模式下PET/CT显像示PET-RV算法对PMMA模体质子放疗空间位置差异的验证精度在1 mm之内,可以满足后期对临床病例分析的精度要求。 | 张建岗 张俊昱 卢艳 盛尹祥子 任炜 施磊俊 孙筠 章英剑 Hsi Wenchien 周荣 蒋国梁 陆嘉德 程竞仪 | 2020 | 中华核医学与分子影像杂志2020,40,10: | 2 |

| 18 | Recent developments in neuropsychological endophenotypes for schizophrenia: Development of the MATRICS battery, liability syndromes and the near future显示文摘Cognitive deficits are now recognized widely as core features of schizophrenia, and as major contributors to the clinical outcome of the disorder. They are also studied widely as 'endophenotypes', reflecting a growing consensus that schizophrenia is a broader, more multidimensional illness than the diagnostic criteria required for its formal diagnosis. This evolving view of cognition underlies its utilization in recent initiatives for intervention and assessment in schizophrenia. Two of these initiatives are reviewed in this paper. The first focuses on the development and validation of the MATRICS Cognitive Consensus Battery, a standardized battery of neuropsychological tests developed to assess the effectiveness of cognitive enhancing treatments in schizophrenia. A part of this effort includes the identification of performance-based, 'co-primary' measures of functional capacity that are related to cognition, and that are likely to show improvement at least partly as a function of improved cognition. The second initiative involves efforts to utilize neuropsychological deficits in the identification, validation and remediation of a liability syndrome for schizophrenia ('schizotaxia'). The discussion of this effort focuses on the development of a syndrome that is both measurable and meaningful clinically, and that may provide useful intervention targets. The utilization of cognition in both of these initiatives underscores its functional importance in the clinical outcome of schizophrenia.
