| 1 | Characterization of the receptor-binding domain(RBD)of 2019 novel coronavirus:implication for development of RBD protein as a viral attachment inhibitor and vaccine显示文摘The outbreak of Coronavirus Disease 2019(COVID-19)has posed a serious threat to global public health,calling for the development of safe and effective prophylactics and therapeutics against infection of its causative agent,severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),also known as 2019 novel coronavirus(2019-nCoV).The CoV spike(S)protein plays the most important roles in viral attachment,fusion and entry,and serves as a target for development of antibodies,entry inhibitors and vaccines.Here,we identified the receptor-binding domain(RBD)in SARS-CoV-2 S protein and found that the RBD protein bound strongly to human and bat angiotensin-converting enzyme 2(ACE2)receptors.SARS-CoV-2 RBD exhibited significantly higher binding affinity to ACE2 receptor than SARS-CoV RBD and could block the binding and,hence,attachment of SARS-CoV-2 RBD and SARS-CoV RBD to ACE2-expressing cells,thus inhibiting their infection to host cells.SARS-CoV RBD-specific antibodies could crossreact with SARS-CoV-2 RBD protein,and SARS-CoV RBD-induced antisera could cross-neutralize SARS-CoV-2,suggesting the potential to develop SARS-CoV RBD-based vaccines for prevention of SARS-CoV-2 and SARS-CoV infection. | Wanbo Tai Lei He Xiujuan Zhang Jing Pu Denis Voronin Shibo Jiang Yusen Zhou Lanying Du | 2020 | Cellular & Molecular Immunology2020,17,6: | 35 |
| 2 | Agricultural non-point nitrogen pollution control function of dierent vegetation types in riparian wetlands: A case study in the Yellow River wetland in China显示文摘Riparian wetland is the major transition zone of matter, energy and information transfer between aquatic and terrestrial ecosystems and has important functions of water purification and non-point pollution control. Using the field experiment method and an isotope tracing technique, the agricultural non-point nitrogen pollution control function of dierent vegetation types in riparian wetland was studied in the Kouma Section of the Yellow River. The results showed that the retention of agricultural non-point nitrogen pollution by riparian wetland soil occurs mainly in top 0-10 cm layer. The amount of nitrogen retained by surface soils associated with three types of vegetation are 0.045 mg/g for Phragmites communis Trin Linn, 0.036 mg/g for Scirpus triqueter Linn, and 0.032 mg/g for Typha angustifolia Linn, which account for 59.21%, 56.25%, and 56.14% of the total nitrogen interception, respectively. Exogenous nitrogen in 0-10 cm soil layer changes more quickly than in other layers. One month after adding K15NO3 to the tested vegetation, nitrogen content was 77.78% for P. communis Trin, 68.75% for T. angustifolia, and 8.33% for S. triqueter in the surface soil. After three months, nitrogen content was 93.33% for P. communis Trin, 72.22% for S. triqueter, and 37.50% for T. Angustifolia. There are large dierences among vegetation communities respecting to purification of agricultural non-point nitrogen pollution. The nitrogen uptake amount decreases in the sequence: new shoots of P. communis Trin (9.731 mg/g) > old P. communis Trin (4.939 mg/g) > S. triqueter (0.620 mg/g) > T. angustifolia (0.186 mg/g). | 4 | TLR4-HMGB1-, MyD88- and TRIF-dependent signaling in mouse intestinal ischemia/reperfusion injury显示文摘AIM: To characterize high-mobility group protein 1-toll-like receptor 4(HMGB1-TLR4) and downstream signaling pathways in intestinal ischemia/reperfusion(I/R) injury.METHODS: Forty specific-pathogen-free male C57BL/6 mice were randomly divided into five groups(n = 8 per group): sham, control, anti-HMGB1, anti-myeloid differentiation gene 88(My D88), and anti-translocatingchain-associating membrane protein(TRIF) antibody groups. Vehicle with the control Ig G antibody, antiHMGB1, anti-My D88, or anti-TRIF antibodies(all 1 mg/kg, 0.025%) were injected via the caudal vein 30 min prior to ischemia. After anesthetization, the abdominal wall was opened and the superior mesenteric artery was exposed, followed by 60 min mesenteric ischemia and then 60 min reperfusion. For the sham group, the abdominal wall was opened for 120 min without I/R. Levels of serum nuclear factor(NF)-κB p65, interleukin(IL)-6, and tumor necrosis factor(TNF)-α were measured, along with myeloperoxidase activity in the lung and liver. Inaddition,morphologic changes that occurred in the lung and intestinal tissues were evaluated. Levels of m RNA transcripts encoding HMGB1 and NF-κB were measured by real-time quantitative PCR, and levels of HMGB1 and NF-κB protein were measured by Western blot. Results were analyzed using one-way analysis of variance.RESULTS: Blocking HMGB 1, MyD 8 8, and TRIF expression by injecting anti-HMGB1, anti-My D88, or anti-TRIF antibodies prior to ischemia reduced the levels of inflammatory cytokines in serum; NF-κB p65: 104.64 ± 11.89, 228.53 ± 24.85, 145.00 ± 33.63, 191.12 ± 13.22, and 183.73 ± 10.81(P < 0.05); IL-6: 50.02 ± 6.33, 104.91 ± 31.18, 62.28 ± 6.73, 85.90 ± 17.37, and 78.14 ± 7.38(P < 0.05); TNF-α, 43.79 ± 4.18, 70.81 ± 6.97, 52.76 ± 5.71, 63.19 ± 5.47, and 59.70 ± 4.63(P < 0.05) for the sham, control, anti-HMGB1, anti-My D88, and anti-TRIF groups, respectively(all in pg/m L).Antibodies also alleviated tissue injury in the lung and small intestine compared with the control group in the mouse intestinal I/R model. | 5 | Correlation between BRAF^(V600E) mutation and clinicopathological features in pediatric papillary thyroid carcinoma显示文摘In adults, the presence of the BRAF^(V600E) mutation in papillary thyroid cancer(PTC) has been demonstrated to be strongly associated with aggressive cancer-cell characteristics and poor patient prognosis. In contrast, the frequency of this mutation in pediatric PTC has undergone limited study, and the few available estimates range from 0 to 63%. Furthermore, the role of the BRAF^(V600E) mutation in pediatric PTC is controversial; thus, the present study aimed to investigate the prevalence and role of the BRAF^(V600E) mutation in48 pediatric patients with PTC, aged 3–13 years. Of these patients, 41 were diagnosed with classic PTC, five were found to have a follicular variant of PTC, and two to exhibit a diffuse sclerosing PTC variant. The BRAF^(V600E) mutation was identified to be present in 35.4% of the 48 analyzed patients, and in 41.5% of the patients diagnosed with classical PTC. Furthermore, the presence of the BRAF^(V600E) mutation was found to be associated with a patient age at diagnosis of less than ten years(P=0.011), the performance of a thyroidectomy(P=0.03), exhibited tumor multifocality(P=0.02) and/or extra-thyroidal invasion(P=0.003), and both a low MACIS(Metastases, Age, Completeness of resection, Invasion, Size)(P=0.036) and AMES(Age, Metastasis, Extent of tumor,Size)(P=0.001)score. | 10 | Steroid elimination within 24 hours after orthotopic liver transplantation:effectiveness and tolerability显示文摘BACKGROUND:Steroids have been the mainstay of immunosuppressive regimen in liver transplantation.However,the use of steroids is associated with various post-transplant complications.This study evaluated the efficacy and safety of reduced immunosuppressive regimen with steroids(steroid elimination within 24 hours post-transplant) in a cohort of Chinese liver transplant recipients.METHODS:Seventy-six patients in line with the selection criteria were enrolled in this prospective study.All patients received anti-IL-2 receptor antibody induction and tacrolimusbased maintenance therapy.The recipients were divided into two groups according to the duration of steroid use:40 transplant in a 3-month withdrawal group and the remaining 36 in a 24-hour elimination group.Recipient survival,postoperative infections,biopsy-proven acute rejection and steroidresistant acute rejection,non-healing wound,recurrence of hepatitis B virus(HBV) and hepatocellular carcinoma(HCC),de novo diabetes,hyperlipidemia and hypertension were assessed in the two groups.RESULTS:There was no significant difference in patient survival,incidence of acute rejection episodes and hyperlipidemia,and recurrence of HBV and HCC between the two groups.However,the incidence rates of post-transplant infection,non-healing wound,de novo diabetes and hypertension were significantly lower in the 24-hour elimination group than in the 3-month withdrawal group(all P values <0.05).CONCLUSION:Under anti-IL-2 receptor antibody induction and tacrolimus-based maintainance,steroid elimination within 24 hours post-transplant is associated with reduced steroidrelated complications without increasing the risk of rejection. | Lin-Wei Wu,Zhi-Yong Guo,Qiang Tai,Wei-Qiang Ju,Dong-Ping Wang,An-Bin Hu,Xiao-Feng Zhu and Xiao-Shun He Organ Transplantation Center, First Affiliated Hospital, Sun Yat-Sen University, Guangzhou 510080, China | 2012 | Hepatobiliary & Pancreatic Diseases International2012,11,2: | 4 |
| 11 | Liver transplantation using organs from deceased organ donors: a single organ transplant center experience显示文摘BACKGROUND: In 2011, a pilot program for deceased organ donation was initiated in China. We describe the first successful series of liver transplants in the pilot program.METHODS: From July 2011 to August 2012, our center performed 26 liver transplants from a pool of 29 deceased donors. All organ donation and allograft procurement were conducted according to the national protocol. The clinical data of donors and recipients were collected and summarized retrospectively.RESULTS: Among the 29 donors, 24 were China Category II donors(organ donation after cardiac death), and five were China Category III donors(organ donation after brain death followed by cardiac death). The recipients were mainly the patients with hepatocellular carcinoma. The one-year patient survival rate was 80.8% with a median follow-up of 422(2-696) days. Among the five mortalities during the follow-up,three died of tumor recurrence. In terms of post-transplant complications, 9 recipients(34.6%) experienced early allograft dysfunction, 1(3.8%) had non-anastomotic biliary stricture,and 1(3.8%) was complicated with hepatic arterial thrombosis.None of these complications resulted in patient death. | 12 | Greenhouse gas emissions from a constructed wetland for municipal sewage treatment显示文摘The fluxes of greenhouse gases (methane and nitrous oxide) emission from a constructed wetland in the Eastern China as municipal sewage treatment were measured from June 1999 to August 2000 by the closed chamber method. The constructed wetland for municipal sewage treatment is a significant source of methane, up to 976 6×10 6 g CH 4/a, which was emitted from the constructed wetland with the area of 495000 m 2 and wastewater loading rate of 12000 m 3/d. Its daily mean methane flux reached 5 22 g CH 4/(m 2·d), 250 times as much as that in natural wetland in the same latitude region. 227 8 mg CH 4 was produced from the treatment of 1 liter wastewater, up to 700—1000 times as much as that in the secondary treatment. | 14 | Cholesterol homeostasis regulated by ABCA1 is critical for retinal ganglion cell survival显示文摘Genome-wide association studies have suggested a link between primary open-angle glaucoma and the function of ABCA1.ABCA1 is a key regulator of cholesterol efflux and the biogenesis of high-density lipoprotein(HDL) particles. Here, we showed that the POAG risk allele near ABCA1 attenuated ABCA1 expression in cultured cells. Consistently, POAG patients exhibited lower ABCA1 expression, reduced HDL, and higher cholesterol in white blood cells. Ablation of Abca1 in mice failed to form HDL, leading to elevated cholesterol levels in the retina. Counting retinal ganglion cells(RGCs) by using an artificial intelligence(AI) program revealed that Abca1-deficient mice progressively lost RGCs with age. Single-cell RNA sequencing(scRNA-seq) revealed aberrant oxidative phosphorylation in the Abca1-/-retina, as well as activation of the mTORC1 signaling pathway and suppression of autophagy. Treatment of Abca1-/-mice using atorvastatin reduced the cholesterol level in the retina,thereby improving metabolism and protecting RGCs from death. Collectively, we show that lower ABCA1 expression and lower HDL are risk factors for POAG. Accumulated cholesterol in the Abca1-/-retina causes profound aberrant metabolism, leading to a POAG-like phenotype that can be prevented by atorvastatin. To investigate the proximate causes of dispersal in female Yunnan snub-nosed monkeys (Rhinopithecus bieti), we observed and analyzed nine years of detailed dispersal and demographic data from a population of R. bieti in Xiangguqing, Baimaxueshan Nature Reserve, Yunnan Province, China. Results showed that females who lived long-term in a one-male unit (OMU), without giving birth and with few or no relatives, were more likely to leave that OMU. In addition, an OMU led by an outgroup male and containing more female relatives was significantly more likely to be chosen for immigration. Conversely, greater male age, longer male tenure, and more potentially fertile females discouraged immigration into an OMU.
