1Effect of Silicon on Leaf Ultrastructure, Chlorophyll Content and Photosynthetic Activity of Barley Under Salt Stress显示文摘Two contrasting cultivars of barley (Horderm vykgare L.): Kepin No. 7 (salt sensitive), and Jian 4 (salt tolerant) were grown in a hydroponics system with 2 NaCl levels: 60 mmol NaCl L-1 and 120 mmol NaCl L-1, and 3 Si levels: 0 mmol Si L-1, 0.5 mmol Si L-1 and 1.0 mmol Si L-1 (as silicic acid). Compared with the plants treated with 60 mmol NaCl L-1 alone, the leaf chlorophyll contents of plants treated with salt and Si increased significantly for salt-sensitive cultivar at tillering stage, but for salt-tolerant cultivar,the addition of Si resulted in an obvious increase in the leaf chlorophyll content of plants exposed to 120 mmol NaCl L-1. However, this Si-enhancement of leaf chlorophyll content was also observed in the salttolerant plants at jointing stage, but not in the salt-sensitive plants. Moreover, leaf chlorophyll content was consistently higher for the salt-tolerant cultivar than for the salt-sensitive cultivar irrespective of salt and/or Si treatment. Compared with the plants treated with salt alone, net CO2 assimilation rate in plant leaves increased significantly for both cultivars when trested with salt and Si. The addition of Si to the salt treatment was found to improve the cell ultrastructure of leaves. Under salt stress condition, the double membranes of chloroplasts disappeared, but membrane integrity was markedly improved in the salt treatment supplemented with Si. Silicon was also found to ameliorate the damage to the ultrastructure of chloroplast granae which appeared to be disintegrated and vague in salt treatments without added Si. 2Relationship between osmotic stress and polyamines conjugated to the deoxyribonucleic acid-protein in wheat seedling roots显示文摘The contents of covalently conjugated polyamines (CC-PAs) and noncovalently conju- gated polyamines (NCC-PAs) to deoxyribonucleic acid-protein (DNP) isolated from wheat (Triticum aestivum L.) seedling roots under osmotic stress were detected. Results showed that after osmotic stress treatment for 7 d, the levels in NCC-spermine (NCC-Spm) and NCC-spermidine (NCC-Spd) of drought-tolerant Yumai No. 18 cv. increased more markedly than that of drought-sensitive Yangmai No. 9 cv., while the NCC-putrescine (NCC-Put) could not be statistically detected in two cultivars. Exogenous Spm treatment alleviated osmotic stress injury to Yangmai No. 9 cv. seedlings, coupled with marked increases of NCC-Spm and NCC-Spd levels of this cultivar. Under PEG osmotic stress, the concomitant treatment of drought-tolerant Yumai No. 18 cv.seedlings with methylglyoxyl-bis (guanylhydrazone) (MGBG), an inhibitor of S-adenosylmethionine decarboxylase (SAMDC), aggravated osmotic stress injury to this cultivar, coupled with market decreases of the NCC-Spm and NCC- Spd levels. The levels in CC-Put and CC-Spd of drought-tolerant Yumai No. 18 cv. increased more markedly than that of drought-sensitive Yangmai No. 9 cv. Under osmotic stress. The treatment of drought-tolerant Yumai No. 18 cv. seedlings with phenanthrolin (o-Phen), an inhibitor of transglutaminase (TGase), aggravated osmotic stress injury to this cultivar, coupled with a reduction of sum contents of CC-Put+CC-Spd. 3Relationships Between Balance of Nitrogen Supply-Demandand Nitrogen Translocation and Senescenceof Different Position Leaves on Rice显示文摘By removing panicles and spikelets, the effects of nitrogen supply-demand balance (NSDB) onnitrogen transiocation and senescence of leaves at different positions of the same stem in rice plant werestudied. The results showed that under NSDB<0 (CK), nitrogen translocation and senescence of the top 4leaves was normal. Nitrogen exported from the top leaves (LFT) especially the top 4 leaves decreased remar-kably with increase of NSDB, thus, their MDA contents and differences in color (SPAD value) decreased,while the activities of SOD and CAT increased. Under NSDB>0 (panicle removal), N accumulation of the top4 leaves increased significantly, and the leaf senescence process was delayed. 4Stress of Cl- is Stronger than That of Na+ on Glycine max Seedlings Under NaCl Stress显示文摘Seedlings of six Glycine max cultivars were used to compare Cl- with Na+ in stress effects onsoybean seedlings under NaCl stress. Results showed that stress of NaCl on G. max seedlings was mainlycaused by Cl- and not by Na+. After treatment with isoosmotic solutions of Cl- (without Na+ ) and Na+ (with-out Cl-) respectively, fresh weight and height of G. max seedlings growing in solution of Cl- were much morestressed than those in solution of Na+. And the roots and leaves electrolyte leakage were much more increasedunder stress of Cl- than those under stress of Na+. 5Effects of Panicle Nitrogen Fertilization on Non-Structural Carbohydrate and Grain Filling in Indica Rice显示文摘Grain filling, a crucial stage of grain yield formation in rice, is usually affected by the panicle nitrogen (N) fertilization. Field and pot culture experiments were conducted to explore the underlying mechanisms of N effect. Two rice cultivars with high lodging resistance were grown in the field and pot. Four panicle N fertilization treatments were conducted in 2006 and repeated in 2007. The result showed that medium level of panicle N fertilization treatment (NM) enhanced the accumulation and translocation of non-structural carbohydrate (NSC) in the stem and sheath. Compared with non-nitrogen treatment (N0), NM promoted the translocation of labeled 13C from stem and sheath to grain. But, low level of panicle N fertilization treatment (NL) and high level of panicle N fertilization treatment (NH) showed the negative effect. The endosperm cell, grain length, and grain width of NM increased more quickly than that of N0 from 4 to 10 d after anthesis. During the early period of grain filling, sucrose-phosphate synthase (EC 2.4.1.14, SPS) activity were significantly higher for the NM treatment than those of the NL and NH treatments. Sucrose synthase (EC 2.4.1.13, SuSase) activity in the grains was substantially enhanced by NM, with the duration of higher activity being longer than those of the other treatments. At maturing stage, NM significantly increased the filled grain number, the seed-setting rate, and the grain weight compared with NL and NH. 6Monitoring Protein and Starch Accumulation in Wheat Grains with Leaf SPAD and Canopy Spectral Reflectance显示文摘The research was conducted to determine the relationships of protein and starch accumulation dynamics in grains of wheat to post-heading leaf SPAD values and canopy spectral reflectance. The results showed that leaf nitrogen accumulation was exponentially related to leaf SPAD values and linearly related to canopy spectral reflectance, and that there was negative linear relationship between leaf nitrogen accumulation and grain protein accumulation, but positive linear relationship between post-heading leaf nitrogen translocation and grain protein accumulation at maturity. In addition, leaf SPAD values were parabolically related with and ratio indices R(1 500,610) and R(1 220,560) were exponentially related with protein and starch accumulation in grains. 7A Dynamic Knowledge Model for Design of Suitable Sowing Date and Sowing Rate in Winter Wheat显示文摘Based on research concerning dynamic relationships of winter wheat growth to environments and production conditions, a winter wheat model for selecting suitable sowing date, population density and sowing rate under different varieties, spatial and temporal environments was developed. 8A Dynamic Knowledge Model for Nitrogen Fertilizationin Wheat Management显示文摘By analyzing and extracting the research progress on nitrogen fertilization in wheat, a dynamic knowledge model for management decision-making on total nitrogen rate, ratios of organic to inorganic and of basal to dressing nitrogen under different environments and cultivars in wheat was developed with principle of nutrient balance and by integrating the quantitative effects of grain yield and quality targets, soil characters, variety traits and water management levels. 9Interactions of Water Management and Nitrogen Fertilizer on Nitrogen Absorption and Utilization in Rice显示文摘The interactions of water management and nitrogen fertilizer on nitrogen absorption and utilization were studied in rice with Wuxiangjing9 (japonica). The results showed that the nitrogen uptake and remaining in straw increased and the percentage of nitrogen translocation (PNT) from vegetative organs, nitrogen dry matter production efficiency (NDMPE) and nitrogen grain production efficiency (NGPE) decreased with nitrogen increasing. The nitrogen uptake and NGPE decreased when severe water stressed. However, rice not only decreased the nitrogen uptake but also increased the PNT from vegetative organs, NDMPE and NGPE when mild water stressed. 10Subtropical Modern Greenhouse Cucumber Canopy Transpiration Under Summer Climate Condition显示文摘Greenhouse canopy transpiration not only has effects on greenhouse air temperature and humidity, but also is important for determining the set-point of fertigation. In this study, Penman-Monteith equation was used to calculate the greenhouse cucumber canopy transpiration under summer climate condition.The effects of greenhouse environmental factors on canopy transpiration were analyzed based on the measurements of greenhouse microclimate factors and canopy transpiration. The results showed that Penman-Monteith equation was reliable and robust in estimating greenhouse cucumber canopy transpiration under summer climate condition. Greenhouse cucumber canopy transpiration rate increased linearly with the increase of net radiation and water vapor pressure deficit (VPD) above the canopy. But the maximum value of the canopy transpiration rate occurred at the same time as that of VPD whereas about two hours later than that of net radiation. 11A Knowledge Model-and Growth Model-Based Decision Support System for Wheat Management显示文摘By applying the system analysis principle and mathematical modeling technique to knowledge expression system for crop cultural management, the fundamental relationships and quantitative algorithms of wheat growth and management indices to variety types, ecological environments and production levels were analysed and extracted, and a dynamic knowledge model with temporal and spatial characters for wheat management (WheatKnow) was developed. By adopting the soft component characteristics as non language rele vance, re-utilization and portable system maintenance, and by further integrating the wheat growth simulation model (WheatGrow) and intelligent system for wheat management, a comprehensive and digital knowledge model, growth model and component-based decision support system for wheat management (MBDSSWM) was established on the platforms of Visual C++ and Visual Basic. 12Construction of sperm-specific lactate dehydrogenase DNA vaccine and experimental study of its immuno-contraceptive effect on mice显示文摘Lactate dehydrogenase C4 (LDHC4) is a key enzyme for sperm metabolism. It is distributed specifically in testis and is highly immunogenic. In this study, two DNA vaccines pVAX1-hLDHC and pVAX1-mLDHC were constructed by inserting coding sequences of human and mice LDHC4 into the eukaryotic ex-pression vector pVAX1. The production of LDHC4 specific antibodies was induced in the sera of vac-cinated mice and the reproductive tract secretions of vaccinated female mice through immunization by mucosal surface instillation. Furthermore, the antibody titer increased with the times of immunization. In the mating experiment, the number of newborns of the vaccinated mice reduced significantly and some immunized female mice even lost the ability to bear any offsprings, suggesting that the difference between the immunized and control mice was statistically significant. Sperm agglutination analysis indicated that both the antisera from immunized mice and the reproductive tract secretions of vacci-nated female mice could agglutinate normal sperms. Results of immunohistochemistry showed that the antibodies present in the sera of immunized mice and the reproductive tract secretions of vaccinated female mice could specifically react with LDHC4 antigen, which mainly locates in the cytoplasm, acrosome membrane externa and acrosome capsule of the sperm. 13Response of the enzymes to nitrogen applications in cotton fiber (Gossypium hirsutum L.) and their relationships with fiber strength显示文摘To investigate the response of key enzymes to nitrogen (N) rates in cotton fiber and its relationship with fiber strength, experiments were conducted in 2005 and 2006 with cotton cultivars in Nanjing. Three N rates 0, 240 and 480 kgN/hm2, signifying optimum and excessive nitrogen application levels were applied.The activities and the gene expressions of the key enzymes were affected by N, and the characteristics of cellulose accumulation and fiber strength changed as the N rate varied. Beta-1,3-glucanase activity in cotton fiber declined from 9 DPA till boll opening, and the beta-1, 3-glucanase coding gene expression also followed a unimodal curve in 12—24 DPA. 14Distribution of Leaf Color and Nitrogen Nutrition Diagnosis in Rice Plant显示文摘Greenness and nitrogen content of each leaf on main stem of different japonica and indica ricevarieties under different nitrogen levels were investigated. Results showed that the fourth leaf from the top ex-hibited active changes with the change of plant nitrogen status. When the plant nitrogen content was low, itscolor and nitrogen content were obviously lower than those of the three top leaves. With the increase of plantnitrogen content, the color and nitrogen content of the fourth leaf increased quickly, and the differences ofcolor and nitrogen content between the fourth leaf and the three top leaves decreased. So, the fourth leaf wasan ideal indication of plant nutrition status. In addition, color difference between the fourth and the third leaffrom the top was highly related to the plant nitrogen content regardless of the variety and development stage.Therefore, color difference between the fourth and the third leaf could be widely used for diagnosis of plantnutrition. Results also indicated that the minimized color difference between the fourth and the third leaf at the criticaleffective tillering, the emergence of the second leaf from the top, and the heading was the symbol of high yield. 15A Process-Based Model for Simulating Phasic Developmentand Phenology in Rice显示文摘A simulation model for phasic and phenological development of rice was developed using the scale of physiological development time, based on the ecophysiological development processes. The interaction of daily thermal effectiveness, photoperiod effectiveness and intrinsic earliness (before heading), and basic filling duration factor (after heading) determined the daily physiological effectiveness, which accumulated to get physiological development time. The Beta and quadratic functions were used to describe daily thermal and photoperiod effectiveness, respectively. Five specific genetic parameters were added to adjust the genotypic differences in rice development so that all different varieties could reach the same physiological development time at a given development stage. The stages of seedling emergence, panicle initiation, heading, and maturity were validated using sowing dates under different ecological environments, with the RMSE of 1.47, 5.10,4.58 and 3.37 days, respectively.
