1The immunotherapeutic effect of dendritic cells vaccine modified with interleukin-18 gene and tumor cell lysate on mice with pancreatic carcinoma显示文摘AIM:To estimate the effect of a therapeutic vaccine against pancreatic carcinoma based on dendritic cell(DC)vaccine modified with tumor lysate and Interleukin-18 gene.METHODS:The BALB/C mice model of pancreatic arcinoma was induced with DMBA,DCvaccine was construted through pulsed with tumor lysate and transfected by the recombinant adenoviral vector encoding IL-18gene.The immnotherapeutic effects of DC vaccine on mice with pancreatic carcinoma were assessed (divided intoD-IL18-Lysate group,DC-Lysate group,DC-IL18 group,DC group,PBSgroup).RESULTS:After vaccination of the DC vaccine,the concentration of IL-18and IFN-γwere 2161±439ng·L^-1and435±72ng·L^-1inDC-IL18-Lysate group and there was significant difference compared with other groups(P<0.01)After vaccination of the DCcvaccine,the transplanted tumors were observed on 30days inDC-Lysate groups.on 16daysinDC-IL18groups.on3days in control group,but mice remained tumor-free for at least50days in DC-IL18-Lysate group and there was significant difference between DC-IL18-Lysate group and other groups(P<0.01).The median survival exceeds62days in DC-IL18-Lysate group.But the medsian surival was 48.6days in DC-Lysate group,33days in DC-IL-18group,17days in PBSgroup.The survival period was obviously prolonged in DC-IL18-Lysate group than in other groups(P<0.05,P<0.01).The weight of pancreatic tumor was0.22±0.083ginDC-IL18-Lysate group,1.45±0.74ginDC-Lysate group,1.89±1.34gin DC-IL18group,3.0±1.6ginDCgroup,2.9±2.0ginPBSgroup and the weight of tumor obviously reduced in DC-IL-18-Lysate group than in other groups(P<0.05,P<0.01).CONCLUSION:DC vaccine modified with tumo lysate and Interleukin-18gene can induce a specific and effective immune response against pancreatic cancinoma,cell.Zhao-Hui Tang Gao-Song Wu Sheng-Quan Zou Fa-Zhu Qiu Department of Surgery of Tong Ji Hospital Wen-Hong Qiu Department of immunology,Tongji Medical College,Huazhong University of Science and Technology,Wuhan 430030,Hubei Province,China Xiang-Ping Yang Department of Biochemistry,Rheinisch-Westfalische Technische Hochschule(RWTH),D-52074 Aachen,Germany 2002World Journal of Gastroenterology2002,8,5:37
2Ensemble projection of 1―3℃ warming in China显示文摘Studies on the influences of climate change on biogeochemical cycles and on the key vulnerabilities and the risk from climate change suggest that annual surface temperature rise of 1℃, 2℃ and 3℃ above the present level would lead to changes in extreme weather and climate events, food production, fresh water resources, biodiversity, human mortality, etc. Here two sets of simulations as performed with seventeen atmosphere-ocean general circulation models (AOGCMs) for the Fourth Assessment Report of the Intergovernmental Panel on Climate Change (IPCC AR4), i.e. the model outputs from the 20th Century Climate in Coupled Models (20C3M) and from the Special Report on Emissions Scenarios (SRES) emission scenarios B1, A1B and A2, are used to analyze spatial and temporal characteristics of the above values in China over the 21st century. The results indicate that the rate of warming varies from region to region. The above values are reached much later (earlier) when emission amount is lower (higher), and spread of the time when the lower (higher) value is exceeded is narrower (wider) among the three scenarios. 3Veno occlusive disease: Update on clinical management显示文摘Hepatic veno-occlusive disease is a clinical syndrome characterized by hepatomegaly, ascites, weight gain and jaundice, due to sinusoidal congestion which can be caused by alkaloid ingestion, but the most frequent cause is haematopoietic stem cell transplantation (STC) and is also seen after solid organ transplantation. The incidence of veno occlusive disease (VOD) after STC ranges from 0 to 70%, but is decreasing. Survival is good when VOD is a mild form, but when it is severe and associated with an increase of hepatic venous pressure gradient > 20 mmHg, and mortality is about 90%. Prevention remains the best therapeutic strategy, by using non-myeloablative conditioning regimens before STC. Prophylactic administration of ursodeoxycholic acid, being an antioxidant and antiapoptotic agent, can have some benefit in reducing overall mortality. Defibrotide, which has pro-fibrinolytic and antithrombotic properties, is the most effective therapy; decompression of the sinusoids by a transjugular intrahepatic portosystemic shunt (TIPS) can be tried, especially to treat VOD after liver transplantation and when multiorgan failure (MOF) is not present. For an environmental magnetic study, it is important to understand the magnetic properties of various magnetic minerals in lacustrine sediments and the processes involved in the formation, transport, and preservation of these magnetic minerals. The magnetic response of lacustrine sediments to climatic change may vary widely from lake to lake. 6VEGF-D expression correlates with colorectal cancer aggressiveness and is downregulated by cetuximab显示文摘AIM:To gain mechanistic insights into the role played by epidermal growth factor receptor (EGFR) in the regulation of vascular endothelial growth factors (VEGFs) in colorectal cancer (CRC). METHODS:The impact of high-level expression of the growth factor receptors EGFR and VEGF receptor (VEGFR)3 and the VEGFR3 ligands VEGF-C and VEGF-D on disease progression and prognosis in human CRC was investigated in 108 patients using immunohistochemistry. Furthermore, the expression of the lymphangiogenic factors in response to the modulation of EGFR signalling by the EGFR-targeted monoclonal antibody cetuximab was investigated at the mRNA and protein level in human SW480 and SW620 CRC cell lines and a mouse xenograft model. RESULTS: Human CRC specimens and cell lines displayed EGFR, VEGF-C and VEGF-D expression with varying intensities. VEGF-C expression was associated with histological grade. Strong expression of VEGF-D was significantly associated with lymph node metastases and linked to a trend for decreased survival in lymph node-positive patients. EGFR blockade with cetuximab resulted in a significant decrease of VEGF-D expression in vitro and in vivo. 7Dislocation structure of the crust-mantle boundary and low-velocity body within the crust beneath the Dabie Shan collision orogen显示文摘From April of 2001 to March of 2002, a passive seismic array experiment was car- ried out in the Dabie Shan and its adjacent region. In this experiment, totally 34 broadband seismic stations were deployed along a profile across the Dabie Shan orogen and North-China platform. This profile is about 500 km long from Cuilin (34°40′N, 114°49′E), Henan Province, to Dajipu (30°20′N, 115°03′E), Hubei Province. The space between stations is about 3?8 km in the Dabie Shan orogenic belt and about 15?20 km in other area. The receiver function profile and S-wave velocity structure of the crust and upper mantle down to 100 km depth along the profile are investigated in terms of the receiver function techniques (Liu et al., 1996, 2000). Our results show that the crust beneath the Dabie Shan orogen has an obviously asymmetric blocked structure in the direction perpendicular to the mountain strike. The maximal crustal thickness reaches to 42 km. The crust-mantle boundary has a dislocation structure correlated to the crustal blocks and the largest offset reaches to 8 km. In the kernel of the orogen exists a low-velocity body inside the crust, which is separated into two parts corresponding to the South Dabie and North Dabie on the surface, respectively. Probably a vertical divergent movement between both took place in history. The crust below this low-velocity area has a positive gradient velocity structure with the depth, and the upper mantle down to the depth of 70 km has the lower S-wave velocity than its both sides. 8Appendicitis in children less than five years old:A challenge for the general practitioner显示文摘Acute appendicitis is one of the most common indications for abdominal surgery in pediatrics with peak incidence in the second decade of life. Acute appendicitis in the first years of life is an uncommon event. The clinical presentation is often varied and the diagnosis may be overshadowed by other medical conditions.Gastroenteritis is the most common misdiagnosis, with a history of diarrhea present in 33% to 41% of patients. Pain is the most common presenting symptom in children less than 5 years old, followed by vomiting, fever, anorexia and diarrhea. The most common physical sign is focal tenderness(61% of the patients) followed by guarding(55%), diffuse tenderness(39%), rebound(32%), and mass(6%). Neonatal appendicitis is a very rare disease with high mortality; presenting symptoms are nonspecific with abdominal distension representing the main clinical presentation. The younger the patient, the earlier perforation occurs: 70% of patients less than 3 years develop a perforation within 48 h of onset of symptoms. A timely diagnosis reduces the risk of complications. 10Heat shock induction of a 65 kDa ATP-binding proteinase in rat C6 glioma cells显示文摘The 45, 55, 65 and 100 kDa ATP-binding proteinases (ATP-BPases) of the heat-shocked (44 ℃ for 30 min, recovery for 12h) rat C6 glioma cells were purified by DEAE-ionexchange and ATP-affinity chromatography. Their molecular masses, isoelectric points (pI), pH-optima and other properties were analyzed by native proteinase gels.It was shown that the 65 kDa ATP-BPase is specifically induced by heat shock and not detectable in control cells.Its N-terminal 1-9 amino acid sequence was determined by Edman degradation, but no homologies to other proteins in the protein data bases were found. 30 and 31 kDa proteinases can be cleaved from the 45, 55 and 65 kDa proteinases to which they are linked. 12Rock-weathering by lichens in Antarctic: patterns and mechanisms显示文摘Saxicolous species of lichens are able to induce and accelerate weathering of their rock substrate, and effects of lichens on substrate can be attributed to both physical and chemical causes. This paper is focused on biotic weathering actions of epilithic and endolithic species on the different rock types (sandstones and volcanogenic rocks) in Antarctica. The patterns, mechanisms, processes and neoformations of rock-weathering resulting from lichen colonization are expounded in detail. 13Determination of Elastic and Creep Properties of Thin-film Systems from indentation Experiments显示文摘Based on the detailed computer .simulation of the indentation testing on the thin-film systems, the present paper explores the detailed procedure of determining elastic properties (elastic modulus E(f) and Poisson ratio v(f) ) and creep parameters (CCREEP (f) and nCREEP (f)) for a simple Norton law (ε=CCREEP (f)σnCREEP(f), where ε is creep strain rate, and σ is the stress) material for a thin film coated on a creep substrate, whose elastic properties(E(s) and v(s)) and creep properties (CCREEP(s) and nCREEP(s) ) of the substrate are known, from indentation elastic and creep testing, respectively. The influences of the thickness of the thin-film and the size of the indenter on the indentation behavior have been discussed. It is shown that the boundary between the thin film and the substrate has great influence on the indentation creep behavior. The relative sizes of indentation systems are chosen so that the behavior of the indentation on the film is influenced by the substrate. The two elastic parameters E(f) and v(f) of the film are coupled on the influence of the elastic behavior of indentation. With the two different size indenters, the two elastic parameters E(f) and v(f) of the film can be uniquely determined by the indentation experimental slopes of depth to applied net section stress results. The procedure of determining of the two Norton law parameters CCREEP (f) and nCREEP(f) includes the following steps by the steady indentation rate d. The first step to calculate the creep indentation rate on certain loads of the two different sizes of indenters on a set of assumed values of CCREEP (f) and nCREEP (f). Then to build relationship between the creep indentation rate and the assumed CCREEP(f) and nCREEP(f). With the experimental creep indentation rate to intersect two sets of CCREEP (f) and nCREEP(f), which have the same values of d. The last step is to build the CCREEP(f) and nCREEP(f)curves from the intersection points for the two indenters.These two curves CCREEP and nCREEP(f) will intersect at one point. The axial values of the point are what we want. 14NaAlSi_3O_8-hollandite and other high-pressure minerals in the shock melt veins of the Suizhou meteorite显示文摘The Suizhou L6 chondrite contains a few very thin shock melt veins of 0.02-0.09 mm in width. In spite of small width of the veins, shock-induced high-pressure phases, such as coarse-grained NaAlSi3O8-hollandite, ringwoodite, majorite and fine-grained matrix majorite-pyropess have been discovered in these veins. NaAlSi3O8-hollandite, the high-pressure phase of plagioclase, in Suizhou shock veins occurs as a single phase mineral, no silicate glassy phase, such as albitic glass, was incorporated with it. The presence of above-mentioned high-pressure phases constrains the high pressure (up to 23-24 GPa) and high temperature (up to 1900-2000℃) regime in Suizhou shock veins, and indicates that the duration of high-pressure regime in the veins should be long enough (a few seconds) for phase transformation and crystallization of minerals under pressure. 16Integral Recrystallization Modelling显示文摘We present models designed to represent the evolution of microstructure and crystallographic texture during recrystallization. For engineering applications the models are tied to a finite element approach. A connection to the deformed microstructure is established by dislocation theory of work hardening and Taylor type texture simulations. 18Liver transplantation for viral hepatitis in 2015显示文摘Liver transplantation(LT) is a life-saving treatment forpatients with end-stage liver disease and for patients with liver cell cancer related to liver disease. Acute and chronic liver diseases related to hepatitis viruses are between the main indications for liver transplantation. The risk of viral reinfection after transplantation is the main limiting factor in these indications. Before the availability of antiviral prophylaxis, hepatitis B virus(HBV) recurrence was universal in patients who were HBV DNA-positive before transplantation. The natural history of recurrent HBV was accelerated by immunosuppression, and it progressed rapidly to graft failure and death. Introduction of post-transplant prophylaxis with immunoglobulin alone first, and associated to antiviral drugs later, drastically reduced HBV recurrence, resulting in excellent long-term outcomes. On the contrary, recurrence of hepatitis C is the main cause of graft loss in most transplant programs. Overall, patient and graft survival after LT for hepatitis C virus(HCV)-associated cirrhosis is inferior compared with other indications. However, successful pretransplant or post transplant antiviral therapy has been associated with increased graft and overall survival. Until recently, the combination of pegylated interferon and ribavirin was the standard of care for the treatment of patients with chronic hepatitis C. 19Markers of acute rejection and graft acceptance in liver transplantation显示文摘The evaluation of the immunosuppression state in liver transplanted patients is crucial for a correct posttransplant management and a major step towards the personalisation of the immunosuppressive therapy. However, current immunological monitoring after liver transplantation relies mainly on clinical judgment and on immunosuppressive drug levels, without a proper assessment of the real suppression of theimmunological system. Various markers have been studied in an attempt to identify a specific indicator of graft rejection and graft acceptance after liver transplantation. Considering acute rejection, the most studied markers are pro-inflammatory and immunoregulatory cytokines and other proteins related to inflammation. However there is considerable overlap with other conditions, and only few of them have been validated. Standard liver tests cannot be used as markers of graft rejection due to their low sensitivity and specificity and the weak correlation with the severity of histopathological findings. Several studies have been performed to identify biomarkers of tolerance in liver transplanted patients. Most of them are based on the analysis of peripheral blood samples and on the use of transcriptional profiling techniques. 20Invasive fungal infection before and after liver transplantation显示文摘Invasive infections are a major complication before liver transplantation(LT)and in the early phase after surgery.There has been an increasing prevalence of invasive fungal disease(IFD),especially among the sickest patients with decompensated cirrhosis and acute-on-chronic liver failure,who suffer from a profound state of immune dysfunction and receive intensive care management.In such patients,who are listed for LT,development of an IFD often worsens hepatic and extra-hepatic organ dysfunction,requiring a careful evaluation before surgery.In the post-transplant setting,the burden of IFD has been reduced after the clinical advent of antifungal prophylaxis,even if several major issues still remain,such as duration,target population and drug type(s).Nevertheless,the development of IFD in the early phase after surgery significantly impairs graft and patient survival.This review outlines presentation,prophylactic and therapeutic strategies,and outcomes of IFD in LT candidates and recipients,providing specific considerations for clinical practice.Alberto Ferrarese Annamaria Cattelan Umberto Cillo Enrico Gringeri Francesco Paolo Russo Giacomo Germani Martina Gambato Patrizia Burra Marco Senzolo 2020World Journal of Gastroenterology2020,26,47:6
