4Experimental and clinic-opathologic study on the relationship between transcription factor Egr-1 and esophageal carcinoma显示文摘AIM To observe the growth suppression effect of exogenous introduction of early growth response gene-1 (Egr-1 gene) on esophageal carcinoma tissue as well as on esophageal carcinoma cell line Eca109 and to explore the potential application of Egr-1 gene in gene therapy of tumor.METHODS Eukaryotic expression vector of PCMV-Egr-1 plasmid was introduced into Eca109 cell line which expressed no Egr-1 protein originally with lipofectamine transfection method. The introduction and expression of PCMV-Egr-1 plasmid into Eca109 cell line was confirmed by G418 selection culture, PCR amplification of neogene contained in the vector, Western blot analysis and immunocytochemical analysis. The cell growth curve,soft agar colony formation rate and tumorigenicity in SClD mice were examined to demonstrate the growth suppression effect of exogenous Egr-1 gene on Eca109 cell line. The Egr-1 mRNA and Egr-1 protein were also detected in 50 surgical specimens of esophageal carcinoma by in situ hybridization and immunohistochemistry.RESULTS Exogenous Egr-1 gene was introduced successfully into Eca109 cell line and expressed Egr-1 protein stably. The transfected Eca109 cell line grew more slowly than control Eca109 as shown by cell growth curves, the soft agar colony formation rate (4.0% vs 6.9%, P<0.01) and the average growth rate of tumor in SCID mice (35.5 ± 7.6 vs 65.8 ± 7.6, P<0.05). The expression level of Egr-1 mRNA and protein significantly increased in dysplastic epithelia adjacent to cancer rather than in cancer tissues (65.8% vs 20.0% by ISH and 57.9% vs 14.0% by IHC, P<0.01).CONCLUSION Exogenous Egr-1 gene shows the strong effect of growth inhibition in Eca109 cell line. 5Killing effect of TNF-related apoptosis inducing ligand regulated by tetracycline on gastric cancer cell line NCI-N87显示文摘AIM To clone the cDNA fragment of human TRAIL (TNFrelated apoptosis inducing ligand) into a tetracyclineregulated gene expression system, the RevTet-On system, transduce expression vectors into a gastric carcinoma cell line-NCl-N87 and examine the effects of controlled expression of TRAIL in vitro on the gastric carcinoma cells.METHODS The full-length cDNA of TRAIL was inserted into a vector under the control of the tetracyclineresponsive element (TRE) to obtain the plasmid pRevTRETRAIL, which was transfected into a packaging cell line PT67. In addition, vector pRev-Tet- On and pRevTRE were also transfected into PT67 separately. After hygromycin and G418 selection, the viral titer was determined. The medium containing retroviral vectors was collected and used to transduce a gastric carcinoma cell line NCI-N87.The resulting cell line NCI-N87-Tet-On-TRE-TRAIL and a control cell line, NCI-N87-Tet-On-TRE, were established.TRAIL expression in the cell line was induced by incubating cells with doxycycline (Dox), which is a tetracycline analogue. The killing effect on gastric carcinoma cells was analyzed after induction.RESULTS The recombinant plasmid pRev-TRE-TRAIL was constructed. After hygromycin or G418 selection, the producer cell lines PT67-TRE, PT67-TRE-TRAIL and PT67TetOn were obtained, with titers of about 108CFU@ L-1 By transducing NCI-N87 cells with retroviral vectors from these cell lines, stable cell lines NCI-N87-Tet-On-TRETRAIL (NN3T) and control cell line NCI-N87-Tet-On-TRE (NN2T) were established. The growth curves of the selected cell lines were the same with the wild type NCIN87. When Dox was added, cell death was obvious in the test groups (29% -77%), whereas no difference was observed in control and wild type cell lines. 8Morphology and mapping analysis of rice(Oryza sativa L.)clustered spikelets(Cl)mutant显示文摘The rice clustered spikelets (Cl) mutant exhib-its a phenotype that most of branch apical have 2-3 spikelets clustered together. SEM (scanning electron microscope) ob-servation suggested that the Cl gene controlled branch apical development, and influenced the terminal spikelets elonga-tion. The spikelet number was reduced in mutant, indicating that Cl may also have an effect on spikelet number. To map Cl locus, two F2 mapping populations derived from the crosses between the Cl and ZhongHua11, and Cl and ZheFu802 were constructed, respectively. The Cl locus was roughly mapped between two CAPS markers, CK0214 and SS0324. A further fine mapping analysis showed that the Cl locus was mapped between makers R0674E and Cl2560, with genetic distances of 0.2 and 2.1 cM, respectively. Then we found a PAC contig spanning Cl locus, the region was delim-ited to 196 kb. This result was useful for cloning of the Cl gene. 10Four types of bounded wave solutions of CH-■ equation显示文摘Recently, many authors have studied the following CH-γequation: ut + c0ux + 3uux -α2(wxxt + uuxxx + 2uxuxx) 4-γuxxx = 0,whereα2, c0 andγare paramters. Its bounded wave solutions have been investigated mainly for the caseα2 > 0. For the caseα2 < 0, the existence of three bounded waves (regular solitary waves, compactons, periodic peakons) was pointed out by Dullin et al. But the proof has not been given. In this paper, not only the existence of four types of bounded waves: periodic waves, compacton-like waves, kink-like waves, regular solitary waves, is shown, but also their explicit expressions or implicit expressions are given for the caseα2 < 0. 13Paleoenvironmental information recorded by pollen in B-3GC gravity core in Okinawa Trough显示文摘Based on pollen records in B-3GC gravity core, environmental change since 9500 aBP of Okinawa Trough and its adjacent islands was derived. The result showed that the most time during this period was in a warm temperate climate except in middle Holocene (6800-4400 aBP) that was under subtropical climate control. During 9500-8300 aBP and 3100-2000 aBP periods, it appeared colder and drier than the rest time. The original area of pollen sources surrounding Okinawa Trough was covered by evergreen and deciduous broadleaf forest alternatively with mixed broad-leaf-conifer forest distributed in high mountainous areas. Usually, these three kinds of forests existed at the same time with difference in altitude. Pollen from subtropical and tropical plants increased obviously in about 5000-6000 aBP, reflecting a great lifting of vegetation zone and expansion of evergreen broad-leaf forest in the study area. 14TWO-DIMENSIONAL SEDIMENT MATHEMATICAL MODEL FORUNSTEADY FLOW IN THE LOWER YELLOW RIVER显示文摘The theoretical frame of a new two-dimensional sediment mathematical model is established on the basis of the resent research achievements about the sediment transport law of the Yellow River. The formulas of sediment carrying capacity of flow and river roughness, which reflect the features of the Yellow River, are applied to it. The finite element scheme is employed to deduce the discretized equations of the model. The pre-estimated-verified substitution is used to solve them. Verifications of the model are carried out with the flood occurring on Jinan reach of the Yellow River during the flood season in 1976. 18Weighted L^p-estimates for Stokes flow in R_+~n with applications to the non-stationary Navier-Stokes flow显示文摘We study the time-decay properties of weighted norms of solutions to the Stokes equations and the Navier-Stokes equations in the half-space Rn+ (n 2). Three kinds of the weighted Lp-Lr estimates are established for the Stokes semigroup generated by the Stokes operator in the half-space R+n (n 2).
