| 2 | Design of a new dynamical core for global atmospheric models based on some efficient numerical methods显示文摘A careful study on the integral properties of the primitive hydrostatic balance equations for baroclinic atmosphere is carried out, and a new scheme todesign the global adiabatic model of atmospheric dynamics ispresented. This scheme includes a method of weighted equal-areamesh and a fully discrete finite difference method with quadraticand linear conservations for solving the primitive equationsystem. Using this scheme, we established a new dynamical corewith adjustable high resolution acceptable to the availablecomputer capability, which can be very stable without anyfiltering and smoothing. Especially, some important integralproperties are kept unchanged, such as the anti-symmetries of thehorizontal advection operators and the vertical convectionoperator, the mass conservation, the effective energy conservationunder the standard stratification approximation, and so on. | 3 | Dysfunction of peripheral blood dendritic cells from patients with chronic hepatitis B virus infection显示文摘AIM To identify the property of dendritic cella (DCs) of peripheral blood monocytes (PBMC) in patlents with chronic HBV infection.METHODS Twenty patients with persistent HBV infectlon were included in this study, 10 healthy subjects being used as a control group. The peripheral blood mononuclear cells (PBMC) of T cell-depleted populations were incubated and induced into mature dendritic cells in the RPMI-1640 medium in the presence of cytokines GMCSF, IL-4, FLt-3, TNF-α and 100 mL@ L-1 of fetal calf serum for a total of 10 - 12 days. The expressions of surface markers on DCs were evaluated using flow cytometric analysis. ELISA method was used to determine the cytokine levels of interleukin-12 (IL-12) and IL-10 in the supernatant produced by DCs. For detection of the stimulatory capacity of DCs to T cell proliferation,mytomycin C-treated DC were incubated with allogenic T cells.RESULTS A typical morphology of mature DCs from healthy subjects and HBV-infected patients was induced in in vitro incubation, but the proliferation ability and cellular number of DCs from HBV-infected patients significantly decreased compared with healthy individuals. In particular, the expression levels of HLADR, CD80 (B7-1) and CD86 (B7-2) on DC surface from patients were also lower than that from healthy individuals (0.46 vs 0.92 for HLA-DR, 0.44 vs 0.88 for CD80 and 0.44 vs 0. 84 for CD86, P< 0.05). | 4 | Quantitative analysis of vascular endothelial growth factor, microvascular density and their clinicopathologic features in human hepatocellular carcinoma显示文摘BACKGROUND: Angiogenesis is known to be essential to the survival, growth, invasion, and metastasis of tumor cells. Vascular endothelial growth factor (VEGF) are an important angiogenic factor regulating tumor angiogenesis, but its significance and tumor pathologic features are un- clear in hepatocellular carcinoma (HCC). In the present study, we analyzed expression of tissue VEGF, alteration of microvascular density (MVD) in microvessel angiogenesis, development and metastasis of HCC, and level of serum VEGF in differential diagnosis of benign and malignant liv- er diseases. METHODS: Tumor specimens were prospectively collected from HCC patients undergoing resection. Total RNAs were extracted and the expression levels were detected from different parts of HCC tissues. The cellular distributions of VEGF and MVD of liver tumors and their paracancerous and distal cancerous tissues were investigated by streptavi- din peroxidase (S-P) immunohistochemistry, respectively. The VEGF levels of circulating blood and hepatoma tissues were measured by enzyme-linked immunosorbent assay. RESULTS: The incidence of VEGF expression was 63.9% in HCCs (23/36 cases), 78.3% in non-encapsulated HCCs (18/23), and 90.9% in HCCs with extrahepatic metastasis (10/11), respectively. The VEGF expression was tightly correlated with MVD (P <0.01). The MVD in HCC with metastasis, low differentiation or non-encapsulation was significantly higher than that in HCC with intact capsule, high differentiation, or no metastasis. No significant diffe- rence was found between VEGF, MVD, tumor size, and hepatitis virus infection. The level of total RNA in HCC tis- sues was significantly lower but the VEGF level significantly higher than those in paracancerous or distal cancerous ones (P<0.01). The abnormal expression levels of VEGF in sera of HCC patients were directly correlated with the me- tastasis and recurrence of tumors. | 5 | Increasing the frequency of CIK cells adoptive immunotherapy may decrease risk of death in gastric cancer patients显示文摘AIM: To analyze the correlation between cytokineinduced killer (CIK) cells adoptive immunotherapy and cancer-related death in gastric cancer patients. METHODS: One hundred and fifty-six gastric cancer patients after operation at the Third Affiliated Hospital of Soochow University were enrolled in this study. Their clinical data including demographic characteristics, operation time, tumor size, pathological type and staging, tumor metastasis, outcome of chemotherapy or CIK cells adoptive immunotherapy, survival time or time of death were collected with a standard structured questionnaire. Kaplan-Meier method was used to estimate the median survival time, and the 2- and 5- year survival rates. Hazard risk (HR) and 95% confidence interval (95% CI) of CIK cells adoptive immunotherapy for gastric cancer were calculated using the two-stage time-dependent covariates Cox model. RESULTS: The survival time of gastric cancer patients was longer after CIK cells adoptive immunotherapy than after chemotherapy (χ 2 = 10.907, P = 0.001). The median survival time of gastric cancer patients was also longer after CIK cells adoptive immunotherapy than after chemotherapy (49 mo vs 27 mo, P < 0.05). The 2- and 5-year survival rates of gastric cancer patients were significantly higher after CIK cells adoptive immunotherapy than after chemotherapy (73.5% vs 52.6%, 40.4% vs 23.9%, P < 0.05). A significant difference was observed in the survival curve for patients who received CIK cells adoptive immunotherapy (0, 1-10, 11-25, and over 25 frequencies) (χ 2 = 14.534, P = 0.002). The frequencies of CIK cells adoptive immunotherapy were significantly related with the decreasing risk of death in gastric cancer patients after adjustment for sex and age of the patients, tumor stage and relapse (HR = 0.54, 95% CI: 0.36-0.80) when the first stage Cox model was used to define the subjects who remained alive beyond 36 mo as survivors. However, no correlation was observed between the frequencies of death in CIK cells adoptive immunotherapy and the risk of gastric cancer patients (HR = 1.09, 95% CI: 0.63-0.89) when the second stage Cox model was used to define the subjects who survived for more than 36 mo as survivors. | 8 | An analysis of 10218 ulcerative colitis cases in China显示文摘AIM: To analyze the characteristics of ulcerative colitis(UC)in Chine.METHODS: From 1981 to 2000, a total of 10218 patients of UCreported in Chinese medical literature and including ourcases diagnosed were analyzed according to the diagnosticcriteria of Lennard-Jones.RESULTS: The number of cases increased by 3.08 times overthe past 10 years (2506 patients were diagnosed from 1981 to1990 while 7512 patients were diagnosed from 1991 to 2000).Lesion renge were described in 7966 patients, 5592 (70.20%)were proctosigmoiditis or proctitis, 1792(22.50%) left-sidedcolitis, 582(7.30%) pancolitis. Among the 8122 patients,2826 (34.8%) had first episole, 4272 (52.6%) had chronicrelapse, 869 (10.7%) were of chronic persist type, 154(1.9%) were of acuts fulminant tyle. The course of theillress were described in 5867 patients, 4427(75.5%) wereless than 5 years, 910 (15.5%) between 5 and 10 years, 530(9.1%) more then 10 years. Six hundred and sixteenpatients patients (6.1% ) had extraintestinal manifestations.The mean age at the diagnosis was 40.7 years( range 6-8 years, and the peak ages 30-49 years). The male to femaleratio was1.09. Among 270 patients diagnosed in ourhospital, 36 had histories of smoking, there was no negativeassociation between the severity of UC and smoking ( P >0.05), 21 smokers were followed up for one year, 15 of themhad given up smoking when the disease were diagnosed,and one year later, 7 patients relapsed, another 6 patientscontinued smoking, and one year later, 2 patients relapsed.Among 270 UC patients diagnosed in our hospital, 4 patients(1.48%) from 2 families had familial history of UC.Treatment was mentioned in 6859 patients, only SASP and/or corticosteroid only in 1276 patients(18.6%), only Chineseherbs in 1377 patients (20.1%), combined Chinese andwestern medicine in 4056 patients (59.1%), surgery wasperformed in 87 patients (1.3%), other treatments in 63patients(0.9%).CONCLUSIONS: In China, number of UC patients increasedsignificantly in the past 10 years. Lesions are commonlylocated to left side colon. The course is short with rereextraintestinal manifestations. The age of onset is relativelyhigh. Males and females are nearly equally affected. Nonegative relation was found between smoking and severityof the disease. | 9 | Antitumor activities of human autologous cytokineinduced killer(CIK)cells against hepatocellular carcinoma cells in vitro and in vivo显示文摘AIM: To characterize the anticancer function of cytokine-induced killer cells (CIK) and develop an adoptiveimmunotherapy for the patients with primary hepatocellularcarcinoma (HCC), we evaluated the proliferation rate,phenotype and the antitumor activity of human CIK cellsfrom healthy donors and HCC patients in vitro and in vivo.METHODS: Peripheral blood mononuclear cells (PBMC) fronhealthy donors and patients with primary HCC were incubatedin vitro and induced into ClK cells in the presence of variouscytokines such as interferon-gamma (IFN-γ), interleukin-1(IL-1), IL-2, and monoclonal antibody (mAb) against CD3.The phenotype and characterization of CIK cells wereidentified by flow cytometric analysis. The cytotoxicity of CIKcells was determined by 51 Cr release assay.RESULTS: The CIK cells were shown to be a heterogeneouspopulation with different cellular phenotypes. Thepercentage of CD3+/CD56+ positive cells, the dominanteffector cells, in total CIK cells from healthy donors andHCC patients, significantly increased from 0.1-0.13 % at day0 to 19.0-20.5 % at day 21 incubation, which suggested thatthe CD3+ CD56+ positive cells proliferated faster than othercell populations of CIK cells in the protocol used in thisstudy. After 28 day in vitro incubation, the ClK cells frompatients with HCC and healthy donors increased by morethan 300-fold and 500-fold in proliferation cell number,respectively. CIK cells originated from HCC patientspossessed a higher in vitro antitumor cytotoxic activity onautologous HCC cells than the autologous lymphokine-activated killer (LAK) cells and PBMC cells. For the optimization in the fractional Fourier domain, an algorithm based on Quasi-Newton method is proposed which consists of two steps of searching, leading to a reduction in computation without loss of accuracy. And for multicomponent signals, we further propose a signal separation technique in the fractional Fourier domain which can effectively suppress the interferences on the detection of the weak components brought by the stronger components. In respect of petrography, geochemistry, Nd isotope, and single grain zircon U-Pb dating, the representative granite bodies of them are studied. The research shows that these granites were emplaced in two stages, namely 228-225 Ma BP and J2-3 159-156 Ma BP, belonging to Indosinian and early Yanshanian periods, respectively, and they have low εNd(t) values (-10.6--11.1), high A/CNK, Rb/Sr ratios and tDM values (1887-1817 Ma), and REE's tetrad effect (TE1, 3=1.13-1.34). In comparison with related geology, petrology and chronology of granites in adjacent regions, it is suggested that Indosinian SPGs of ENR formed in the circumstance of post-collisional extension 20 Ma after the major collision of Indosinian Movement (258-243 Ma BP) in Indo-China Peninsula, and early Yanshanian SPGs formed in the background of back-arc extension setting controlled by paleo-Pacific tectonic domain, and J1, the interval of two stages, is the interim from Tethyan to Pacific tectonic domains in South China. | 17 | Effect of phosphorylation of MAPK and Stat3 and expression of c-fos and c-jun proteins on hepatocarcinogenesis and their clinical significance显示文摘AIM To study the effect of phosphorylation ofMAPK and Stat3 and the expression of c-fos andc-jun proteins on hepatocellular carcinogenesisand their clinical significance.METHODS SP immunohistochemistry was usedto detect the expression of p42/44MAPK, p-Stat3,c-fos and c-jun proteins in 55 hepatocellularcarcinomas (HCC) and their surrounding livertissues.RESULTS The positive rates and expressionlevels of p42/44MAPK, p-Stat3, c-fos and c-junproteins in HCCs were significantly higher thanthose in pericarcinomatous liver tissues (PCLT).A positive correlation was observed between theexpression of p42/44MAPK and c-fos proteins, andbetween p-Stat3 and c-jun, but there was nosignificant correlation between p42/44MAPK and p-Stat3 in HCCs and their surrounding livertissues.CONCLUSION The abnormalities of Ras/Rat/MAPK and JAKs/ Stat3 cascade reaction maycontribute to malignant transformation ofhepatocytes. | 18 | Modulation of postoperative immune and inflammatory response by immune-enhancing enteral diet in gastrointestinal cancer patients显示文摘AIM To evaluate if the administration of anenteral diet supplemented with glutamine,arginine and ω-3-fatty acids modulatesinflammatory and immune responses aftersurgery.METHODS A prospective randomized double-blind, clinical trial was performed. Forty-eightpatients with gastrointestinal cancer wererandomized into two groups, one group wasgiven an isocaloric and isonitrogenous standarddiet and the other was fed with the supplementeddiet with glutamine, arginine and ωo-3-fattyacids. Feedings were started within 48 hoursafter operation, and continued until day 8. Allvariables were measured before operation andon postoperative day 1 and 8. Immune responseswere determined by phagocytosis ability,respiratory burst of polymorphonuclear cells,total lymphocytes lymphocyte subsets, nitricoxide, cytokines concentration, andinflammatory responses by plasma levels of C-reactive protein, prostaglandin E2 level.RESULTS Tolerance of both formula diets wasexcellent. There were siagnificant differences inthe immunological and inflammatory responsesbetween the two groups. In supplementedgroup, phagocytosis and respiratory burst aftersurgery was higher and C-reactive protein levelwas lower (P<0.01) than in the standard group.The supplemented group had higher levels ofnitric oxide, total Iymphocytes, T lymphocytes,T-helper cells, and NK cells. | 19 | Growth inhibition and apoptosis induction Sulindac on Human gastric cancer cells显示文摘AIM: To evaluate the effects of sulindac in inducing growth inhibition and apoptosis of human gastric cancer cells in comparison with human hepatocellular carcinoma (HCC)cells.METHODS: The human gastric cancer cell lines MKN45 and MKN28 and human hepatocellular carcinoma cell lines HepG2and SMMC7721 were used for the study. Anti-proliferative effect was measured by MTT assay, and apoptosis was determined by Hoechst-33258 staining, electronography and DNA fragmentation. The protein of cyclooxygenase-2 (COX(2) and Bcl-2 were detected by Westem dot blotting.RESULTS: Sulindac could initiate growth inhibition and apoptosis of MKN45, MKN28, HepG2 and SMMC7721 cells in a dose-and time-dependent manner. Growth inhibitory activity and apoptosis were more sensitive in HepG2 cells than in SMMC7721 cells, MKN45 and MKN28 cells. After 24hours incubation with sulindac at 2mmol. L-1 and 4mmol.L-1, the level of COX-2 and Bcl-2 protein were lowered in MKN45, SMMC7721 and HepG2 cells but not in MKN28 cells.CONCLUSION: Sulindac could inhibit the growth of gastric cancer cells and HCC cells effectively in vitro by apoptosis induction, which was associated with regression of COX-2and Bcl-2 expression. The growth inhibition and apoptosis of HCC cells were greater then that of human gastric cancer cells. | 20 | Association of H.pylori infection with gastric carcinoma:a Meta analysis显示文摘AIM: To follow the principles of evidence based medicine to reach the integrated results of these studies.METHODS: Twenty-one papers of case-control studies were selected, including 11 on gastric cancer, 7 on precancerous lesion of stomach and 3 on lymphoma of stomach: Meta analysis was used to sum up the odds ratios (OR) of these studies.RESULTS: H. Pylori vsgastric cancer (intestinal and diffuse type): the odds ratio from the fixed effect model is 3.0016(95% Cl 2.4197-3.7234, P < 0.001 ). H. Pylori vs precancerous lesion of stomach: a random effect model was used to calculate the summary odds ratio and its value is 2.5635 (95% Cl: 1.8477-3.5566, P < 0.01). H. Pylori vs lymphoma of stomach: though the quantity of literature is too small to make Meta analysis, the data of these 3 studies show that lymphoma of stomach is highly associated with H. Pylori infections.CONCLUSION: Since it had been revealed that H. Pylori infection pre-exists in gastric carcinoma and precancerous lesions, the results of Meta analysis present a strong evidence to support the conclusion that H.
