2Nonalcoholic fatty liver disease and aging: Epidemiology to management显示文摘Nonalcoholic fatty liver disease(NAFLD) is common in the elderly, in whom it carries a more substantial burden of hepatic(nonalcoholic steatohepatitis, cirrhosis and hepatocellular carcinoma) and extra-hepatic manifestations and complications(cardiovascular disease, extrahepatic neoplasms) than in younger age groups. Therefore, proper identification and management of this condition is a major task for clinical geriatricians and geriatric hepatologists. In this paper, the epidemiology and pathophysiology of this condition are reviewed, and a full discussion of the link between NAFLD and the aspects that are peculiar to elderly individuals is provided; these aspects include frailty, multimorbidity, polypharmacy and dementia. The proper treatment strategy will have to consider the peculiarities of geriatric patients, so a multidisciplinary approach is mandatory. Non-pharmacological treatment(diet and physical exercise) has to be tailored individually considering the physical limitations of most elderly people and the need for an adequate caloric supply. Similarly, the choice of drug treatment must carefully balance the benefits and risks in terms of adverse events and pharmacological interactions in the common context of both multiple health conditions and polypharmacy. In conclusion, further epidemiological and pathophysiological insight is warranted. 3Pathogenesis and significance of hepatitis C virus steatosis:An update on survival strategy of a successful pathogen显示文摘Hepatitis C virus(HCV)is a successful pathogen on the grounds that it exploits its host’s metabolism to build up viral particles;moreover it favours its own survival by inducing chronic disease and the development of specific anatomic changes in the infected organ.Steatosis,therefore,is associated with HCV infection by necessity rather than by chance alone.Approximately6%of HCV patients have steatohepatitis.Interestingly,HCV steatosis occurs in the setting of multiple metabolic abnormalities(hyperuricemia,reversible hypocholesterolemia,insulin resistance,arterial hypertension and expansion of visceral adipose tissue)collectively referred to as'hepatitis C-associated dysmetabolic syndrome'(HCADS).General,nonalcoholic fatty liver disease(NAFLD)-like,mechanisms of steatogenesis(including increased availability of lipogenic substrates and de novo lipogenesis;decreased oxidation of fatty substrates and export of fatty substrates)are shared by all HCV genotypes.However,genotype 3 seemingly amplifies such steatogenic molecular mechanisms reported to occur in NAFLD via more profound changes in microsomal triglyceride transfer protein;peroxisome proliferator-activated receptor alpha;sterol regulatory element-binding proteins and phosphatase and tensin homologue.HCV steatosis has a remarkable clinical impact in as much as it is an acknowledged risk factor for accelerated fibrogenesis;for impaired treatment response to interferon and ribavirin;and development of hepatocellular carcinoma.Recent data,moreover,suggest that HCV-steatosis contributes to premature atherogenesis via both direct and indirect mechanisms.In conclusion,HCV steatosis fulfills all expected requirements necessary to perpetuate the HCV life cycle.A better understanding of the physiology of HCADS will likely result in a more successful handling of disease with improved antiviral success rates.Amedeo Lonardo Luigi Elio Adinolfi Luciano Restivo Stefano Ballestri Dante Romagnoli Enrica Baldelli Fabio Nascimbeni Paola Loriaca 2014World Journal of Gastroenterology2014,20,23:14

5Relationship between air temperature oscillations and solar variability on short and medium time scales显示文摘The contemporary science of climate change is increasingly focusing on the temporal and spatial characteristics of temperature oscillations and determining possible underlying causes.In particular,the effect of variations in solar irradiance on the variability of the climate remains a hot topic of debate.Most studies focus on the effects of solar variation on the Earth's climate on long time scales.This study presents the responses of regional climates to solar variations on shorter time scales using two datasets:one for the air temperature in Nanjing and the Greenwich sunspot number,and the other for the air temperature in Shijiazhuang and the United States sunspot number.Employing empirical mode decomposition,both the 11-year quasi-period of the sunspot number and similar periods including approximately 5.5-and 10.5-year cycles of the air temperature in Nanjing and Shijiazhuang are obtained.However,correlation analysis of similar periodic components for the sunspot number and air temperature indicates that changes in the air temperature on short and medium time scales are not linked to solar variations.This is further confirmed by a test of whether a mode component is a stochastic noise signal.Many shorter periods are also found at the 95% confidence level;in particular,the 3.1-year period of the Nanjing air temperature coincides with a previously obtained empirical result.Moreover,no temperature variations on shorter time scales correlate with solar variability.YANG ChunXia WU HongFa HU DanTing 2011Science China Earth Sciences2011,54,6:6
6Mapping the immune microenvironment for mandibular alveolar bone homeostasis at single-cell resolution显示文摘Alveolar bone is the thickened ridge of jaw bone that supports teeth.It is subject to constant occlusal force and pathogens invasion,and is therefore under active bone remodeling and immunomodulation.Alveolar bone holds a distinct niche from long bone considering their different developmental origin and postnatal remodeling pattern.However,a systematic explanation of alveolar bone at single-cell level is still lacking.Here,we construct a single-cell atlas of mouse mandibular alveolar bone through single-cell RNA sequencing(scRNA-seq).A more active immune microenvironment is identified in alveolar bone,with a higher proportion of mature immune cells than in long bone.Among all immune cell populations,the monocyte/macrophage subpopulation most actively interacts with mesenchymal stem cells(MSCs)subpopulation.Alveolar bone monocytes/macrophages express a higher level of Oncostatin M(Osm)compared to long bone,which promotes osteogenic differentiation and inhibits adipogenic differentiation of MSCs.In summary,our study reveals a unique immune microenvironment of alveolar bone,which may provide a more precise immune-modulatory target for therapeutic treatment of oral diseases.Weimin Lin Qiwen Li Danting Zhang Xiaohan Zhang Xingying Qi Qian Wang Yaqian Chen Caojie Liu Hanwen Li Shiwen Zhang Yuan Wang Bin Shao Li Zhang Quan Yuan 2021Bone Research2021,9,1:6
7A case of biliary gastric fistula following percutaneous radiofrequency thermal ablation of hepatocellular carcinoma显示文摘Percutaneous radiofrequency thermal ablation (RFA) is an effective and safe therapeutic modality in the management of liver malignancies, performed with ultrasound guidance. Potential complications of RFA include liver abscess, ascites, pleural effusion, skin burn, hypoxemia, pneumothorax, subcapsular hematoma, hemoperitoneum, liver failure, tumour seeding, biliary lesions. Here we describe for the first time a case of biliary gastric fistula occurred in a 66-year old man with a Child’s class A alcoholic liver cirrhosis as a complication of RFA of a large hepatocellular carcinoma lesion in the Ⅲ segment. 8Clinical features and natural history of cryptogenic cirrhosis compared to hepatitis C virus-related cirrhosis显示文摘AIM To characterize natural history of cryptogenic cirrhosis(CC) and compare its clinical features and outcomes to those of hepatitis C virus(HCV)-related cirrhosis.METHODS A prospective cohort of 102 consecutive patients at their first diagnosis of CC were enrolled in this study. The clinical data and outcomes were compared to an ageand Child-pugh class-matched cohort of 110 patients with HCV-related cirrhosis. Diagnosis of cirrhosis was based on compatible clinical and laboratory parameters, ultrasound/endoscopic parameters and, whenever possible, on histological grounds and transient elastography. All cases of cirrhosis without a definite etiology were enrolled in the CC group. The parameters assessed were:(1) severity of liver disease at the time of first diagnosis;(2) liver decompensation during follow-up;(3) hepatocellular carcinoma(HCC);(4) orthotopic liver transplantation; and(5) death. The independent associated factors were evaluated by multiple logistic regression analysis, and survival and its determinants by the Kaplan-Meier model, log-rank test and Cox regression.RESULTS At the first observation, median age was 66 and 65 years and male gender was 36% and 58% for CC and HCV cirrhosis, respectively. CC showed Child-pugh class A/B/C of 47%/31%/22%, respectively. Compared to HCV cirrhosis, CC exhibited a significantly higher prevalence of metabolic syndrome(12% vs 54%, respectively), overweight/obesity, high BMI, impaired glucose tolerance, high blood pressure, dyslipidemia, hyperuricemia, cardiovascular diseases, extrahepatic cancer, and gallstones. Over a median period of 42 mo of follow-up, liver decompensation, HCC development and death for CC and HCV-related cirrhosis were 60.8%, and 54.4%, 16.7% and 17.2%, 39.2% and 30%, respectively. The median survival was 60 mo for CC. Independent predictors of death were age and Childpugh class at diagnosis. 9Imaging shallow structure with active-source surface wave signal recorded by distributed acoustic sensing arrays显示文摘Distributed acoustic sensing(DAS) is one recently developed seismic acquisition technique that is based on fiber-optic sensing. DAS provides dense spatial spacing that is useful to image shallow structure with surface waves.To test the feasibility of DAS in shallow structure imaging,the PoroTomo team conducted a DAS experiment with the vibroseis truck T-Rex in Brady's Hot Springs, Nevada, USA.The Rayleigh waves excited by the vertical mode of the vibroseis truck were analyzed with the Multichannel Analysis of Surface Waves(MASW) method. Phase velocities between5 and 20 Hz were successfully extracted for one segment of cable and were employed to build a shear-wave velocity model for the top 50 meters. The dispersion curves obtained with DAS agree well with the ones extracted from co-located geophones data and from the passive source Noise Correlation Functions(NCF). Comparing to the co-located geophone array, the higher sensor density that DAS arrays provides help reducing aliasing in dispersion analysis, and separating different surface wave modes. 10Functional metabolomics reveal the role of AHR/GPR35 mediated kynurenic acid gradient sensing in chemotherapy-induced intestinal damage显示文摘Intestinal toxicity induced by chemotherapeutics has become an important reason for the interruption of therapy and withdrawal of approved agents. In this study, we demonstrated that chemotherapeutics-induced intestinal damage were commonly characterized by the sharp upregulation of tryptophan(Trp)àkynurenine(KYN)àkynurenic acid(KA) axis metabolism. Mechanistically,chemotherapy-induced intestinal damage triggered the formation of an interleukin-6(IL-6)àindoleamine2,3-dioxygenase 1(IDO1)àaryl hydrocarbon receptor(AHR) positive feedback loop, which accelerated kynurenine pathway metabolism in gut. Besides, AHR and G protein-coupled receptor 35(GPR35) negative feedback regulates intestinal damage and inflammation to maintain intestinal integrity and homeostasis through gradually sensing kynurenic acid level in gut and macrophage, respectively. Moreover, based on virtual screening and biological verification, vardenafil and linagliptin as GPR35 and AHR agonists respectively were discovered from 2388 approved drugs. 12Fatty liver is associated with an increased risk of diabetes and cardiovascular disease- Evidence from three different disease models: NAFLD, HCV and HIV显示文摘Fatty liver, which frequently coexists with necroinflammatory and fibrotic changes, may occur in the setting of nonalcoholic fatty liver disease(NAFLD) and chronic infections due to either hepatitis C virus(HCV) or human immunodeficiency virus(HIV). These three pathologic conditions are associated with an increased prevalence and incidence of cardiovascular disease(CVD) and type 2 diabetes(T2D). In this multidisciplinary clinical review, we aim to discuss the ever-expanding wealth of clinical and epidemiological evidence supporting a key role of fatty liver in the development of T2 D and CVD in patients with NAFLD and in those with HCV or HIV infections. For each of these three common diseases, the epidemiological features, pathophysiologic mechanisms and clinical implications of the presence of fatty liver in predicting the risk of incident T2 D and CVD are examined in depth. Collectively, the data discussed in this updated review, which follows an innovative comparative approach, further reinforce the conclusion that the presence of fatty/inflamed/fibrotic liver might be a shared important determinant for the development of T2 D and CVD in patients with NAFLD, HCV or HIV. 13The association of cerebrospinal fluid pressure with optic nerve head and macular vessel density显示文摘The present study aims to investigate the effect of temporary cerebrospinal fluid pressure(CSFP) reduction on optic nerve head(ONH) and macular vessel density(VD) using optical coherence tomography angiography.Forty-four eyes of 44 adults with diagnostic lumbar puncture and CSFP reduction were recruited.Thirty-two eyes of 32 healthy volunteers were controls.ONH and macular VD images were evaluated differences between baseline and after CSFP reduction.The results showed that the mean CSFP decreased from(11.6±2.1) mm Hg to(8.2±3.4) mm Hg(P<0.001).VD in the macular regions decreased significantly after CSFP reduction in the study group(all P<0.05).The control group showed no significant changes in macular VD(all P>0.05).In the study group,decreased VD in the macular parainferior region was associated with CSFP reduction(R^(2)=0.192,P=0.003),the reduction of macular VD in parafoveal(R^(2)=0.098,P=0.018),parainferior(R^(2)=0.104,P=0.021),parasuperior(R^(2)=0.059,P=0.058),paranasal(R^(2)=0.057,P=0.042),paratemporal(R^(2)=0.079,P=0.026) was associated with mean ocular perfusion pressure decrease following CSFP reduction.ONH vessel density did not differ after CSFP reduction(all P>0.05).In conclusion,macular vessel density decreased in association with CSFP reduction.Retinal vessel density in the macular region is more sensitive than that in peripapillary region after CSFP reduction.Xiangxiang Liu Mohamed M.Khodeiry Danting Lin Yunxiao Sun Qing Zhang Jiawei Wang Richard K.Lee Ningli Wang 2022Science China(Life Sciences)2022,65,6:3
14Drought and Spatiotemporal Variability of Forest Fires Across Mexico显示文摘Understanding the spatiotemporal links between drought and forest fire occurrence is crucial for improving decision-making in fire management under current and future climatic conditions. We quantified forest fire activity in Mexico using georeferenced fire records for the period of 2005–2015 and examined its spatial and temporal relationships with a multiscalar drought index, the Standardized Precipitation-Evapotranspiration Index(SPEI). A total of 47975 fire counts were recorded in the 11-year long study period, with the peak in fire frequency occurring in 2011. We identified four fire clusters, i.e., regions where there is a high density of fire records in Mexico using the Getis-Ord G spatial statistic. Then, we examined fire frequency data in the clustered regions and assessed how fire activity related to the SPEI for the entire study period and also for the year 2011. Associations between the SPEI and fire frequency varied across Mexico and fire-SPEI relationships also varied across the months of major fire occurrence and related SPEI temporal scales. In particular, in the two fire clusters located in northern Mexico(Chihuahua, northern Baja California), drier conditions over the previous 5 months triggered fire occurrence. In contrast, we did not observe a significant relationship between drought severity and fire frequency in the central Mexico cluster, which exhibited the highest fire frequency. We also found moderate fire-drought associations in the cluster situated in the tropical southern Chiapas where agriculture activities are the main causes of forest fire occurrence. 15Driving Factors for Forest Fire Occurrence in Durango State of Mexico:A Geospatial Perspective显示文摘Forest fire is one of the major causes of forest loss and therefore one of the main constraints for sustainable forest management worldwide.Identifying the driving factors and understanding the contribution of each factor are essential for the management of forest fire occurrence.The objective of this study is to identify variables that are spatially related to the occurrence and incidence of the forest fire in the State of Durango,Mexico.For this purpose,data from forest fire records for a five-year period were analyzed.The spatial correlations between forest fire occurrence and intensity of land use,susceptibility of vegetation,temperature,precipitation and slope were tested by Geographically Weighted Regression(GWR) method,under an Ordinary Least Square estimator.Results show that the spatial pattern of the forest fire in the study area is closely correlated with the intensity of land use,and land use change is one of the main explanatory variables.In addition,vegetation type and precipitation are also the main driving factors.The fitting model indicates obvious link between the variables.Forest fire was found to be the consequence of a particular combination of the environmental factors,and when these factors coexist with human activities,there is high probability of forest fire occurrence.Mandatory regulation of human activities is a key strategy for forest fire prevention.Diana Avila-Flores Marin Pompa-Garcia Xanat Antonio-Nemiga Dante A Rodriguez-Trejo Eduardo Vargas-Perez Javier Santillan-Perez 2010Chinese Geographical Science2010,20,6:3

17Guanylyl cyclase C signaling axis and colon cancer prevention显示文摘Colorectal cancer(CRC) is a major cause of cancerrelated mortality and morbidity worldwide. While improved treatments have enhanced overall patient outcome, disease burden encompassing quality of life, cost of care, and patient survival has seen little benefit. Consequently, additional advances in CRC treatments remain important, with an emphasis on preventative measures. Guanylyl cyclase C(GUCY2C), a transmembrane receptor expressed on intestinal epithelial cells, plays an important role in orchestrating intestinal homeostatic mechanisms. These effects are mediated by the endogenous hormones guanylin(GUCA2A) and uroguanylin(GUCA2B), which bind and activate GUCY2 C to regulate proliferation, metabolism and barrier function in intestine. Recent studies have demonstrated a link between GUCY2 C silencing and intestinal dysfunction, including tumorigenesis. Indeed, GUCY2 C silencing by the near universal loss of its paracrine hormone ligands increases colon cancer susceptibility in animals and humans. GUCY2C's role as a tumor suppressor has opened the door to a new paradigm for CRC prevention by hormone replacement therapy using synthetic hormone analogs, such as the FDA-approved oral GUCY2 C ligand linaclotide(Linzess^(TM)). 20Priming of NLRP3 inflammasome activation by Msn kinase MINK1 in macrophages显示文摘The nucleotide-binding domain,leucine-rich-repeat containing family,pyrin domain-containing 3(NLRP3)inflammasome is essential in inflammation and inflammatory disorders.Phosphorylation at various sites on NLRP3 differentially regulates inflammasome activation.The Ser725 phosphorylation site on NLRP3 is depicted in multiple inflammasome activation scenarios,but the importance and regulation of this site has not been clarified.The present study revealed that the phosphorylation of Ser725 was an essential step for the priming of the NLRP3 inflammasome in macrophages.We also showed that Ser725 was directly phosphorylated by misshapen(Msn)/NIK-related kinase 1(MINK1),depending on the direct interaction between MINK1 and the NLRP3 LRR domain.MINK1 deficiency reduced NLRP3 activation and suppressed inflammatory responses in mouse models of acute sepsis and peritonitis.Reactive oxygen species(ROS)upregulated the kinase activity of MINK1 and subsequently promoted inflammasome priming via NLRP3 Ser725 phosphorylation.Eliminating ROS suppressed NLRP3 activation and reduced sepsis and peritonitis symptoms in a MINK1-dependent manner.Altogether,our study reveals a direct regulation of the NLRP3 inflammasome by Msn family kinase MINK1 and suggests that modulation of MINK1 activity is a potential intervention strategy for inflammasomerelated diseases.Kaixiang Zhu Xuexiao Jin Zhexu Chi Sheng Chen Songquan Wu Richard D.Sloan Xuai Lin Dante Neculai Di Wang Hu Hu Linrong Lu 2021Cellular & Molecular Immunology2021,18,10:2
