1Triptolide (PG-490) induces apoptosis of dendritic cells through sequential p38 MAP kinase phosphorylation and caspase 3 activation显示文摘Dendritic cells (DCs) are the most potent antigen-presen ting cells that play crucial roles in the regulation of immune response. Triptol ide, an active component purified from the medicinal plant Tripterygium wilfor dii Hook F., has been demonstrated to act as a potent immunosuppressive drug c apab le of inhibiting T cell activation and proliferation. However, little is known a bout the effects of triptolide on DCs. The present study shows that triptolide d oes not affect phenotypic differentiation and LPS-induced maturation of murine DCs. But triptolide can dramatically reduce cell recovery by inducing apoptosis of DCs at concentration as low as 10 ng/ml, as demonstrated by phosphatidylserin e exposure, mitochondria potential decrease, and nuclear DNA condensation. Tript olide induces activation of p38 in DCs, which precedes the activation of caspase 3. SB203580, a specific kinase inhibitor for p38, can block the activation of caspase 3 and inhibit the resultant apoptosis of DCs. 2Identification of an HLA-A~* 0201-restricted CD8^+ T-cell epitope SSp-1 of SARS-CoV spike protein显示文摘A novel coronavirus, severe acute respiratory syndrome (SA RS)-associated coronavirus (SARS-CoV), has been identified as the causal agent of SARS. Spike (S) protein is a major structural glycoprotein of the SARS virus and a potential target for SARS-specific cell-mediated immune responses. A pa nel of S protein-derived peptides was tested for their binding affinity to HLA -A *0201 molecules. Peptides with high affinity for HLA-A *0201 were then as se ssed for their capacity to elicit specific immune responses mediated by cytotoxi c T lymphocytes (CTLs) both in vivo, in HLA-A2.1/K b transgenic mice, a nd in vitro, from peripheral blood lymphocytes (PBLs) harvested from healthy HLA-A 2.1 + donors. SARS-CoV protein-derived peptide-1 (SSp-1 RLNEVAKNL), induced pepti de-specific CTLs both in vivo (transgenic mice) and in vitro (human PBL s), which specifically released interferon-gamma (IFN-gamma) upon stimulation with SSp-1-pulsed autologous dendritic cells (DCs) or T2 cells. SSp-1-specif ic CTLs also lysed major histocompatibility complex (MHC)-matched tumor cell lines engineered to express S proteins. HLA-A *0201-SSp-1 tetramer staining re vealed the presence of significant populations of SSp-1-specific CTLs in SSp- 1-induced CD8 + T cells. 3De novo assembly of a new Olea europaea genome accession using nano pore seque ncing显示文摘Olive(Olea europaea L.)is internationally renowned for its high-end product,extra virgin olive oil.An incomplete genome of O.europaea was previously obtained using shotgun sequencing in 2016.To further explore the genetic and breeding utilization of olive,an updated draft genome of olive was obtained using Oxford Nanopore third-generation sequencing and Hi-C technology.Seven different assembly strategies were used to assemble the fi nal genome of 1.30 Gb,with contig and scaffold N50 sizes of4.67 Mb and 42.60 Mb,respectively.This greatly increased the quality of the olive genome.We assembled 1.1 Gb of sequences of the total olive genome to 23 pseudochromosomes by Hi-C,and 53,518 protein-coding genes were predicted in the current assembly.Comparative genomics analyses,including gene family expansion and contraction,whole-genome replication,phylogenetic analysis,and positive selection,were performed.Based on the obtained high-quality olive genome,a total of nine gene families with 202 genes were identi fi ed in the oleuropein biosynthesis pathway,which is twice the number ofgenes identi fi ed from the previous data.This new accession of the olive genome is of suf fi cient quality for genome-wide studies on gene function in olive and has provided a foundation for the molecular breeding of olive species.Guodong Rao Jianguo Zhang Xiaoxia Liu Chunfu Lin Huaigen Xin Li Xue Chenhe Wang 2021Horticulture Research2021,8,1:4
4High-resolution digital beamforming of UWB signals based on Carathéodory representation for delay compensation and array extrapolation显示文摘To realize high-resolution digital beamforming(DBF)of ultra-wideband(UWB) signals, we propose a DBF method based on Carath ′eodory representation for delay compensation and array extrapolation. Delay compensation by Carath ′eodory representation could achieve high interpolation accuracy while using the single channel sampling technique. Array extrapolation by Carath ′eodory representation reformulates and extends each snapshot, consequently extends the aperture of the original uniform linear array(ULA) by several times and provides a better realtime performance than the existing aperture extrapolation utilizing vector extrapolation based on the two dimensional autoregressive(2-D AR) model. The UWB linear frequency modulated(LFM) signal is used for simulation analysis.
