1积雪草酸对结肠癌HCT116细胞增殖及自噬水平的影响显示文摘目的:观察积雪草酸对结肠癌HCT116细胞增殖和自噬水平的影响。方法:用10、30、60、90μmol/L的积雪草酸作用于结肠癌HCT116细胞24、48 h,用四甲基偶氮唑盐比色法(MTT法)检测细胞存活率;用MDC自噬特异性荧光染料观察自噬囊泡的形成情况;蛋白质印迹法检测30、60、90μmol/L的积雪草酸作用12 h和90μmol/L积雪草酸作用4、8、12 h的细胞自噬相关蛋白LC3-Ⅱ、p62、p-m TOR、p-4EBP1的表达。结果:积雪草酸90μmol/L对结肠癌HCT116细胞有明显抑制作用;30、60、90μmol/L积雪草酸均可以诱导结肠癌细胞发生自噬,90μmol/L积雪草酸作用4、8、12 h,自噬蛋白LC3-Ⅱ生成明显增加;90μmol/L积雪草酸与氯喹5μmol/L共同作用于癌细胞12 h,能促进自噬潮的产生;积雪草酸可抑制p-m TOR、p-4EBP1的表达。结论:积雪草酸能明显抑制结肠癌HCT116细胞的增殖,并呈时间、剂量依赖性地抑制m TOR-4EBP1通路来诱导自噬潮的产生。耿骥 郭文洁 刘佳 谈德斐 Daniel Boison 高静 2015江苏大学学报(医学版)2015,25,2:10
