| 1 | Short-chain fatty acids act as antiinflammatory mediators by regulating prostaglandin E_2 and cytokines显示文摘AIM:To investigate the effect of short-chain fatty ac-ids (SCFAs) on production of prostaglandin E2 (PGE2),cytokines and chemokines in human monocytes.METHODS:Human neutrophils and monocytes were isolated from human whole blood by using 1-Step Polymorph and RosetteSep Human Monocyte Enrich-ment Cocktail,respectively. Human GPR41 and GPR43 mRNA expression was examined by quantitative real-time polymerase chain reaction. The calcium flux assay was used to examine the biological activities of SCFAs in human neutrophils and monocytes. The effect ofSCFAs on human monocytes and peripheral blood mononuclear cells (PBMC) was studied by measuring PGE2,cytokines and chemokines in the supernatant. The effect of SCFAs in vivo was examined by intraplantar injection into rat paws.RESULTS:Human GPR43 is highly expressed in human neutrophils and monocytes. SCFAs induce robust calcium flux in human neutrophils,but not in human monocytes. In this study,we show that SCFAs can in-duce human monocyte release of PGE2 and that this effect can be enhanced in the presence of lipopolysac-charide (LPS). In addition,we demonstrate that PGE2 production induced by SCFA was inhibited by pertussis toxin,suggesting the involvement of a receptor-mediated mechanism. Furthermore,SCFAs can specifi cally inhibit constitutive monocyte chemotactic protein-1 (MCP-1) production and LPS-induced interleukin-10 (IL-10) production in human monocytes without affecting the secretion of other cytokines and chemokines examined. Similar activities were observed in human PBMC for the release of PGE2,MCP-1 and IL-10 after SCFA treatment. In addition,SCFAs inhibit LPS-induced production of tumor necrosis factor-α and interferon-γ in human PBMC. Finally,we show that SCFAs and LPS can induce PGE2 production in vivo by intraplantar injection into rat paws (P < 0.01). | 2 | Estrogen deficiency reversibly induces telomere shortening in mouse granulosa cells and ovarian aging in vivo显示文摘Estrogen is implicated as playing an important role in aging and tumorigenesis of estrogen responsive tissues;however the mechanisms underlying the mitogenic actions of estrogen are not fully understood.Here we report that estrogen deficiency in mice caused by targeted disruption of the aromatase gene results in a signi-ficant inhibition of telomerase maintenance of telomeres in mouse ovaries in a tissue-specific manner.The inhibition entails a significant shortening of telomeres and compromised proliferation in the follicular granulosa cell compartment of ovary.Gene expression analysis showed decreased levels of proto-oncogene c-Myc and the telomerase catalytic subunit,telomerase reverse transcriptase(TERT),in response to estrogen deficiency.Estrogen replacement therapy led to increases in TERT gene expression,telomerase activity,telomere length and ovarian tissue growth,thereby reinstating ovary development to normal in four weeks.Our data demonstrate for the first time that telomere maintenance is the primary mechanism mediating the mitogenic effect of estrogen on ovarian granulosa cell proliferation by upregulating the genes of c-Myc and TERT in vivo.Estrogen deficiency or over-activity may cause ovarian tissue aging or tumorigenesis,respectively,through estrogen regulation of telomere remodeling. | Sharyn Bayne He Li Margaret E.E.Jones Alex R.Pinto Michelle van Sinderen Ann Drummond Evan R.Simpson Jun-Ping Liu | 2011 | Protein & Cell2011,2,4: | 8 |

| 4 | Dealing with detection error in site occupancy surveys: what can we do with a single survey?显示文摘Aim Site occupancy probabilities of target species are commonly used in various ecological studies,e.g.to monitor current status and trends in biodiversity.Detection error introduces bias in the estimators of site occupancy.Existing methods for estimating occupancy probability in the presence of detection error use replicate surveys.These methods assume population closure,i.e.the site occupancy status remains constant across surveys,and independence between surveys.We present an approach for estimating site occupancy probability in the presence of detection error that requires only a single survey and does not require assumption of population closure or independence.In place of the closure assumption,this method requires covariates that affect detection and occupancy.Methods Penalized maximum-likelihood method was used to estimate the parameters.Estimability of the parameters was checked using data cloning.Parametric boostrapping method was used for computing confidence intervals.Important Findings The single-survey approach facilitates analysis of historical datasets where replicate surveys are unavailable,situations where replicate surveys are expensive to conduct and when the assumptions of closure or independence are not met.This method saves significant amounts of time,energy and money in ecological surveys without sacrificing statistical validity.Further,we show that occupancy and habitat suitability are not synonymous and suggest a method to estimate habitat suitability using single-survey data. | Subhash R.Lele Monica Moreno Erin Bayne | 2012 | Journal of Plant Ecology2012,5,1: | 2 |
