1Antioxidant activity and phytochemical screening of the methanol extracts of Euphorbia hirta L显示文摘Objective:To assess antioxidant activities of different parts of Euphorbia hirta(E.hirta),and to search for new sources of safe and inexpensive antioxidants.Methods:Samples of leaves,stems, flowers and roots from E.hirta were tested for total phenolic content,and flavonoids content and in vitro antioxidant activity by diphenyl-1-picrylhydrazyl(DPPH) assay and reducing power was measured using cyanoferrate method.Results:The leaves extract exhibited a maximum DPPH scavenging activity of(72.96±0.78)%followed by the flowers,roots and stems whose scavenging activities were(52.45±0.66)%,(48.59±0.97)%,and(44.42±0.94)%,respectively.The standard butylated hydroxytoluene(BHT) was(75.13±0.75)%.The IC50,for leaves,flowers,roots,stems and BHT were 0.803,0.972,0.989,1.358 and 0.794 mg/mL,respectively.The reducing power of the leaves extract was comparable with that of ascorbic acid and found to be dose dependent. Leaves extract had the highest total phenolic content[(206.17±1.95) mg GAE/g],followed by flowers,roots and stems extracts which were(117.08±3.10) mg GAE/g,(83.15±1.19) mg GAE/g,and (65.70±1.72) mg GAE/g,respectively.On the other hand,total flavonoids content also from leave had the highest value[(37.970±0.003) mg CEQ/g],followed by flowers,roots and stems extracts which were(35.200±0.002) mg CEQ/g,(24.350±0.006) mg CEQ/g,and(24.120±0.004) mg CEQ/ g,respectively.HPTLC bioautography analysis of phenolic and antioxidant substance revealed phenolic compounds.Phytochemical screening of E.hirta leaf extract revealed the presence of reducing sugars,terpenoids,alkaloids,steroids,tannins,flavanoids and phenolic compounds. 2Association between low molecular polypeptide 7 single nucleotide polymorphism and response to therapy in hepatitis C virus infection显示文摘AIM: To investigate the relationship between low molecular polypeptide-7 (LMP-7) gene polymorphism and response to interferon (IFN) therapy in chronic hepatitis C virus (HCV) patients. METHODS: LMP-7 polymorphism at codon 49 with nucleotide substitution from A to C was amplified in 104 chronic HCV patients of genotype 4. The amplicons were digested with restriction endonuclease Bsm I and the produced restriction fragment length polymorphism was analyzed. Patients received IFN + regional blood volume therapy for 48 wk and the frequency of thissingle nucleotide polymorphism (SNP) was statistically correlated with treatment response. The exclusion criteria for these patients were stated by the national health program for treating viral hepatitis. Main exclusion criteria included co-infection with hepatitis B virus or schistosomiasis, thyroid dysfunction, uncontrolled diabetes mellitus, history of long term drug or alcohol intake and autoimmune hepatitis. Multivariate analyses were done to correlate LMP-7 SNP plus several factors such as age, gender, weight, serum alpha-fetoprotein (AFP) and alanine aminotransferase levels, liver activity, fibrosis score and viral load with response to therapy. RESULTS: The data presented in this study clearly demonstrated statistically significant differences between sustained virological response (SVR) (defined as the absence of HCV RNA levels in the patient's sera at least 6 mo after discontinuation of treatment) and non-response (NR) (where HCV RNA levels in the patient's sera never become undetectable for 6 mo during or after treatment). Variables were described as odds ratio with 95%CI. The data were considered significant if P values were ≤ 0.05; highly significant if P < 0.01 and very highly significant if P < 0.001. Current data showed that 91.7% of patients carrying LMP-7 C/C allele were associated with SVR, while the other two genotypes C/A and A/A were associated with NR patients, 83.3% and 64.3% respectively, showing that genotype CC was strongly associated with response to interferon (95%CI: 12.0719-134.6572, P = 0.0001). The majority of parameters recorded in SVR and NR patients included higher values of mean age (P = 0.004), alanine aminotransferase (P = 0.001), AFP (P = 0.001), body weight (P = 0.025), viral load (P = 0.025), higher fibrosis and histological activity index indices among NR vs SVR patients. Also, the multivariate statistical analysis of the different factors of fibro-sis score, liver activity grade, genotypes and alleles of LMP-7 gene polymorphism in responders and NRs of HCV patients in this study showed that HCV patients with A allele had a very highly significant association with the NRs, high fibrosis and higher liver activity, while the C allele had a very highly significant association with the responders, low fibrosis and lower liver activity (95%CI: 3.5800-13.2519, P = 0.0001).CONCLUSION: LMP-7 SNP is a candidate gene that should be considered when designing a mathematical model for predicting response to therapy and disease progression in HCV patients.Moataza H Omran Basma E Fotouh Samar S Youssef Noha E Ibrahim Wael Nabil EL-Sayed M Mahdy Wafaa G Shosha Mostafa K El-Awady 2013World Journal of Hepatology2013,5,3:4
3Acid leaching of heavy metals from contaminated soil collected from Jeddah,Saudi Arabia:kinetic and thermodynamics studies显示文摘Urban soils polluted with heavy metals are of increasing concern because it is greatly affecting human health and the ecological systems.Hence,it is mandatory to understand the reasons behind this pollution and remediate the contaminated solid.The removal of heavy metals from contaminated soil samples collected from the vicinity of the sewage lake in Jeddah,Saudi Arabia,was explored.The leaching process was studied kinetically and thermodynamically for better understanding of the remediation process.The results showed that the soil samples were slightly basic in nature,and tend to be more neutral away from the main contaminated sewage lake area.The total metal content in the soil samples was measured using the aqua regia extractions by ICPOES and the results showed that many of the heavy metals present have significant concentrations above the tolerable limits.In general,the metal concentrations at different sites indicated that the heavy metal pollution is mainly due to the sewage discharge to the lake.The results showed excellent correlation between the concentrations of Co,As,and Hg with the distance from the main contaminated area.The leaching of Co,As,and Hg using 1.0 M hydrochloric acid from the soil was studied kinetically at different temperatures and the experimental results were fitted using different kinetics models.The experimental data were best described with two-constant rate and Elovich equation kinetic models.Also,the thermodynamic study showed that the leaching process was spontaneous,endothermic and accompanied with increase in the entropy.In general,the polluted soil could be remediated successfully from the heavy metals using the acid leaching procedure in a short period of time.Shorouq I.Alghanmi Amani F.Al Sulami Tahani A.El-Zayat Basma G.Alhogbi Mohamed Abdel Salam 2015International Soil and Water Conservation Research2015,3,3:4

5Role for urinary biomarkers in diagnosis of acute rejection in the transplanted kidney显示文摘Despite the introduction of potent immunosuppressive medications within recent decades, acute rejection still accounts for up to 12% of all graft losses, and is generally associated with an increased risk of late graft failure. Current detection of acute rejection relies on frequent monitoring of the serum creatinine followed by a diagnostic renal biopsy. This strategy is flawed since an alteration in the serum creatinine is a late clinical event and significant irreversible histologic damage has often already occurred. Furthermore, biopsies are invasive procedures that carry their own inherent risk. The discovery of non-invasive urinary biomarkers to help diagnose acute rejection has been the subject of a significant amount of investigation. We review the literature on urinary biomarkers here, focusing on specific markers perforin and granzyme B m RNAs, FOXP3 m RNA, CXCL9/CXCL10 and mi RNAs. 6A transmission electron microscopy study of the diversity of Candida albicans cells induced by Euphorbia hirta L.leaf extract in vitro显示文摘Objective:To determine the major changes in the microstructure of Candida albicans(C. albicans) after treatment with Euphorbia hirta(E.hirta) L.leaf extract.Methods:Transmission electron microscopy was used to study the ultrastructural changes caused by E.hirta extract on C. albicans cells al various exposure time.Results:It was found that the main abnormalities were the alterations in morphology,lysis and complete collapse of the yeast cells after 36 h of exposure to the extract.Whereas the control cultures showed a typical morphology of Candida with a uniform central density,typically structured nucleus,and a cytoplasm with several elements of endomembrane system and enveloped by a regular,intact cell wall.Conclusions:The significant antifungal activity shown by this methanol extract of E.hirta L.suggests its potential against infections caused by C.albicans.The extract may be developed as an anticandidal agent.Abu Arra Basma Zakaria Zuraini Sreenivasan Sasidharan 2011Asian Pacific Journal of Tropical Biomedicine2011,1,1:3
